Long-term Lactobacillus rhamnosus BMX 54 application to restore a balanced vaginal ecosystem: a promising solution against HPV-infection
Background: Over recent years, a growing interest has developed in microbiota and in the concept of maintaining a special balance between Lactobacillus and other bacteria species in order to promote women's well-being. The aim of our study was to confirm that vaginal Lactobacilli long-lasting implementation in women with HPV-infections and concomitant bacterial vaginosis or vaginitis might be able to help in solving the viral infection, by re-establishing the original eubiosis.Methods: A total of 117 women affected by bacterial vaginosis or vaginitis with concomitant HPV-infections were enrolled at Department of Gynecological Obstetrics and Urological Sciences, La Sapienza University, Rome, Italy between February 2015 and March 2016. Women were randomized in two groups, standard treatment (metronidazole 500 mg twice a day for 7 days or fluconazole 150 mg orally once a day for 2 consecutive days) plus short-term (3 months) vaginal Lactobacillus implementation (group 1, short probiotics treatment protocol group, n = 60) versus the same standard treatment plus long-lasting (6 months) vaginal Lactobacillus rhamnosus BMX 54 administration (group 2, treatment group, n = 57).Results: After a median follow up of 14 months (range 9-30 months) the chance to solve HPV-related cytological anomalies was twice higher in probiotic long-term users (group 2) versus short probiotics implementation group (group 1) (79.4% vs 37.5%, p = 0.041). Moreover, a total HPV-clearance was shown in 11.6% of short schedule probiotics implementation patients compared to a percentage of 31.2% in vaginal Lactobacilli long term users (p = 0.044), assessed as negative HPV-DNA test documented at the end of the study period.Conclusions: The consistent percentage of clearance of PAP-smear abnormalities and HPV-clearance obtained in long-term treatment group has been interestingly high and encouraging. Obviously, larger and randomized studies are warranted to confirm these encouraging results, but we believe that eubiosis re-establishment is the key to tackle effectively even HPV-infection
The sit up test to exhaustion as a test for muscular endurance evaluation
AIMS/HYPOTHESIS The aim of this study was to examine the sit up test to exhaustion as a field test for muscular endurance evaluation in a sample of sedentary people of both sexes.
METHODS:
A cross-sectional study was performed. Three-hundred-eighty-one participants volunteered for the study (28.5 \ub1 10.0 years; 168.2 \ub1 8.9 cm; 65.1 \ub1 11.1 kg), of which 194 males (27.5 \ub1 10.2 years; 173.6 \ub1 7.0 cm; 71.2 \ub1 5.2 kg) and 187 females (29.6 \ub1 10.1 years; 162.6 \ub1 7.1 cm; 58.7 \ub1 8.9 kg). Each subject voluntarily and randomly performed: a sit up test (SUT), a push up test (PUT), and a free weight squat test (ST), all till exhaustion. A multiple regression analysis was adopted for data analysis. Subsequently a percentile model for muscle endurance was developed. The 25th, 50th, and 75th percentile were identified as upper limit for low muscular endurance, average muscular endurance, and lower limit for high muscular endurance, respectively.
RESULTS:
Considering the sit up test as the dependent variable, the coefficients (R(2) = 0.23; r = 0.49; p < 0.001), and (R(2) = 0.31; r = 0.57; p < 0.001) emerged from a multiple regression analysis applied with respect to the push up test and the squat test, respectively. Gender stratification showed regression coefficients of (R(2) = 0.19; r = 0.44; p < 0.001) for SUT vs. PUT, and (R(2) = 0.30; r = 0.56; p < 0.001) for SUT vs. ST in male; and (R(2) = 0.23; r = 0.49; p < 0.001) for SUT vs. PUT, and (R(2) = 0.34; r = 0.59; p < 0.001) for SUT vs. ST in female.
