    How to build a functional connectomic biomarker for mild cognitive impairment from source reconstructed MEG resting-state activity: the combination of ROI representation and connectivity estimator matters

    Our work aimed to demonstrate the combination of machine learning and graph theory for the designing of a connectomic biomarker for mild cognitive impairment (MCI) subjects using eyes-closed neuromagnetic recordings. The whole analysis based on source-reconstructed neuromagnetic activity. As ROI representation, we employed the principal component analysis (PCA) and centroid approaches. As representative bi-variate connectivity estimators for the estimation of intra and cross-frequency interactions, we adopted the phase locking value (PLV), the imaginary part (iPLV) and the correlation of the envelope (CorrEnv). Both intra and cross-frequency interactions (CFC) have been estimated with the three connectivity estimators within the seven frequency bands (intra-frequency) and in pairs (CFC), correspondingly. We demonstrated how different versions of functional connectivity graphs single-layer (SL-FCG) and multi-layer (ML-FCG) can give us a different view of the functional interactions across the brain areas. Finally, we applied machine learning techniques with main scope to build a reliable connectomic biomarker by analyzing both SL-FCG and ML-FCG in two different options: as a whole unit using a tensorial extraction algorithm and as single pair-wise coupling estimations. We concluded that edge-weighed feature selection strategy outperformed the tensorial treatment of SL-FCG and ML-FCG. The highest classification performance was obtained with the centroid ROI representation and edge-weighted analysis of the SL-FCG reaching the 98% for the CorrEnv in α1:α2 and 94% for the iPLV in α2. Classification performance based on the multi-layer participation coefficient, a multiplexity index reached 52% for iPLV and 52% for CorrEnv. Selected functional connections that build the multivariate connectomic biomarker in the edge-weighted scenario are located in default-mode, fronto-parietal and cingulo-opercular network. Our analysis supports the notion of analysing FCG simultaneously in intra and cross-frequency whole brain interactions with various connectivity estimators in beamformed recordings

    O projeto de implantação da indústria automotiva no Brasil: por uma abordagem sob a ótica da teoria dos stakeholders

    A indústria automotiva representa um marco na história da industrialização brasileira. Com a sua realização, na gestão de Juscelino Kubitschek, o Brasil implantou a produção nacional de veículos. Este trabalho apresenta uma contribuição inicial para a busca de integração de duas áreas com pesquisas ainda incipientes no universo dos estudos organizacionais: gerenciamento de projetos e história da gestão. O objetivo é descrever e analisar, à luz da teoria dos stakeholders, a criação da indústria automotiva no Brasil. O estudo parte das ações relacionadas às tentativas de desenvolvimento e industrialização do Brasil pós-Segunda Guerra Mundial, que resultaram no Plano de Metas, o qual teve sua execução iniciada no governo de Juscelino Kubitscheck. Com dados documentais e uma entrevista em profundidade, foram apresentadas a formação do Grupo Executivo da Indústria Automotiva (GEIA) e suas ações de 1956 a 1960. Na análise das partes interessadas, constatou-se a existência de stakeholders opositores de alto poder, como as três maiores empresas multinacionais do setor e a própria equipe econômica do governo. Por outro lado, o Presidente JK era o stakeholder entusiasta. A estrutura do GEIA representou um marco na formação da visão tecnocrata brasileira, com uma equipe enxuta, ágil, com poder decisório e regras preestabelecidas

    Vocal caricatures reveal signatures of speaker identity

    What are the features that impersonators select to elicit a speaker’s identity? We built a voice database of public figures (targets) and imitations produced by professional impersonators. They produced one imitation based on their memory of the target (caricature) and another one after listening to the target audio (replica). A set of naive participants then judged identity and similarity of pairs of voices. Identity was better evoked by the caricatures and replicas were perceived to be closer to the targets in terms of voice similarity. We used this data to map relevant acoustic dimensions for each task. Our results indicate that speaker identity is mainly associated with vocal tract features, while perception of voice similarity is related to vocal folds parameters. We therefore show the way in which acoustic caricatures emphasize identity features at the cost of loosing similarity, which allows drawing an analogy with caricatures in the visual space.Fil: Lopez, Sabrina Laura.     Modulation of Cell Surface Receptor Expression by Modified Vaccinia Virus Ankara in Leukocytes of Healthy and HIV-Infected Individuals

    Viral vectors are increasingly used as delivery means to induce a specific immunity in humans and animals. However, they also impact the immune system, and it depends on the given context whether this is beneficial or not. The attenuated vaccinia virus strain modified vaccinia virus Ankara (MVA) has been used as a viral vector in clinical studies intended to treat and prevent cancer and infectious diseases. The adjuvant property of MVA is thought to be due to its capability to stimulate innate immunity. Here, we confirmed that MVA induces interleukin-8 (IL-8), and this chemokine was upregulated significantly more in monocytes and HLA-DR(bright)dendritic cells (DCs) of HIV-infected patients on combined antiretroviral therapy (ART) than in cells of healthy persons. The effect of MVA on cell surface receptors is mostly unknown. Using mass cytometry profiling, we investigated the expression of 17 cell surface receptors in leukocytes afterex vivoinfection of human whole-blood samples with MVA. We found that MVA downregulates most of the characteristic cell surface markers in particular types of leukocytes. In contrast, C-X-C motif chemokine receptor 4 (CXCR4) was significantly upregulated in each leukocyte type of healthy persons. Additionally, we detected a relative higher cell surface expression of the HIV-1 co-receptors C-C motif chemokine receptor 5 (CCR5) and CXCR4 in leukocytes of HIV-ART patients than in healthy persons. Importantly, we showed that MVA infection significantly downregulated CCR5 in CD4+ T cells, CD8+ T cells, B cells, and three different DC populations. CD86, a costimulatory molecule for T cells, was significantly upregulated in HLA-(DRDCs)-D-bright after MVA infection of whole blood from HIV-ART patients. However, MVA was unable to downregulate cell surface expression of CD11b and CD32 in monocytes and neutrophils of HIV-ART patients to the same extent as in monocytes and neutrophils of healthy persons. In summary, MVA modulates the expression of many different kinds of cell surface receptors in leukocytes, which can vary in cells originating from persons previously infected with other pathogens

