    Performance of an innovative multi-stage anaerobic reactor during start-up period

    Start-up of an anaerobic reactor is a relatively delicate process and depends on various factors such as wastewater composition, available inoculum, operating conditions and reactor configuration. Accordingly, systematized operational procedures are important, mainly during the start-up of an anaerobic reactor. In this paper, the start-up performance of an innovative multi-stage anaerobic reactor using synthetic wastewater at various organic loading rates (OLRs) was investigated. In Phase 1 of the experimental study, the reactor was operated at hydraulic retention time (HRT) of 1 day with corresponding OLR of 1.07 kg COD.m-3.d-1. Thereafter, the reactor was operated at intermittent feeding (Phase 2), with HRT of 1.4 day and OLR of 0.82 to 2.45 kg COD.m-3.d-1. Results showed up to 71% COD reduction in the Phase 1 of the experimental study. However, in Phase 2, when the reactor was operated at intermittent feeding, the COD removal efficiency increased from 75 to 92%. It can be concluded that the multi-stage anaerobic reactor system performed better at intermittent feeding, indicating that the reactor required low loading rate and sufficient HRT for gradual acclimatization for reactor start-up. The reduction of the period necessary for the start-up and improved operational control are important factors to increase the efficiency the reactor system.Key words: Anaerobic reactor start-up, biomass, glucose wastewater, intermittent feeding, multi-stage anaerobic reactor

    Influence of citronella and chlorpyrifos on Chrysomya megacephala (Fabricius) and Chrysomya rufifacies (Macquart) (Diptera: Calliphoridae) infesting rabbit carcasses

    Influence of citronella and chlorpyrifos on oviposition and duration for completing life cycles for Chrysomya megacephala and Chrysomya rufifacies infesting decomposing rabbit carcasses was studied. Male rabbit carcasses (n = 12) were equally divided into control, citronella-and chlorpyrifos–treated groups, and left to decompose for 14 consecutive days. C. megacephala was the first necrophagous fly oviposited in all control and citronella-treated carcasses followed by C. rufifacies. Although initial oviposition of C. megacephala was delayed (4-6 hours) in citronella-treated carcasses (P < 0.05), prolongation in completing its life cycle was not observed. Neither delayed initial oviposition nor prolonged life cycle for C. rufifacies in citronella-treated carcasses was observed. Oviposition was delayed for chlorpyrifos-treated carcasses (0.42 g/L), and eclosion of eggs remained unsuccessful. The findings deserve consideration because these chemicals are easily accessible and can be used by cunning criminals to confuse forensic entomologists while estimating minimum postmortem interval

    Pengembangan Koleksi Tumbuhan Kebun Raya Daerah Dalam Kerangka Strategi Konservasi Tumbuhan Di Indonesia

    Sebagai bagian dari kebun raya dunia, Kebun Raya Indonesia (KRI) memiliki tanggung jawab untuk melaksanakan upaya konservasi tumbuhan yang ada di Indonesia. Hingga tahun 2010, empat kebun raya yang dikelola oleh LIPI hanya mampu mengkonservasi sekitar 21% dari seluruh tumbuhan terancam Indonesia. Oleh karena itu, diperlukan pengembangan Kebun Raya Daerah (KRD) untuk mengkonservasi tumbuhan pada tiap daerah di Indonesia. Makalah ini bertujuan untuk untuk mengetahui capaian KRD dalam konservasi tumbuhan dan menentukan strategi pengelolaan koleksi tumbuhan tiap daerah. Pada akhir 2012, KRI yang didukung KRD telah berhasil mengoleksi 24% dari tumbuhan terancam Indonesia versi IUCN Red List 2013, dan berhasil membudidayakan 25% termasuk koleksi pembibitan. Beberapa strategi untuk pengembangan koleksi KRD antara lain: penguatan sistem data base koleksi; pengembangan koleksi tumbuhan di setiap KRD dengan mengacu pada IUCN Red List; dan penentuan spesies prioritas untuk konservasi terkait dengan kebijakan dan kondisi alam di Indonesia

    Novel safranin-tinted Candida rugosa lipase nanoconjugates reagent for visualizing latent fingerprints on stainless steel knives immersed in a natural outdoor pond

    Waterways are popular locations for the disposition of criminal evidence because the recovery of latent fingerprints from such evidence is difficult. Currently, small particle reagent is a method often used to visualize latent fingerprints containing carcinogenic and hazardous compounds. This study proposes an eco-friendly, safranin-tinted Candida rugosa lipase (triacylglycerol ester hydrolysis EC 3.1.1.3) with functionalized carbon nanotubes (CRL-MWCNTS/GA/SAF) as an alternative reagent to the small particle reagent. The CRL-MWCNTS/GA/SAF reagent was compared with the small particle reagent to visualize groomed, full fingerprints deposited on stainless steel knives which were immersed in a natural outdoor pond for 30 days. The quality of visualized fingerprints using the new reagent was similar (modified-Centre for Applied Science and Technology grade: 4; p > 0.05) to small particle reagent, even after 15 days of immersion. Despite the slight decrease in quality of visualized fingerprints using the CRL-MWCNTS/GA/SAF on the last three immersion periods, the fingerprints remained forensically identifiable (modified-Centre for Applied Science and Technology grade: 3). The possible chemical interactions that enabled successful visualization is also discussed. Thus, this novel reagent may provide a relatively greener alternative for the visualization of latent fingerprints on immersed non-porous objects

