    Remote monitoring of industrial frequency converters

    Frequency converters are sometimes mounted at places, in which they have the most processing power of the surrounding devices. Often the remote monitoring of frequency converters has been implemented in an external programmable logic controller PC which is connected to the Internet. Sometimes it is not reasonable to use an extra computer at the location, which means the remote connection should be directly connectible to the frequency converter. This master’s thesis studies the possibility to use SOAP for a remote connection, established from the frequency converter to an external database server. The objective is to create a remote monitoring connection which is easily deployable for the end user.     Prevention of Rat Intestinal Injury with a Drug Combination of Melatonin and Misoprostol

    A healthy intestinal barrier prevents uptake of allergens and toxins, whereas intestinal permeability increases following chemotherapy and in many gastrointestinal and systemic diseases and disorders. Currently, there are no approved drugs that target and repair the intestinal epithelial barrier while there is a medical need for such treatment in gastrointestinal and related conditions. The objective of this single-pass intestinal perfusion study in rats was to investigate the preventive cytoprotective effect of three mucosal protective drugs¿melatonin, misoprostol, and teduglutide¿with different mechanisms of action on an acute jejunal injury induced by exposing the intestine for 15 min to the anionic surfactant, sodium dodecyl sulfate (SDS). The effect was evaluated by monitoring intestinal clearance of 51Cr-labeled ethylenediaminetetraacetate and intestinal histology before, during, and after luminal exposure to SDS. Our results showed that separate pharmacological pretreatments with luminal misoprostol and melatonin reduced acute SDS-induced intestinal injury by 47% and 58%, respectively, while their use in combination abolished this injury. This data supports further development of drug combinations for oral treatments of conditions and disorders related to a dysregulated or compromised mucosal epithelial barrier

    Background: The clinical behaviour of colon cancer is heterogeneous. Five-year overall survival is 50-65% with all stages included. Recurring somatic chromosomal alterations have been identified and some have shown potential as markers for dissemination of the tumour, which is responsible for most colon cancer deaths. We investigated 115 selected stage II-IV primary colon cancers for associations between chromosomal alterations and tumour dissemination. Methods: Follow-up was at least 5 years for stage II-III patients without distant recurrence. Affymetrix SNP 6.0 microarrays and allele-specific copy number analysis were used to identify chromosomal alterations. Fisher's exact test was used to associate alterations with tumour dissemination, detected at diagnosis (stage IV) or later as recurrent disease (stage II-III). Results: Loss of 1p36.11-21 was associated with tumour dissemination in microsatellite stable tumours of stage II-IV (odds ratio = 5.5). It was enriched to a similar extent in tumours with distant recurrence within stage II and stage III subgroups, and may therefore be used as a prognostic marker at diagnosis. Loss of 1p36.11-21 relative to average copy number of the genome showed similar prognostic value compared to absolute loss of copies. Therefore, the use of relative loss as a prognostic marker would benefit more patients by applying also to hyperploid cancer genomes. The association with tumour dissemination was supported by independent data from the The Cancer Genome Atlas. Conclusion: Deletions on 1p36 may be used to guide adjuvant treatment decisions in microsatellite stable colon cancer of stages II and III

    KRAS-G12C Mutation in One Real-Life and Three Population-Based Nordic Cohorts of Metastatic Colorectal Cancer

    BackgroundKRAS mutations, present in over 40% of metastatic colorectal cancer (mCRC), are negative predictive factors for anti-EGFR therapy. Mutations in KRAS-G12C have a cysteine residue for which drugs have been developed. Published data on this specific mutation are conflicting; thus, we studied the frequency and clinical characteristics in a real-world and population-based setting. MethodsPatients from three Nordic population-based cohorts and the real-life RAXO-study were combined. RAS and BRAF tests were performed in routine healthcare, except for one cohort. The dataset consisted of 2,559 patients, of which 1,871 could be accurately classified as KRAS, NRAS, and BRAF-V600E. Demographics, treatments, and outcomes were compared using logistic regression. Overall survival (OS) was estimated with Kaplan-Meier, and differences were compared using Cox regression, adjusted for baseline factors. ResultsThe KRAS-G12C frequency was 2%-4% of all tested in the seven cohorts (mean 3%) and 4%-8% of KRAS mutated tumors in the cohorts (mean 7%). Metastasectomies and ablations were performed more often (38% vs. 28%, p = 0.040), and bevacizumab was added more often (any line 74% vs. 59%, p = 0.007) for patients with KRAS-G12C- vs. other KRAS-mutated tumors, whereas chemotherapy was given to similar proportions. OS did not differ according to KRAS mutation, neither overall (adjusted hazard ratio (HR) 1.03; 95% CI 0.74-1.42, reference KRAS-G12C) nor within treatment groups defined as "systemic chemotherapy, alone or with biologics", "metastasectomy and/or ablations", or "best supportive care", RAS and BRAF wild-type tumors (n = 548) differed similarly to KRAS-G12C, as to other KRAS- or NRAS-mutated (n = 66) tumors. ConclusionsIn these real-life and population-based cohorts, there were no significant differences in patient characteristics and outcomes between patients with KRAS-G12C tumors and those with other KRAS mutations. This contrasts with the results of most previous studies claiming differences in many aspects, often with worse outcomes for those with a KRAS-G12C mutation, although not consistent. When specific drugs are developed, as for this mutation, differences in outcome will hopefully emerge.Peer reviewe

