    Monitoring of ivermectin residues in bovine and pork tissues

    Argentina is traditionally a beef-producing country. However, in recent years other productions have grown exponentially. Due to improvements in technology, changes in customs, or economic issues, consumer preferences have changed for cheaper meats. Animal production systems, both extensive and intensive, are closely linked to veterinary drug use for prevention, control, or treatment of parasitic diseases. Consequently, if good agricultural practices are not respected, products obtained from these productions could present residues above the Maximum Residue Limits (MRLs). In this context, ivermectin (IVM), an endectocide drug belonging to the macrocyclic lactone group, is one of the most widely used to treat parasitic diseases. IVM residues above the MRLs in bovine and pork tissues may constitute a risk to consumer health. Consequently, the current study aimed to assess the presence of ivermectin residues in bovine and pork tissues for local consumption in Buenos Aires province (Argentina). Samples of bovine/pork meat and fat, and bovine liver and kidney were taken for 3 years during the winter and summer seasons in 5 cities of Buenos Aires province. Tissue samples were analyzed by HPLC with a fluorescence detector. Of a total of 691 samples analyzed, 87 of them presented residues of IVM. However, only 13 samples from the positive ones showed concentrations above the Codex MRL. Mean IVM concentrations (range) were 42.18 (0.11?587.15), 31.66 (2.96?283.33), 162.61 (1.32?516.55), 22.78 (1.51?65.40), 15.26 (0.07?194.25) and 22.14 (1.58?126.76) ppb for bovine meat, bovine fat, bovine liver, bovine kidney, pork meat, and pork fat, respectively. In conclusion, bovine and pork tissue ingestion is safe for the consumer. However, 1.88% of the samples showed IVM concentrations above de MRL, thus the implementation of residue surveillance programs at the local level guaranteeing consumer health is strongly recommended.Fil: Cantón, Lucila.     Dietary geraniol by oral or enema administration strongly reduces dysbiosis and systemic inflammation in dextran sulfate sodium-treated mice

    (Trans)-3,7-Dimethyl-2,6-octadien-1-ol, commonly called geraniol (Ge-OH), is an acyclic monoterpene alcohol with well-known anti-inflammatory, antitumoral, and antimicrobial properties. It is widely used as a preservative in the food industry and as an antimicrobial agent in animal farming. The present study investigated the role of Ge-OH as an anti-inflammatory and anti-dysbiotic agent in the dextran sulfate sodium (DSS)-induced colitis mouse model. Ge-OH was orally administered to C57BL/6 mice at daily doses of 30 and 120 mg kg(-1) body weight, starting 6 days before DSS treatment and ending the day after DSS removal. Furthermore, Ge-OH 120 mg kg(-1) dose body weight was administered via enema during the acute phase of colitis to facilitate its on-site action. The results show that orally or enema-administered Ge-OH is a powerful antimicrobial agent able to prevent colitis-associated dysbiosis and decrease the inflammatory systemic profile of colitic mice. As a whole, Ge-OH strongly improved the clinical signs of colitis and significantly reduced cyclooxygenase-2 (COX-2) expression in colonocytes and in the gut wall. Ge-OH could be a powerful drug for the treatment of intestinal inflammation and dysbiosis

    A temporal record of microplastic pollution in Mediterranean seagrass soils

In the current study, we selected seagrass meadows, known to efficiently trap organic and inorganic particles, to investigate the concentrations and dynamics of microplastics in their soil. We assessed microplastic contamination and accumulation in 210Pb dated soil cores collected in Posidonia oceanica meadows at three locations along the Spanish Mediterranean coast, with two sites located in the Almería region (Agua Amarga and Roquetas) and one at Cabrera Island (Santa Maria). Almería is known for its intense agricultural industry with 30 000 ha of plastic-covered greenhouses, while the Cabrera Island is situated far from urban areas. Microplastics were extracted using enzymatic digestion and density separation. The particles were characterized by visual identification and with Fourier-transformed infrared (FTIR) spectroscopy, and related to soil age-depth chronologies. Our findings showed that the microplastic contamination and accumulation was negligible until the mid-1970s, after which plastic particles increased dramatically, with the highest concentrations of microplastic particles (MPP) found in the recent (since 2012) surface soil of Agua Amarga (3819 MPP kg−1), followed by the top-most layers of the soil of the meadows in Roquetas (2173 kg−1) and Santa Maria (68–362 kg−1). The highest accumulation rate was seen in the Roquetas site (8832 MPP m−2 yr−1). The increase in microplastics in the seagrass soil was associated to land-use change following the intensification of the agricultural industry in the area, with a clear relationship between the development of the greenhouse industry in Almería and the concentration of microplastics in the historical soil record. This study shows a direct linkage between intense anthropogenic activity, an extensive use of plastics and high plastic contamination in coastal marine ecosystems such as seagrass meadows. We highlight the need of proper waste management to protect the coastal environment from continuous pollution

    Evaluating the extent and impact of the extreme Storm Gloria on Posidonia oceanica seagrass meadows

