Effect of immobilization stress on some toxicokinetics parametres of metals (cadmium, aluminium) in males of laboratory rats
The aim of this study was to evaluate the effects of stress on the kinetic of cadmium and aluminum on the male of laboratory rats. Information-analytical assessment systems for perinatal outcomes and children's health status born by assisted reproductive technologies
Purpose: To integrate clinical databases based on the information and analytical systems of medical organizations (MO) and to evaluate outcomes and health of children born by assisted reproductive technologies (ART). Research methods: To form database by integrating information and analytical systems of the medical organizations. To search and unify the data by means of freeware relational database management system (DBMS) - MySQL. Results. A prototype application for the management and support of a unified clinical database to analyze and evaluate the outcomes of ART. To compare information from individual databases of MO one need to match fields in the form of unique personal records, field validation, processing of missing data. Significant (p <0.001) differences in gestational age and anthropometric data in the databases of medical organizations (76% of full-term children from the Automated System βRegional obstetric monitoringβ (AS ROM) database compared with 18% according to the IS βRegister of Children requiring Early Careβ) were determined by the introduction of the conception of ART, along with the presence of perinatal risk factors in children. The created software can be used to create registers of medical organizations, as well as to support clinical decision-making in forecasting, modeling outcomes and children's health after ART, developing personalized treatment and rehabilitation programs. Conclusion. It is necessary to develop and implement information systems, to create ART registers on the basis of regional and national registers, to analyze the outcomes of the use of reproduction methods. Fragmentation of DNA in the male germ cells: impact on reproduction, the causes of origin and methods of diagnosis
The studying of DNA fragmentation in human sperm is necessary, because the DNA breaks, inherited from the paternal genome, could impact the embryo development. In this review the known mechanisms that cause DNA fragmentation in human sperm are considered. The mechanisms examined include: defects in chromatin remodeling during the process of spermiogenesis, apoptosis, oxygen radical-induced DNA damage. The different tests currently used for sperm DNA fragmentation analysis and the factors that determine the predictive value of sperm DNA fragmentation testing are also discussed.ΠΠ΅ΠΎΠ±Ρ

βUsage a CRISPR/Cas9-based for obtaining knockouts of economically significant cattle genesβ
An experimental work dealing with the gene modification using the Cas9 RNA-based editing system was performed. Point site-specific breakpoints in gDNA were introduced at the zygote stage by microinjection of spCas9 mRNA protein and guide RNAs into the zygote cytoplasm. Oocytes that extruded the first and second polar bodies were used for the injection. 2 series of microinjections of gene editing designs for early bovine embryos were made. The degeneration ranged from 10% to 56% in different groups. A total of 100 injections were performed. Cleavage was started by 78% of the surviving oocytes; 5 embryos reached the blastocyst stage, which was 16% of the number of dividing embryos. All the resulting embryos were analyzed to evaluate the efficiency of editing. gDNA was isolated from all embryos that had reached the blastocyst stage. Using Sanger sequencing of genes of interest in pre-implantation bovine embryos and biopsies from them, it was shown that in 5 out of 17 embryos resulting from microinjections of guide RNA against the BLG gene and SpCas9 mRNA, and in 2 out of 9 embryos after microinjections of guide RNA against CD209 gene and SpCas9 mRNA, the required genome modifications were found. This is indicative of the high efficiency of this delivery method of the editing system
Potentiometric method for evaluating the Oxidant/Antioxidant activity of seminal and follicular fluids and clinical significance of this parameter for human reproductive function
Potentiometric method based on the sharp change of electrode potential for the ferricyanide/ferrocyanide mediator system after addition of the seminal or follicular fluid sample into the measuring electrochemical cell has been developed to enable the easy assessment of the oxidant/antioxidant status of these kind of biological fluids. Platinum screen printed electrode has been used as an indicator. Essential advantage of the proposed method is simultaneous use of the both forms of the ferricyanide/ferrocyanide mediator system (oxidized and reduced ones) that allows estimating oxidant and antioxidant components of the seminal or follicular fluids samples which were obtained, respectively, from 99 male and 27 female subjects. Oxidant activity of seminal fluids has been observed in pathology conditions of reproductive system of male studied subjects and related with possible male infertility symptoms. Β© Brainina et al