    The Duodenal Mucosal Bicarbonate Secretion : Role of Melatonin in Neurohumoral Control and Cellular Signaling

    The duodenal lumen is exposed to aggressive factors with a high potential to cause damage to the mucosa. Bicarbonate secretion by the duodenal mucosa is accepted as the primary important defense mechanism against the hydrochloric acid intermittently expelled from the stomach. The present thesis concerns the influence of the central nervous system and the effects of the hormone melatonin on bicarbonate secretion in anesthetized rats in vivo. Effects of melatonin on intracellular calcium signaling by duodenal enterocyte in vitro were examined in tissues of both human and rat origin. The main findings were as follows: Melatonin is a potent stimulant of duodenal mucosal bicarbonate secretion and also seems to be involved in the acid-induced stimulation of the secretion. Stimulation elicited in the central nervous system by the α1-adrenoceptor agonist phenylephrine induced release of melatonin from the intestinal mucosa and a four-fold increase in alkaline secretion. The melatonin antagonist luzindole abolished the duodenal secretory response to administered melatonin and to central nervous phenylephrine but did not influence the release of intestinal melatonin. Central nervous stimulation was also abolished by synchronous ligation of the vagal trunks and the sympathetic chains at the sub-laryngeal level. Melatonin induced release of calcium from intracellular stores and also influx of extracellular calcium in isolated duodenal enterocytes. Enterocytes in clusters functioned as a syncytium. Overnight fasting rapidly and profoundly down-regulated the responses to the duodenal secretagogues orexin-A and bethanechol but not those to melatonin or vasoactive intestinal polypeptide. In conclusion, the results strongly suggest that intestinal melatonin plays an important role in central nervous elicited stimulation of duodenal mucosal bicarbonate secretion. Sensitivity of this alkaline secretion to some peripheral stimulators markedly depends on the feeding status

    Industrial Sheet Metal Forming Simulation with Elastic Dies

    As part of the development process for new stamping dies, in the automotive sheet metal forming (SMF) industry, the majority of all forming operations are simulated with the Finite Element Method (FEM) before the dies are manufactured. Today, these simulations are conducted with rigid tools under the assumption that there are no tool deformations. However, research shows that tool deformations have an influence on the finished product. In real production these deformations are compensated by manual rework during the try-out. Additional reason for simulating with rigid dies is that there are non-existing simulation methods elaborated for elastic stamping dies. Also, simulation of elastic tools requires high computational power.     Since simulations today are performed with rigid stamping dies the purpose of this work is to investigate the conditions of how to conduct SMF-simulations with elastic stamping dies. The object that will be studied is a stamping die for a Volvo XC90 inner door used in a single-action press. This work is part of the development to minimize the manual rework, with the goal to compensate for tool deformations in a virtual environment.    Results for rigid stamping dies in LS-Dyna was compared to currently used AutoForm as a pre-study. A simple model was then created to find a suitable method while using elastic stamping dies. The developed method was used for an industrial size stamping die.     Since there are little amount of research performed on simulations using elastic stamping dies, elasticity and complexity were gradually introduced into the FE-model. As a first step, only the punch was included as an elastic solid. Secondly, the die was added. Finally, the entire die was simulated as elastic together with the hydraulic cushion of the press. When the FE-model worked as expected a suitable method for minimizing the simulation time with acceptable results was studied.     Comparisons of measured- and simulation results show a high correlation. To improve the results from the FE-model factors such as press deformations, advanced friction models, etc. should be included.    Conclusions from this work shows that it is possible to perform SMF-simulations with elastic stamping dies. As the computational time normally is high this work also presents a method first step to reduce the computational time with acceptable results. Comparisons between simulations with rigid and elastic stamping dies proves that there are significant differences in the outcome of the two methods. Reduced Lead Time through Advanced Die Structure Analysis - Vinnov