However, for its success, an effective communication is essential. It is understood by any kind of information transmission. In this sense, it becomes crucial to understand how communications work in Brazilian projects and how these are perceived by the communications manager Brazilian projects. The present study aims to understand how the communication is made in Brazilian projects and how the project manager realizes this area of knowledge. The results indicate the strategic view, the process view, the objectives of the manager’s objectives with communication, the adherence to best practice guides, the barriers and what is not done in communications management.A nova configuração competitiva do mercado corporativo obriga as empresas a buscarem soluções inovadoras para sua própria sobrevivência. Nesse contexto ganha importância dentro das organizações o gerenciamento de projetos, uma vez que qualquer mudança substancial ou incremental em uma organização seja lançamento de produtos, reestruturações, fusões ou aquisições se dá por meio de projetos. Contudo, para o sucesso de um projeto, é importante que haja uma comunicação eficaz. Entenda-se por comunicação qualquer tipo de transmissão de informações. Neste sentido, torna-se fundamental compreender como funcionam as comunicações nos projetos brasileiros e como essas comunicações são percebidas pelo gerente de projetos brasileiro. O presente trabalho buscou compreender como o é feita a comunicação nos projetos brasileiros e como o gerente de projetos percebe essa área do conhecimento.     Antiproliferative Activity And Induction Of Apoptosis In Pc-3 Cells By The Chalcone Cardamonin From Campomanesia Adamantium (myrtaceae) In A Bioactivity-guided Study.

    The Myrtaceae family is a common source of medicines used in the treatment of numerous diseases in South America. In Brazil, fruits of the Campomanesia species are widely used to make liqueurs, juices and sweets, whereas leaves are traditionally employed as a medicine for dysentery, stomach problems, diarrhea, cystitis and urethritis. Ethanol extracts of Campomanesia adamantium (Myrtaceae) leaves and fruits were evaluated against prostate cancer cells (PC-3). The compound (2E)-1-(2,4-dihydroxy-6-methoxyphenyl)-3-phenylprop-2-en-1-one, cardamonin) was isolated from ethanol extracts of C. adamantium leaves in a bioactivity-guided study and quantified by UPLC-MS/MS. In vitro studies showed that the isolated chalcone cardamonin inhibited prostate cancer cell proliferation and decreased the expression of NFkB1. Moreover, analysis by flow cytometry showed that this compound induced DNA fragmentation, suggesting an effect on apoptosis induction in the PC-3 cell line.191843-5

    Differential effects of the second SARS-CoV-2 mRNA vaccine dose on T cell immunity in naive and COVID-19 recovered individuals

    The rapid development of mRNA-based vaccines against the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) led to the design of accelerated vaccination schedules that have been extremely effective in naive individuals. While a two-dose immunization regimen with the BNT162b2 vaccine has been demonstrated to provide a 95% efficacy in naive individuals, the effects of the second vaccine dose in individuals who have previously recovered from natural SARS-CoV-2 infection has not been investigated in detail. In this study, we characterize SARS-CoV-2 spike-specific humoral and cellular immunity in naive and previously infected individuals during and after two doses of BNT162b2 vaccination. Our results demonstrate that, while the second dose increases both the humoral and cellular immunity in naive individuals, COVID-19 recovered individuals reach their peak of immunity after the first dose.     Juxtaposing BTE and ATE – on the role of the European insurance industry in funding civil litigation

    One of the ways in which legal services are financed, and indeed shaped, is through private insurance arrangement. Two contrasting types of legal expenses insurance contracts (LEI) seem to dominate in Europe: before the event (BTE) and after the event (ATE) legal expenses insurance. Notwithstanding institutional differences between different legal systems, BTE and ATE insurance arrangements may be instrumental if government policy is geared towards strengthening a market-oriented system of financing access to justice for individuals and business. At the same time, emphasizing the role of a private industry as a keeper of the gates to justice raises issues of accountability and transparency, not readily reconcilable with demands of competition. Moreover, multiple actors (clients, lawyers, courts, insurers) are involved, causing behavioural dynamics which are not easily predicted or influenced. Against this background, this paper looks into BTE and ATE arrangements by analysing the particularities of BTE and ATE arrangements currently available in some European jurisdictions and by painting a picture of their respective markets and legal contexts. This allows for some reflection on the performance of BTE and ATE providers as both financiers and keepers. Two issues emerge from the analysis that are worthy of some further reflection. Firstly, there is the problematic long-term sustainability of some ATE products. Secondly, the challenges faced by policymakers that would like to nudge consumers into voluntarily taking out BTE LEI