    Extreme storms can trigger abrupt and often lasting changes in ecosystems by affecting foundational (habitat-forming) species. While the frequency and intensity of extreme events are projected to increase under climate change, its impacts on seagrass ecosystems remain poorly documented. In January 2020, the Spanish Mediterranean coast was hit by Storm Gloria, one of the most devastating recent climate events in terms of intensity and duration. We conducted rapid surveys of 42 Posidonia oceanica meadows across the region to evaluate the extent and type of impact (burial, unburial and uprooting). We investigated the significance of oceanographic (wave impact model), geomorphological (latitude, depth, exposure), and structural (patchiness) factors in predicting impact extent and intensity. The predominant impact of Storm Gloria was shoot unburial. More than half of the surveyed sites revealed recent unburial, with up to 40 cm of sediment removed, affecting over 50 % of the meadow. Burial, although less extensive, was still significant, with 10–80 % of meadow cover being buried under 7 cm of sediment, which is considered a survival threshold for P. oceanica. In addition, we observed evident signs of recently dead matte in some meadows and large amounts of detached drifting shoots on the sea bottom or accumulated as debris on the beaches. Crucially, exposed and patchy meadows were much more vulnerable to the overall impact than sheltered or continuous meadows. Given how slow P. oceanica is able to recover after disturbances, we state that it could take from decades to centuries for it to recoup its losses. Seagrass ecosystems play a vital role as coastal ecological infrastructure.     Optimization of universal allogeneic CAR-T cells combining CRISPR and transposon-based technologies for treatment of acute myeloid leukemia

    Despite the potential of CAR-T therapies for hematological malignancies, their efficacy in patients with relapse and refractory Acute Myeloid Leukemia has been limited. The aim of our study has been to develop and manufacture a CAR-T cell product that addresses some of the current limitations. We initially compared the phenotype of T cells from AML patients and healthy young and elderly controls. This analysis showed that T cells from AML patients displayed a predominantly effector phenotype, with increased expression of activation (CD69 and HLA-DR) and exhaustion markers (PD1 and LAG3), in contrast to the enriched memory phenotype observed in healthy donors. This differentiated and more exhausted phenotype was also observed, and corroborated by transcriptomic analyses, in CAR-T cells from AML patients engineered with an optimized CAR construct targeting CD33, resulting in a decreased in vivo antitumoral efficacy evaluated in xenograft AML models. To overcome some of these limitations we have combined CRISPR-based genome editing technologies with virus-free gene-transfer strategies using Sleeping Beauty transposons, to generate CAR-T cells depleted of HLA-I and TCR complexes (HLA-IKO/TCRKO CAR-T cells) for allogeneic approaches. Our optimized protocol allows one-step generation of edited CAR-T cells that show a similar phenotypic profile to non-edited CAR-T cells, with equivalent in vitro and in vivo antitumoral efficacy. Moreover, genomic analysis of edited CAR-T cells revealed a safe integration profile of the vector, with no preferences for specific genomic regions, with highly specific editing of the HLA-I and TCR, without significant off-target sites. Finally, the production of edited CAR-T cells at a larger scale allowed the generation and selection of enough HLA-IKO/TCRKO CAR-T cells that would be compatible with clinical applications. In summary, our results demonstrate that CAR-T cells from AML patients, although functional, present phenotypic and functional features that could compromise their antitumoral efficacy, compared to CAR-T cells from healthy donors. The combination of CRISPR technologies with transposon-based delivery strategies allows the generation of HLA-IKO/TCRKO CAR-T cells, compatible with allogeneic approaches, that would represent a promising option for AML treatment

    Impact of COVID-19 on cardiovascular testing in the United States versus the rest of the world

    Objectives: This study sought to quantify and compare the decline in volumes of cardiovascular procedures between the United States and non-US institutions during the early phase of the coronavirus disease-2019 (COVID-19) pandemic. Background: The COVID-19 pandemic has disrupted the care of many non-COVID-19 illnesses. Reductions in diagnostic cardiovascular testing around the world have led to concerns over the implications of reduced testing for cardiovascular disease (CVD) morbidity and mortality. Methods: Data were submitted to the INCAPS-COVID (International Atomic Energy Agency Non-Invasive Cardiology Protocols Study of COVID-19), a multinational registry comprising 909 institutions in 108 countries (including 155 facilities in 40 U.S. states), assessing the impact of the COVID-19 pandemic on volumes of diagnostic cardiovascular procedures. Data were obtained for April 2020 and compared with volumes of baseline procedures from March 2019. We compared laboratory characteristics, practices, and procedure volumes between U.S. and non-U.S. facilities and between U.S. geographic regions and identified factors associated with volume reduction in the United States. Results: Reductions in the volumes of procedures in the United States were similar to those in non-U.S. facilities (68% vs. 63%, respectively; p = 0.237), although U.S. facilities reported greater reductions in invasive coronary angiography (69% vs. 53%, respectively; p < 0.001). Significantly more U.S. facilities reported increased use of telehealth and patient screening measures than non-U.S. facilities, such as temperature checks, symptom screenings, and COVID-19 testing. Reductions in volumes of procedures differed between U.S. regions, with larger declines observed in the Northeast (76%) and Midwest (74%) than in the South (62%) and West (44%). Prevalence of COVID-19, staff redeployments, outpatient centers, and urban centers were associated with greater reductions in volume in U.S. facilities in a multivariable analysis. Conclusions: We observed marked reductions in U.S. cardiovascular testing in the early phase of the pandemic and significant variability between U.S. regions. The association between reductions of volumes and COVID-19 prevalence in the United States highlighted the need for proactive efforts to maintain access to cardiovascular testing in areas most affected by outbreaks of COVID-19 infection

