    A genetic polymorphism in the CYP1B1 gene in patients with squamous cell carcinoma of the esophagus: an Iranian Mashhad cohort study recruited over 10 years

    Background: Esophageal-cancer is the seventh most common-cause of cancer-related-deaths in men. Cytochrome-P450-family-1-subfamily-B-polypeptide-1 (CYP1B1) plays a role in the metabolism of xenobiotics, and is associated with several cancers. Here we investigated the association between a genetic-variant, CYP1B1-rs1056836, with the clinical-characteristics of patients with esophagus-squamous-cell-carcinoma (ESCC). Method: 117-patients with ESCC and 208 healthy-subjects were recruited. DNA was extracted and genotyped. Kaplan-Meier curves were utilized to assess overall and progression-free survival. The relationship between clinicopathological-data, disease-prognosis, and survival, were evaluated with the genotypes. Results: the genotypic frequency for GG, GC, and CC were 58.6%, 29.8%, 11.5% respectively in the healthy subjects and 51.8%, 36.14% and 12% in the ESCC group. An association between the GG genotype and stage of ESCC was found. Conclusion: Our findings suggest a relationship between the CYP1B1-rs1056836 genetic polymorphism and clinical features of ESCC, supporting further studies in larger-populations in different-ethnic groups, taking into account potentially important environmental-factors

    Association of age related macular degeneration and age related hearing impairment

    Purpose: To evaluate the association between age-related macular degeneration (ARMD) and sensory neural hearing impairment (SHI). Methods: In this case-control study, hearing status of 46 consecutive patients with ARMD were compared with 46 age-matched cases without clinical ARMD as a control group. In all patients, retinal involvements were confirmed by clinical examination, fluorescein angiography (FA) and optical coherence tomography (OCT). All participants were examined with an otoscope and underwent audiological tests including pure tone audiometry (PTA), speech reception threshold (SRT), speech discrimination score (SDS), tympanometry, reflex tests and auditory brainstem response (ABR). Results: A significant (P = 0.009) association was present between ARMD, especially with exudative and choroidal neovascularization (CNV) components, and age-related hearing impairment primarily involving high frequencies. Patients had higher SRT and lower SDS against anticipated presbycusis than control subjects. Similar results were detected in exudative, CNV and scar patterns supporting an association between late ARMD with SRT and SDS abnormalities. ABR showed significantly prolonged wave I and IV latency times in ARMD (P = 0.034 and 0.022, respectively). Average latency periods for wave I in geographic atrophy (GA) and CNV, and that for wave IV in drusen patterns of ARMD were significantly higher than controls (P = 0.030, 0.007 and 0.050, respectively). Conclusion: The association between ARMD and age-related SHI may be attributed to common anatomical components such as melanin in these two sensory organs

    Surveying Mutagenic and Anti-mutagenic Effects of Citrus Limon

    Abstract Background and objective: Cancer is one of the main causes of mortality in the world and countless mutagenic chemical compound kill millions of cancer patients every years. Scientists are searching for natural food stuff to prevent cancer. The present paper examines anti-mutagenic and anti-carcinogenic effects of citrus lemon. Methodology: Human astrocytoma cancerous cell line was cultured in DMEM (Gibco) with FBS10% (fetal bovine serum), L-glutamine, penicillin, and streptomycin at 37ºC for two days. Then the samples were treated with citrus lemon juice and biotic potential of cells was examined through MTT method. Anti-mutagenic and anti-carcinogenic effects of citrus lemon were studied through back mutation assessment standard method (Ames test). The test was done using salmonella typhimurium strain (TA100) with mutation in histidine operon that needs external histidine source. The strain creates reverse colony in presence of carcinogenic agents (sodium azide). Findings: Based on MTT method, human astrocytoma cancerous cell line demonstrated significant cellular death comparing with the control groups (p<0.01). Ames test showed that the juice prevented back mutation and rate of inhibition, based on the anti-mutagenic effect, of half-ripen and ripen citrus lemon were 71.7% and 34.3% respectively. In addition, anti-carcinogenic effect of half-ripen and ripen citrus lemon were 83.3% and 50% respectively. Conclusion: Anti-mutagenic and anti-carcinogenic effects of citrus lemon were examined for the first time. The results showed that in this regard half-ripen citrus lemon was more effective than ripen citrus lemon