    Assessments on the catalytic and kinetic properties of beta-glucosidase isolated from a highly efficient antagonistic fungus Trichoderma harzianum

    Due to the toxicity and inefficiency of chemical fungicides to control infestation of Macrophomina phaseolina (Tassi) Goid which causes charcoal rot in plants, a biotechnological approach using β-glucosidase (EC.3.2.1) as the alternative bioactive ingredient in fungicide is hereby, proposed. The extracellular enzyme was isolated from a highly efficient fungal antagonist, Trichoderma harzianum T12. The highly similar molecular masses obtained using SDS-PAGE (96 kDa) and MALDI-TOF mass spectrometry (98.3 kDa) affirmed that the β-glucosidase was purified to homogeneity. Consequently, optimum catalytic parameters that rendered the highest enzyme activity were found to be: 45˚C, pH 7, inoculum size of 10 % (w/v), supplementation with metal ions Zn2+ and Mn2+ ions, and Tween 80. Addition of wheat bran and (NH4)2SO4 as carbon and nitrogen sources also improved enzyme activity. BLASTn showed the sequence of β-glucosidase T12 was highly identical to other β-glucosidases viz. T. harzianum strain IOC-3844 (99%), T. gamsii and T. virens bgl1 (86 %) as well as T. reesei strain SJVTR and T. viride strain AS 3.3711 (84 %). Kinetic assessment showed that β-glucosidase T12 catalyzes hydrolytic activity is characterized by a Km of 0.79 mM and Vmax of 8.45 mM min-1 mg-1 protein, with a corresponding kcat of 10.69 s-1

    Estimation of stature from hand and handprint measurements in Iban population in Sarawak, Malaysia and its applications in forensic investigation

    Handprints and dismembered hands are commonly found during crime scene investigations and disaster victim identifications, respectively. It has been indicated that the accuracy of handprint and hand measurements for estimating stature maybe population specific. Since Iban is the largest ethnic population in Sarawak, Malaysia and because the application of anthropometry of hand and handprint within this population as well as other populations within the Southeast Asian countries remain unreported, this present study that investigated the reliability and accuracy of these two anthropometric aspects acquires forensic significance. Upon measuring the height, 21 measurements were recorded on each hand and the corresponding handprint of 50 male and 52 female consented adult Iban subjects. Using univariate statistics as well as simple and multiple regression analyses, interpretation of the measurements examined here was attempted. Results revealed that lengths of hand and handprint are the more reliable traits for estimating stature in both the male and female Iban subjects (p < 0.05) with correlation strength ranging from 0.60 to 0.76. Comparable to the established skeletal standards for hand, the stature prediction accuracy using hand and handprint measurements investigated in this research ranged between 4.29 and 5.78 cm. Hence, this research provided the first forensic standard for estimation of stature among the Iban population in Sarawak that may prove useful for crime scene investigations and disaster victim identifications in Malaysia

    Diversification of the Caenorhabditis heat shock response by Helitron transposable elements.

    Heat Shock Factor 1 (HSF-1) is a key regulator of the heat shock response (HSR). Upon heat shock, HSF-1 binds well-conserved motifs, called Heat Shock Elements (HSEs), and drives expression of genes important for cellular protection during this stress. Remarkably, we found that substantial numbers of HSEs in multiple Caenorhabditis species reside within Helitrons, a type of DNA transposon. Consistent with Helitron-embedded HSEs being functional, upon heat shock they display increased HSF-1 and RNA polymerase II occupancy and up-regulation of nearby genes in C. elegans. Interestingly, we found that different genes appear to be incorporated into the HSR by species-specific Helitron insertions in C. elegans and C. briggsae and by strain-specific insertions among different wild isolates of C. elegans. Our studies uncover previously unidentified targets of HSF-1 and show that Helitron insertions are responsible for rewiring and diversifying the Caenorhabditis HSR

    Cobalt containing glass fibres and their synergistic effect on the HIF-1 pathway for wound healing applications

    Introduction and Methods: Chronic wounds are a major healthcare problem, but their healing may be improved by developing biomaterials which can stimulate angiogenesis, e.g. by activating the Hypoxia Inducible Factor (HIF) pathway. Here, novel glass fibres were produced by laser spinning. The hypothesis was that silicate glass fibres that deliver cobalt ions will activate the HIF pathway and promote the expression of angiogenic genes. The glass composition was designed to biodegrade and release ions, but not form a hydroxyapatite layer in body fluid. Results and Discussion: Dissolution studies demonstrated that hydroxyapatite did not form. When keratinocyte cells were exposed to conditioned media from the cobalt-containing glass fibres, significantly higher amounts of HIF-1α and Vascular Endothelial Growth Factor (VEGF) were measured compared to when the cells were exposed to media with equivalent amounts of cobalt chloride. This was attributed to a synergistic effect of the combination of cobalt and other therapeutic ions released from the glass. The effect was also much greater than the sum of HIF-1α and VEGF expression when the cells were cultured with cobalt ions and with dissolution products from the Co-free glass, and was proven to not be due to a rise in pH. The ability of the glass fibres to activate the HIF-1 pathway and promote VEGF expression shows the potential for their use in chronic wound dressings.National Institute for Health Research (Reino Unido) | Ref. In this edition, the 400-µg/L drinking-water guideline for manganese (Mn) was discontinued with the assertion that because “this health-based value is well above concentrations of manganese normally found in drinking water, it is not considered necessary to derive a formal guideline value.