    Lesioni funzionali della cuffia dei rotatori; ruolo del “rotator cable” e del “rotator crescent”

    OBIETTIVO DIDATTICO. Descrivere due importanti componenti anatomiche del tendine del sovraspinato, il “rotator cable” ed il “rotator crescent”, ed il loro ruolo nelle lesioni della cuffia. Valutare le importanti implicazioni biomeccaniche e cliniche attribuite a questa struttura e la correlazione negativa tra "lesione della cuffia" e quadro clinico in termini di dolore e impotenza funzionale. INTRODUZIONE. La cuffia dei rotatori è un complesso funzionale composto da muscoli e tendini che provvedono a mantenere stabile la spalla. Il tendine del sovraspinato ne rappresenta la componente principale, e nel suo contesto si trova uno spesso gruppo di fibre perpendicolari al suo asse, descritto per la prima volta da Clark e denominato “rotator cable” da Burkhart. Questa struttura circonda la zona distale del tendine del sovraspinato, una zona ipovascolare chiamata “rotator crescent” che facilmente va incontro a lesione. Il rotator cable gioca un ruolo centrale nella biomeccanica della spalla, ed è stato descritto come una struttura simile ad un cavo di supporto di un ponte in sospensione dove le forze sono trasferite dalla cuffia al cable, che funge da “stress shielding”. DESCRIZIONE. Per lesione funzionale di cuffia si intende una lesione valida dal punto di vista meccanico. L'azione di protezione del cable nei confronti del tendine distale ("crescent" - zona avascolare), frequente sede di lesioni, fa riflettere su una maggiore responsabilità biologica più che meccanica, nella patogenesi delle lesioni della cuffia. Alcuni studi sperimentali evidenzierebbero due diversi morfotipi di cuffia, a "Crescent" dominante" e a "Cable dominante" forse influenzati dall'età, nel tentativo di ridurre le sollecitazioni su un "crescent" più degenerato. CONCLUSIONI. Lesioni parziali articolari in posizione centrale e in pazienti “cable dominante” possono essere tollerate e rimanere stabili nel tempo perchè localizzate nella zona crescent protetta, grazie al cable, da tutte le sollecitazioni muscolari e/o meccaniche, mentre le lesioni a posizione eccentrica e/o in pazienti “crescent dominanti” l'evoluzione sarà più rapida e meno tollerata

    Dietary Supplementation with a Blend of Hydrolyzable and Condensed Tannins Ameliorates Diet-Induced Intestinal Inflammation in Zebrafish (<i>Danio rerio</i>)

    The current study evaluated the effects of hydrolyzable and condensed tannins from chestnut and quebracho wood, respectively (TSP, Silvafeed®), on zebrafish with intestinal inflammation induced by a plant-based diet (basal diet). Four experimental diets were prepared as follows: the basal diet + 0 TSP, the basal diet + TSP at 0.9 g/kg of feed, the basal diet + TSP at 1.7 g/kg of feed, and the basal diet + TSP at 3.4 g/kg of feed. Eighty-four zebrafish (Danio rerio) were fed for 12 days with the experimental diets. In zebrafish fed the basal diet, intestine integrity appeared to be altered, with damaged intestinal villi, high immunoexpression of tumor necrosis factor-α (TNFα) and cyclooxygenase 2 (COX2), and high expression of the cox2, interleukin 1 (il-1b), interleukin 8 (cxcl8-l1), and tnfα genes. The tannin treatment partially restored intestinal morphology and downregulated the expression of cytokines. The best activity was detected with 1.7 and 3.4 g/kg of feed. In the guts of all groups, Proteobacteria, Fusobacteria, Firmicutes, and Bacteroidetes were the most represented phyla. The most represented genera were Plesiomonas and Sphingomonas, belonging to the Proteobacteria phylum; Cetobacterium, belonging to the Fusobacteria phylum; and Lactobacillus, belonging to the Firmicutes phylum. No significant differences were detected among groups, except for a slight decrease in the Fusobacteria phylum and slight increases in the Shewanella and Bacteroides genera with TSP. In conclusion, these results suggest that tannins can improve the zebrafish intestinal inflammation caused by a terrestrial-plant-based diet in a dose-dependent manner
