Recherche et analyse d'étoiles naines blanches chimiquement stratiées dans le Sloan Digital Sky Survey
Ce mémoire présente une recherche détaillée et une analyse des étoiles naines blanches hybrides chimiquement stratifiées dans le Sloan Digital Sky Survey (SDSS). Une seule étoile stratifiée, PG 1305-017, était connue avant notre recherche. L'objectif principal est de confirmer l'existence de plusieurs nouvelles étoiles stratifiées. Pour ce faire, il a fallu dans un premier temps développer une nouvelle génération de modèles d'atmosphère à partir de ceux de Bergeron et al. (1991) et Tremblay & Bergeron (2009). Nous y avons ajouté l'opacité de toutes les raies d'hélium et les calculs nécessaires pour tenir compte de la stratification chimique de l'atmosphère, où une mince quantité d’hydrogène flotte en équilibre diffusif au-dessus d’une enveloppe massive d’hélium. En parallèle, nous avons aussi calculé des modèles standards, chimiquement homogènes. Ensuite, nous avons sélectionné des naines blanches chaudes (Teff > 30,000 K) de type spectral hybride (traces d'hélium et d'hydrogène) parmi les ~38,000 naines blanches répertoriées dans le SDSS. Un total de 52 spectres d'étoile a été retenu dans notre échantillon final. La technique spectroscopique, c'est-à-dire l'ajustement des raies spectrales des modèles sur un spectre observé, a été appliquée à toutes les étoiles de notre échantillon. Nous avons ainsi mesuré la température effective, la gravité de surface et la composition chimique de l'atmosphère de ces étoiles. Par l'ajustement simultané de modèles stratifiés et homogènes, nous avons aussi pu déterminer si les étoiles étaient stratifiées ou non. Nous identifions ainsi 14 naines blanches stratifiées. Nous tirons de ces résultats plusieurs conclusions sur les processus physiques expliquant la présence d'hélium dans l'atmosphère.We present a detailed research and analysis of chemically stratified hybrid white dwarf stars in the Sloan Digital Sky Survey (SDSS). Only one stratified star, PG 1305-017, was known before our analysis. The main objective is to confirm the existence of several new stratified stars. To do so, we initially had to develop a new generation of model atmospheres from those of Bergeron et al. (1991) and Tremblay & Bergeron (2009). We added the opacity of all the helium lines and the necessary calculations to account for the chemical stratification of the atmosphere, where a thin hydrogen layer floats in diffusive equilibrium on top of a more massive helium layer. In parallel, we also calculated a grid of standard models, which are chemically homogeneous. Then, we selected hot white dwarfs (Teff > 30,000 K) with a hybrid spectral type (traces of helium and hydrogen) from the ~38,000 white dwarfs listed in the SDSS. A total of 52 spectra were retained in our final sample. The spectroscopic technique, i.e. the fit of model spectra to an observed spectrum, was applied to all stars in our sample. Thereby, we have measured the effective temperature, the surface gravity and the chemical composition of these stars. By simultaneously fitting stratified and homogeneous models, we have also been able to determine if the stars were stratified or not. We identify 14 stratified white dwarfs. From these results, we draw several conclusions on the physical processes explaining the presence of helium in the atmosphere
La conception d’un répertoire d’outils didactiques pour soutenir la lecture d’œuvres littéraires complètes en formation générale des adultes
Cet essai professionnel porte sur la conception de matériel pédagogique destiné à la clientèle principalement jeune adulte et issue de l’immigration des centres d’éducation des adultes en région urbain et périurbaine. Le matériel pédagogique a été conçu pour répondre aux besoins des élèves et des personnes enseignantes qui suivent ou donnent le cours FRA-5203-2. Il concerne précisément le développement de la compétence à lire des œuvres littéraires complètes en amont de la rédaction d’une critique littéraire, rédaction qui sert d’épreuve ministérielle à ce niveau. Le premier chapitre présente les différents contextes qu’il fallait caractériser pour bien en saisir les enjeux : la clientèle ciblée, le contexte d’enseignement particulier de la formation générale des adultes et le contexte programmatique du cours FRA-52302-2. Dans le cadre de référence, la théorie de la complexité de Morin (2000, 2005) a été mise à contribution afin de lier diverses théories, comme celles relatives à la lecture littéraire, au sujet lecteur, aux conditions motivationnelles de Viau (2009), à la spécificité de la lecture d’œuvres littéraires complètes et à la différenciation pédagogique inclusive. L’appui sur ces théories a été opérationnalisé à travers des éléments empruntés à l’approche pédagogique par projet, à l’enseignement stratégique et au dispositif didactique du carnet de lecture. Cette opérationnalisation est encadrée par une méthodologie qui a mené à la création d’une grille critériée ayant servi à la fois à l’évaluation du matériel existant pour le cours FRA-5203-2, à notre conception du matériel pédagogique et à la rétroaction qu’en ont faite les personnes enseignantes qui ont été consultées. Ces rétroactions ont mené à des modifications du matériel, présenté en annexe. En guise de conclusion, nous avons souligné les retombées positives de l’essai sur le développement de nos compétences professionnelles, que ce soit en lien avec la collaboration avec nos pairs, avec l’engagement dans le développement professionnel, avec notre capacité à réfléchir aux savoirs enseignés où aux finalités de l’acte d’évaluer