CONCLUSIONS/INTERPRETATION:
The SUT showed low inter-relation with the other proposed tests indicating that the adoption of a single test for the global evaluation of muscle endurance is not the optimal approach. Moreover, the SUT was found to be inexpensive, safe, and appropriate for core muscle endurance measurement for both male and female
Novel Penicillin-Type Analogues Bearing a Variable Substituted 2-Azetidinone Ring at Position 6: Synthesis and Biological Evaluation
The synthesis and the biological activity of novel semi-synthetic β-lactam compounds containing an azetidinone moiety joined to the amino-nitrogen of the (+)-6-aminopenicillanic acid (6-APA) as new antibacterial agents is reported. The synthesized compounds were screened for their in vitro antimicrobial activity against a panel of Gram positive and Gram negative pathogens and environmental bacteria. Tested compounds displayed good antimicrobial activity against all tested Gram positive bacteria and for Staphylococcus aureus and Staphylococcus epidermidis antimicrobial activity resulted higher than that of the reference antibiotic. Additionally, in vitro cytotoxic screening was also carried out indicating that the compounds do not cause a cell vitality reduction effective at concentration next to and above those shown to be antimicrobial

Use of cactus pear pruning waste to improve soil properties and to produce high-quality compost
Bio-fertilizers could be a possible solution to help manage bio-waste problems and to maintain soil health conditions, especially in organic farms. Pruning of cactus pear produces from 6 to 10 tons/yearly of cladodes per hectare in specialized Italian orchards, which represents waste and a cost for disposal to farmers. Therefore, the aim of the work was to investigate the effects on soil physical properties, microflora activity, and basil plant growth of powdered pruning waste from cactus pear incorporated in the soil (10%, 20%, 30% 40% w/w). Moreover, we studied a dynamic composting process from fresh cladodes to produce stabilized end-products at the farm level. Our studies demonstrated that holding water ability and bulk density of soil were ameliorated by supplementing dried cladodes. Thus, gravitational and gravimetric water was positively correlated with the increase of added dried cladode in the soil (0.1% and 6.2 g H2O +/- 0.3 in samples of 40% mixed soil versus 68% and 3.3 g H2O +/- 0.3 in control). Furthermore, bulk density was reduced limiting soil compaction. Preliminary results on microbial activity suggested a possible selection/inhibition of some bacterial strains correlated with the increment of dry cladode supplementation. The basil plants grown in 20% mixed soil showed an increased biomass when compared to the control (+ 21%). Finally, raw cladode proved to be an excellent substrate for aerobic fermentation providing a final product of good quality and high moisture content (65%). These results are particularly relevant in organic agriculture where bio-fertilizers are recommended as economical, eco-friendly solution accessible also to marginal and small farms
Soluble beta amyloid evokes alteration in brain norepinephrine levels: role of nitric oxide and interleukin-1
Strong evidence showed neurotoxic properties of beta amyloid (Aβ) and its pivotal role in the Alzheimer's disease (AD) pathogenesis. Beside, experimental data suggest that Aβ may have physiological roles considering that such soluble peptide is produced and secreted during normal cellular activity. There is now suggestive evidence that neurodegenerative conditions, like AD, involve nitric oxide (NO) in their pathogenesis. Nitric oxide also possess potent neuromodulatory actions in brain regions, such as prefrontal cortex (PFC), hippocampus (HIPP), and nucleus accumbens (NAC). In the present study, we evaluated the effect of acute Aβ injection on norepinephrine (NE) content before and after pharmacological manipulations of nitrergic system in above mentioned areas. Moreover, effects of the peptide on NOS activity were evaluated. Our data showed that 2 h after i.c.v. soluble Aβ administration, NE concentrations were significantly increased in the considered areas along with increased iNOS activity. Pre-treatment with NOS inhibitors, 7-Nitroindazole (7-NI), and N6-(1-iminoethyl)-L-lysine-dihydrochloride (L-NIL), reversed Aβ-induced changes. Ultimately, pharmacological block of interleukin1 (IL-1) receptors prevented NE increase in all brain regions. Taken together our findings suggest that NO and IL-1 are critically involved in regional noradrenergic alterations induced by soluble Aβ injection
Genetic investigations on 8 patients affected by ring 20 chromosome syndrome
