Proteomic analysis of Chromobacterium violaceum and its adaptability to stress
Chromobacterium violaceum (C. violaceum) occurs abundantly in a variety of ecosystems, including ecosystems that place the bacterium under stress. This study assessed the adaptability of C. violaceum by submitting it to nutritional and pH stresses and then analyzing protein expression using bi-dimensional electrophoresis (2-DE) and Maldi mass spectrometry. Chromobacterium violaceum grew best in pH neutral, nutrient-rich medium (reference conditions); however, the total protein mass recovered from stressed bacteria cultures was always higher than the total protein mass recovered from our reference culture. The diversity of proteins expressed (repressed by the number of identifiable 2-DE spots) was seen to be highest in the reference cultures, suggesting that stress reduces the overall range of proteins expressed by C. violaceum. Database comparisons allowed 43 of the 55 spots subjected to Maldi mass spectrometry to be characterized as containing a single identifiable protein. Stress-related expression changes were noted for C. violaceum proteins related to the previously characterized bacterial proteins: DnaK, GroEL-2, Rhs, EF-Tu, EF-P; MCP, homogentisate 1,2-dioxygenase, Arginine deiminase and the ATP synthase β-subunit protein as well as for the ribosomal protein subunits L1, L3, L5 and L6. The ability of C. violaceum to adapt its cellular mechanics to sub-optimal growth and protein production conditions was well illustrated by its regulation of ribosomal protein subunits. With the exception of the ribosomal subunit L3, which plays a role in protein folding and maybe therefore be more useful in stressful conditions, all the other ribosomal subunit proteins were seen to have reduced expression in stressed cultures. Curiously, C. violeaceum cultures were also observed to lose their violet color under stress, which suggests that the violacein pigment biosynthetic pathway is affected by stress. Analysis of the proteomic signatures of stressed C. violaceum indicates that nutrient-starvation and pH stress can cause changes in the expression of the C. violaceum receptors, transporters, and proteins involved with biosynthetic pathways, molecule recycling, energy production. Our findings complement the recent publication of the C. violeaceum genome sequence and could help with the future commercial exploitation of C. violeaceum
Low-frequency physical variations in the coastal zone of Ubatuba, northern coast of São Paulo State, Brazil
Sea level (SL), wind, air temperature (AT) and sea surface temperature (SST) variations in the coastal region of Ubatuba, northern coast of São Paulo, are assessed. A Lanczos-square cosine filter, with a 40-hour window, was applied over the SL time series between 1978 and 2000, except for the period comprising 1984 to 1986. In order to study subtidal effects on mean sea level (MSL), SL numerical filtering indicated that there was a virtually complete removal of semidiurnal and diurnal astronomical tidal components over the period of study. Results indicated an average raw SL rise of 2.3 mm/year, whereas average filtered MSL was of the order of 0.7 mm/year. Despite the overall positive MSL trend, the lunar nodal cycle of 18.6 years seemed to be the explanation for the SL series pattern. Correlations between MSL and parallel wind had a maximum correlation coefficient around 0.6, with 99% statistical confidence, while MSL and perpendicular wind correlations were not statistically significant. These results may be explained by Ekman dynamics. Data records of AT and SST between 1990 and 2003 showed positive trends for both variables. During this period, AT rose about 0.087 ºC /year for the raw series and 0.085 ºC /year for the monthly time series, and SST showed a rise of 0.047 ºC /year and 0.046 ºC/year, for the raw and monthly time series, respectively. The monthly climatology for both AT and SST showed higher values in February with 27.79 ºC and 28.59 ºC for AT and SST, respectively, and the lowest in July with 21.12 ºC for AT and 21.91 ºC for SST
Evaluation of the genetic diversity of Histoplasma capsulatum var. capsulatum isolates from north-eastern Brazil
Since the beginning of the HIV epidemic, there has been a significant increase in the number of histoplasmosis cases in Ceara, a state in north-east Brazil. The lack of epidemiological data on the genotypes circulating in the north-east region shows the importance of more detailed studies on the molecular epidemiology of Histoplasma capsulatum var. capsulatum in this region. Different molecular techniques have been used to better characterize the genetic profile of H. capsulatum var. capsulatum strains. The aim of this study was to analyse the genetic diversity of H. capsulatum var. capsulatum isolates in Fortaleza, the capital of Ceara, through the sequencing of the internal transcribed spacer (ITS)1-5.8S-ITS2 region, and establish the molecular profile of these isolates, along with strains from south-east Brazil, by RAPD analysis, featuring the different clusters in those regions. The isolates were grouped into two clusters. Cluster 1 included strains from the south-east and north-east regions with separation of isolates into three distinct subgroups (subgroups 1a, 1 b and 1 c). Cluster 2 included only samples from north-east Brazil. Sequencing of the ITS1 -5.8S-ITS2 region allowed the detection of two major clades, which showed geographical correlation between them and their subgroups. Therefore, it can be concluded that the H. capsulatum var. capsulatum isolates from Ceara have a high degree of genetic polymorphism.