An evaluation of the stimulants and impediments to innovation within PFI/PPP projects
This paper identifies the theoretical stimulants and impediments associated with the implementation of PFI/PPP (Private Finance Initiative/Public Private Partnership) projects. A current defect of this procurement approach is the unintentional constraint upon the innovations incorporated into the development of PFI projects. A critical evaluation of the published literature has been utilized to synthesize a theoretical model. The paper proposes a theoretical model for the identification of potential innovation stimulants and impediments within this type of procurement. This theoretical model is then utilised to evaluate four previously completed PFI projects. These project case-studies have been examined in detail. The evaluation demonstrates how ineffective current procedures are. The application of this model before project letting could eliminate unintentional constraints and stimulate improved innovation within the process. The implementation of the model could improve the successful delivery of innovation within the entire PFI/PPP procurement process
The biogeography of the caribou lungworm, Varestrongylus eleguneniensis (Nematoda: Protostrongylidae) across northern North America
Varestrongylus eleguneniensis (Nematoda; Protostrongylidae) is a recently described species of lungworm that infects caribou (Rangifer tarandus), muskoxen (Ovibos moschatus) and moose (Alces americanus) across northern North America. Herein we explore the geographic distribution of V. eleguneniensis through geographically extensive sampling and discuss the biogeography of this multi-host parasite. We analyzed fecal samples of three caribou subspecies (n = 1485), two muskox subspecies (n = 159), and two moose subspecies (n = 264) from across northern North America. Protostrongylid dorsal-spined larvae (DSL) were found in 23.8%, 73.6%, and 4.2% of these ungulates, respectively. A portion of recovered DSL were identified by genetic analyses of the ITS-2 region of the nuclear rDNA or the cytochrome oxidase c subunit I (COI) region of the mtDNA. We found V. eleguneniensis widely distributed among caribou and muskox populations across most of their geographic prange in North America but it was rare in moose. Parelaphostrongylus andersoni was present in caribou and moose and we provide new geographic records for this species. This study provides a substantial expansion of the knowledge defining the current distribution and biogeography of protostrongylid nematodes in northern ungulates. Insights about the host and geographic range of V. eleguneniensis can serve as a geographically extensive baseline for monitoring current distribution and in anticipating future biogeographic scenarios under a regime of accelerating climate and anthropogenic perturbation
Vascular Permeability Factor/Vascular Endothelial Growth Factor Induces Lymphangiogenesis as well as Angiogenesis
Vascular permeability factor/vascular endothelial growth factor (VPF/VEGF, VEGF-A) is a multifunctional cytokine with important roles in pathological angiogenesis. Using an adenoviral vector engineered to express murine VEGF-A164, we previously investigated the steps and mechanisms by which this cytokine induced the formation of new blood vessels in adult immunodeficient mice and demonstrated that the newly formed blood vessels closely resembled those found in VEGF-A–expressing tumors. We now report that, in addition to inducing angiogenesis, VEGF-A164 also induces a strong lymphangiogenic response. This finding was unanticipated because lymphangiogenesis has been thought to be mediated by other members of the VPF/VEGF family, namely, VEGF-C and VEGF-D. The new “giant” lymphatics generated by VEGF-A164 were structurally and functionally abnormal: greatly enlarged with incompetent valves, sluggish flow, and delayed lymph clearance. They closely resembled the large lymphatics found in lymphangiomas/lymphatic malformations, perhaps implicating VEGF-A in the pathogenesis of these lesions. Whereas the angiogenic response was maintained only as long as VEGF-A was expressed, giant lymphatics, once formed, became VEGF-A independent and persisted indefinitely, long after VEGF-A expression ceased. These findings raise the possibility that similar, abnormal lymphatics develop in other pathologies in which VEGF-A is overexpressed, e.g., malignant tumors and chronic inflammation
Coordination of microtubule and microfilament dynamics by Drosophila Rho1, Spire, and Cappuccino
The actin nucleation factors Spire and Cappuccino regulate the onset of ooplasmic
streaming in Drosophila1-5. Although this streaming event is microtubule-based, actin
assembly is required for its timing. It is not understood how the interaction of
microtubules and microfilaments is mediated in this context. Here we demonstrate that
Cappuccino and Spire have microtubule and microfilament crosslinking activity. The spire
locus encodes several distinct protein isoforms (SpireA, SpireC, and SpireD). SpireD was
recently shown to nucleate actin, but the activity of the other isoforms has not been