<p>Abstract</p> <p>Background</p> <p>Mosaic Chromosome 20 ring [r(20)] is a chromosomal disorder associated with a rare syndrome characterized by a typical seizure phenotype, a particular electroclinical pattern, cognitive impairment, behavioural problems and absence of a consistent pattern of dysmorphology. The pathogenic mechanism underlying seizures disorders in r(20) syndrome is still unknown. We performed a detailed clinical and genetic study on 8 patients with r(20) chromosome, aimed at detecting the genetic mechanism underlying r(20) syndrome.</p> <p>Methods</p> <p>We submitted 8 subjects with a previous diagnosis of ring 20 chromosome mosaicism to a clinical re-evaluation, followed by cytogenetic, FISH, array-CGH and molecular analyses. The genetic study was also extended to their available parents.</p> <p>Results</p> <p>FISH and array-CGH experiments indicate that cryptic deletions on chromosome 20 are not the cause of the r(20) chromosome associated disease. Moreover, no evidence of chromosome 20 uniparental disomy was found. Analysis of FISH signals given by variant in size alphoid tandem repeats probes on the normal chromosome 20 and the r(20) chromosome in the mosaic carriers suggests that the r(20) chromosome is the same chromosome not circularized in the "normal" cell line.</p> <p>Conclusions</p> <p>Higher percentages of r(20) chromosome cells were observed to be related with precocious age at seizure onset and with resistance to antiepileptic drug treatment. Behavioural problems also seem to be associated with higher percentages of r(20) chromosome cells. Our results suggest that an epigenetic mechanism perturbing the expression of genes close to the telomeric regions, rather than deletion of genes located at the distal 20p and/or 20q regions, may underlie the manifestation of r(20) syndrome.</p
Partial T cell defects and expanded CD56bright NK cells in an SCID patient carrying hypomorphic mutation in the IL2RG gene
X-linked severe combined immunodeficiency (X-SCID) caused by full mutation of the IL2RG gene leads to T- B+ NK- phenotype and is usually associated with severe opportunistic infections, diarrhea, and failure to thrive. When IL2RG hypomorphic mutation occurs, diagnosis could be delayed and challenging since only moderate reduction of T and NK cells may be present. Here, we explored phenotypic insights and the impact of the p.R222C hypomorphic mutation (IL2RGR222C ) in distinct cell subsets in an 8-month-old patient with atypical X-SCID. We found reduced CD4+ T cell counts, a decreased frequency of naĂŻve CD4+ and CD8+ T cells, and an expansion of B cells. Ex vivo STAT5 phosphorylation was impaired in CD4+ CD45RO+ T cells, yet compensated by supraphysiological doses of IL-2. Sanger sequencing on purified cell subsets showed a partial reversion of the mutation in total CD3+ cells, specifically in recent thymic emigrants (RTE), effector memory (EM), and CD45RA+ terminally differentiated EM (EMRA) CD4+ T cells. Of note, patient's NK cells had a normal frequency compared to age-matched healthy subjects, but displayed an expansion of CD56bright cells with higher perforin content and cytotoxic potential, associated with accumulation of NK-cell stimulatory cytokines (IL-2, IL-7, IL-15). Overall, this report highlights an alteration in the NK-cell compartment that, together with the high disease-phenotype variability, should be considered in the suspicion of X-SCID with hypomorphic IL2RG mutation
Impact of Foods and Dietary Supplements Containing Hydroxycinnamic Acids on Cardiometabolic Biomarkers: A Systematic Review to Explore Inter-Individual Variability
Plant-based diets rich in bioactive compounds such as polyphenols have been shown to positively modulate the risk of cardiometabolic (CM) diseases. The inter-individual variability in the response to these bioactives may affect the findings. This systematic review aimed to summarize findings from existing randomized clinical trials (RCTs) evaluating the effect of hydroxycinnamic acids (HCAs) on markers of CM health in humans. Literature searches were performed in PubMed and the Web of Science. RCTs on acute and chronic supplementation of HCA-rich foods/extracts on CM biomarkers were included. Forty-four RCTs (21 acute and 23 chronic) met inclusion criteria. Comparisons were made between RCTs, including assessments based on population health status. Of the 44 RCTs, only seven performed analyses on a factor exploring inter-individual response to HCA consumption. Results demonstrated that health status is a potentially important effect modifier as RCTs with higher baseline cholesterol, blood pressure and glycaemia demonstrated greater overall effectiveness, which was also found in studies where specific subgroup analyses were performed. Thus, the effect of HCAs on CM risk factors may be greater in individuals at higher CM risk, although future studies in these populations are needed, including those on other potential determinants of inter-individual variability.
