    Dual Promoters Improve the Rescue of Recombinant Measles Virus in Human Cells

    Reverse genetics is a technology that allows the production of a virus from its complementary DNA (cDNA). It is a powerful tool for analyzing viral genes, the development of novel vaccines, and gene delivery vectors. The standard reverse genetics protocols are laborious, time-consuming, and inefficient for negative-strand RNA viruses. A new reverse genetics platform was established, which increases the recovery efficiency of the measles virus (MV) in human 293-3-46 cells. The novel features compared with the standard system involving 293-3-46 cells comprise (a) dual promoters containing the RNA polymerase II promoter (CMV) and the bacteriophage T7 promoter placed in uni-direction on the same plasmid to enhance RNA transcription; (b) three G nucleotides added just after the T7 promoter to increase the T7 RNA polymerase activity; and (c) two ribozymes, the hairpin hammerhead ribozyme (HHRz), and the hepatitis delta virus ribozyme (HDVrz), were used to cleavage the exact termini of the antigenome RNA. Full-length antigenome cDNA of MV of the wild type IC323 strain or the vaccine AIK-C strain was inserted into the plasmid backbone. Both virus strains were easily rescued from their respective cloned cDNA. The rescue efficiency increased up to 80% compared with the use of the standard T7 rescue system. We assume that this system might be helpful in the rescue of other human mononegavirales

    Photometric and Spectroscopic Analysis of Cool White Dwarfs with Trigonometric Parallax Measurements

    A photometric and spectroscopic analysis of 152 cool white dwarf stars is presented. The discovery of 7 new DA white dwarfs, 2 new DQ white dwarfs, 1 new magnetic white dwarf, and 3 weak magnetic white dwarf candidates, is reported, as well as 19 known or suspected double degenerates. The photometric energy distributions, the Halpha line profiles, and the trigonometric parallax measurements are combined and compared to model atmosphere calculations to determine the effective temperature and the radius of each object, and also to constrain the atmospheric composition. New evolutionary sequences with C/O cores with thin and thick hydrogen layers are used to derive masses and ages. We confirm the existence of a range in Teff between 5000 and 6000K where almost all white dwarfs have H-rich atmospheres. There is little evidence for mixed H/He dwarfs, with the exception of 2 He-rich DA stars, and 5 C2H white dwarfs which possibly have mixed H/He/C atmospheres. The DQ sequence terminates near 6500K, below which they are believed to turn into C2H stars. True DC stars slightly above this temperature are found to exhibit H-like energy distributions despite the lack of Halpha absorption. Attempts to interpret the chemical evolution show the problem to be complex. Convective mixing is necessary to account for the non-DA to DA ratio as a function of temperature. The presence of helium in cool DA stars, the existence of the non-DA gap, and the peculiar DC stars are also explained in terms of convective mixing, although our understanding of how this mechanism works needs to be revised. The oldest object in our sample is about 7.9 Gyr or 9.7 Gyr old depending on whether thin or thick hydrogen layer models are used.     Variety of symptoms and atypical clinical course of Crohn’s disease (CD) often create the need for additional diagnostic procedures. In the described case of woman with CD, there was a suspicion of coexistence of ovarian cancer. This issue is particularly important in patients treated with immunosuppressants and biological agents. The discussion focused on the usefulness of CA125 (cancer antigen 125, mucin 16) serum level estimation in clinical practice and draws attention to the possible reasons for the increase of its value which is not associated to ovarian cancer

    Heavy Element Nucleosynthesis in the Brightest Galactic Asymptotic Giant Branch stars

    We present updated calculations of stellar evolutionary sequences and detailed nucleosynthesis predictions for the brightest asymptotic giant branch (AGB) stars in the Galaxy with masses between 5Msun to 9Msun, with an initial metallicity of Z =0.02 ([Fe/H] = 0.14). In our previous studies we used the Vassiliadis & Wood mass-loss rate, which stays low until the pulsation period reaches 500 days after which point a superwind begins. Vassiliadis & Wood noted that for stars over 2.5Msun the superwind should be delayed until P ~ 750 days at 5Msun. We calculate evolutionary sequences where we delay the onset of the superwind to pulsation periods of P ~ 700-800 days in models of M = 5, 6, and 7Msun. Post-processing nucleosynthesis calculations show that the 6 and 7Msun models produce the most Rb, with [Rb/Fe] ~ 1 dex, close to the average of most of the Galactic Rb-rich stars ([Rb/Fe] ~ 1.4 plus or minus 0.8 dex). Changing the rate of the 22Ne + alpha reactions results in variations of [Rb/Fe] as large as 0.5 dex in models with a delayed superwind. The largest enrichment in heavy elements is found for models that adopt the NACRE rate of the 22Ne(a,n)25Mg reaction. Using this rate allows us to best match the composition of most of the Rb-rich stars. A synthetic evolution algorithm is then used to remove the remaining envelope resulting in final [Rb/Fe] of ~ 1.4 dex although with C/O ratios > 1.     Depressive and anxiety symptoms among patients with inflammatory bowel diseases

    Introduction. This study was conducted on a population of patients with inflammatory bowel disease (IBD) and was based on an assessment of the prevalence and severity of depression and anxiety symptoms in various clinical aspects. The psychological features of IBD patients are very important in the perception of symptoms, but crucial as triggers of IBD or as a releasing factor for IBD symptoms recurrence. Methods. The study included 130 patients with IBD, including 68 with Crohn’s disease (CD) and 62 with ulcerative colitis (UC). The severity of anxiety and depression symptoms were examined by the Hospital Anxiety and Depression Scale (HADS). Patients were also subjected to assessment of anthropometric attributes, clinical factors, quality of life, and symptoms of the disease, with dedicated clinical scales. Results. The occurrence of significant symptoms of anxiety was estimated at 45.5% in patients with CD and 30.5% in the UC patients. Significant depressive symptoms related to 20.5% of people with CD and 17.5% of patients with UC. The parameters of anxiety and depression showed significant associations with parameters of quality of life, BMI, and the scales describing the exacerbation of the diseases. Conclusions. The analyses did not reveal significant differences in the severity and prevalence of symptoms of anxiety and depression in subgroups with IBD. The expansion of relevant symptoms of anxiety and depression in this population was greater than in the general population. In addition, there was a significant correlation between parameters of HADS and clinical factors

    Hepatic abscess in a pre-existed simple hepatic cyst as a late complication of sigmoid colon ruptured diverticula: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Hepatic abscesses have been reported as a rare complication of diverticulitis of the bowel. This complication is recognized more commonly at the time of the diagnosis of diverticulitis, or ruptured diverticula, but also can be diagnosed prior to surgery, or postoperatively.</p> <p>Case presentation</p> <p>This report describes a man who developed an hepatic abscess within a simple hepatic cyst, two months after operation for ruptured diverticula of the sigmoid colon. The abscess was drained surgically and the patient made a complete recovery.</p> <p>Conclusion</p> <p>The development of an hepatic abscess in a pre-existing hepatic cyst, secondary to diverticulitis, is a rare complication. A high degree of clinical suspicion is required for immediate diagnosis and treatment.</p

    This report is the second part of a comprehensive two-part series aimed at reviewing an extensive and diverse toolkit of novel methods to explore brain health and function. While the first report focused on neurophotonic tools mostly applicable to animal studies, here, we highlight optical spectroscopy and imaging methods relevant to noninvasive human brain studies. We outline current state-of-the-art technologies and software advances, explore the most recent impact of these technologies on neuroscience and clinical applications, identify the areas where innovation is needed, and provide an outlook for the future directions