addressed. We find that SpireD does not have crosslinking activity, while SpireC is a
potent crosslinker. We show that SpireD binds to Cappuccino and inhibits Factin/
microtubule crosslinking, and activated Rho1 abolishes this inhibition, establishing a
mechanistic basis for the regulation of Capu and Spire activity. We propose that Rho1,
cappuccino and spire are elements of a conserved developmental cassette that is capable of
directly mediating crosstalk between microtubules and microfilaments
The FGLamide-Allatostatins Influence Foraging Behavior in Drosophila melanogaster
Allatostatins (ASTs) are multifunctional neuropeptides that generally act in an inhibitory fashion. ASTs were identified as inhibitors of juvenile hormone biosynthesis. Juvenile hormone regulates insect metamorphosis, reproduction, food intake, growth, and development. Drosophila melanogaster RNAi lines of PheGlyLeu-amide-ASTs (FGLa/ASTs) and their cognate receptor, Dar-1, were used to characterize roles these neuropeptides and their respective receptor may play in behavior and physiology. Dar-1 and FGLa/AST RNAi lines showed a significant reduction in larval foraging in the presence of food. The larval foraging defect is not observed in the absence of food. These RNAi lines have decreased for transcript levels which encodes cGMP- dependent protein kinase. A reduction in the for transcript is known to be associated with a naturally occuring allelic variation that creates a sitter phenotype in contrast to the rover phenotype which is caused by a for allele associated with increased for activity. The sitting phenotype of FGLa/AST and Dar-1 RNAi lines is similar to the phenotype of a deletion mutant of an AST/galanin-like receptor (NPR-9) in Caenorhabditis elegans. Associated with the foraging defect in C. elegans npr-9 mutants is accumulation of intestinal lipid. Lipid accumulation was not a phenotype associated with the FGLa/AST and Dar-1 RNAi lines
Functional dissection of the Drosophila Kallmann's syndrome protein DmKal-1
BACKGROUND: Anosmin-1, the protein implicated in the X-linked Kallmann's syndrome, plays a role in axon outgrowth and branching but also in epithelial morphogenesis. The molecular mechanism of its action is, however, widely unknown. Anosmin-1 is an extracellular protein which contains a cysteine-rich region, a whey acidic protein (WAP) domain homologous to some serine protease inhibitors, and four fibronectin-like type III (FnIII) repeats. Drosophila melanogaster Kal-1 (DmKal-1) has the same protein structure with minor differences, the most important of which is the presence of only two FnIII repeats and a C-terminal region showing a low similarity with the third and the fourth human FnIII repeats. We present a structure-function analysis of the different DmKal-1 domains, including a predicted heparan-sulfate binding site. RESULTS: This study was performed overexpressing wild type DmKal-1 and a series of deletion and point mutation proteins in two different tissues: the cephalopharyngeal skeleton of the embryo and the wing disc. The overexpression of DmKal-1 in the cephalopharyngeal skeleton induced dosage-sensitive structural defects, and we used these phenotypes to perform a structure-function dissection of the protein domains. The reproduction of two deletions found in Kallmann's Syndrome patients determined a complete loss of function, whereas point mutations induced only minor alterations in the activity of the protein. Overexpression of the mutant proteins in the wing disc reveals that the functional relevance of the different DmKal-1 domains is dependent on the extracellular context. CONCLUSION: We suggest that the role played by the various protein domains differs in different extracellular contexts. This might explain why the same mutation analyzed in different tissues or in different cell culture lines often gives opposite phenotypes. These analyses also suggest that the FnIII repeats have a main and specific role, while the WAP domain might have only a modulator role, strictly connected to that of the fibronectins
A Versatile ΦC31 Based Reporter System for Measuring AP-1 and Nrf2 Signaling in Drosophila and in Tissue Culture
This paper describes the construction and characterization of a system of transcriptional reporter genes for monitoring the activity of signaling pathways and gene regulation mechanisms in intact Drosophila, dissected tissues or cultured cells. Transgenic integration of the reporters into the Drosophila germline was performed in a site-directed manner, using ΦC31 integrase. This strategy avoids variable position effects and assures low base level activity and high signal responsiveness. Defined integration sites furthermore enable the experimenter to compare the activity of different reporters in one organism. The reporter constructs have a modular design to facilitate the combination of promoter elements (synthetic transcription factor binding sites or natural regulatory sequences), reporter genes (eGFP, or DsRed.T4), and genomic integration sites. The system was used to analyze and compare the activity and signal response profiles of two stress inducible transcription factors, AP-1 and Nrf2. To complement the transgenic reporter fly lines, tissue culture assays were developed in which the same synthetic ARE and TRE elements control the expression of firefly luciferase
