Pulaski County Education Report Card 2021
The goal of this report is to help parents, school personnel, community members, and policy makers understand how effectively the students in their community are being served by public schools. The past two years have been difficult for families, teachers, and students, and throughout the challenging context, Pulaski County students demonstrated lower growth in achievement on the ACT Aspire than students in the state overall, and are performing below the state average in achievement, graduation rates, and School Quality and Student Success scores. Acknowledging the unprecedented circumstances presented by the COVID virus, Arkansas schools were not assigned an overall letter grade in 2020 or 2021

Satellite-Based Assessment of Grassland Conversion and Related Fire Disturbance in the Kenai Peninsula, Alaska
Spruce beetle-induced (Dendroctonus rufipennis (Kirby)) mortality on the Kenai Peninsula has been hypothesized by local ecologists to result in the conversion of forest to grassland and subsequent increased fire danger. This hypothesis stands in contrast to empirical studies in the continental US which suggested that beetle mortality has only a negligible effect on fire danger. In response, we conducted a study using Landsat data and modeling techniques to map land cover change in the Kenai Peninsula and to integrate change maps with other geospatial data to predictively map fire danger for the same region. We collected Landsat imagery to map land cover change at roughly five-year intervals following a severe, mid-1990s beetle infestation to the present. Land cover classification was performed at each time step and used to quantify grassland encroachment patterns over time. The maps of land cover change along with digital elevation models (DEMs), temperature, and historical fire data were used to map and assess wildfire danger across the study area. Results indicate the highest wildfire danger tended to occur in herbaceous and black spruce land cover types, suggesting that the relationship between spruce beetle damage and wildfire danger in costal Alaskan forested ecosystems differs from the relationship between the two in the forests of the coterminous United States. These change detection analyses and fire danger predictions provide the Kenai National Wildlife Refuge (KENWR) ecologists and other forest managers a better understanding of the extent and magnitude of grassland conversion and subsequent change in fire danger following the 1990s spruce beetle outbreak
High-density rhesus macaque oligonucleotide microarray design using early-stage rhesus genome sequence information and human genome annotations
BACKGROUND: Until recently, few genomic reagents specific for non-human primate research have been available. To address this need, we have constructed a macaque-specific high-density oligonucleotide microarray by using highly fragmented low-pass sequence contigs from the rhesus genome project together with the detailed sequence and exon structure of the human genome. Using this method, we designed oligonucleotide probes to over 17,000 distinct rhesus/human gene orthologs and increased by four-fold the number of available genes relative to our first-generation expressed sequence tag (EST)-derived array. RESULTS: We constructed a database containing 248,000 exon sequences from 23,000 human RefSeq genes and compared each human exon with its best matching sequence in the January 2005 version of the rhesus genome project list of 486,000 DNA contigs. Best matching rhesus exon sequences for each of the 23,000 human genes were then concatenated in the proper order and orientation to produce a rhesus "virtual transcriptome." Microarray probes were designed, one per gene, to the region closest to the 3' untranslated region (UTR) of each rhesus virtual transcript. Each probe was compared to a composite rhesus/human transcript database to test for cross-hybridization potential yielding a final probe set representing 18,296 rhesus/human gene orthologs, including transcript variants, and over 17,000 distinct genes. We hybridized mRNA from rhesus brain and spleen to both the EST- and genome-derived microarrays. Besides four-fold greater gene coverage, the genome-derived array also showed greater mean signal intensities for genes present on both arrays. Genome-derived probes showed 99.4% identity when compared to 4,767 rhesus GenBank sequence tag site (STS) sequences indicating that early stage low-pass versions of complex genomes are of sufficient quality to yield valuable functional genomic information when combined with finished genome information from a closely related species. CONCLUSION: The number of different genes represented on microarrays for unfinished genomes can be greatly increased by matching known gene transcript annotations from a closely related species with sequence data from the unfinished genome. Signal intensity on both EST- and genome-derived arrays was highly correlated with probe distance from the 3' UTR, information often missing from ESTs yet present in early-stage genome projects
Therapeutically engineered induced neural stem cells are tumour-homing and inhibit progression of glioblastoma
Transdifferentiation (TD) is a recent advancement in somatic cell reprogramming. The direct conversion of TD eliminates the pluripotent intermediate state to create cells that are ideal for personalized cell therapy. Here we provide evidence that TD-derived induced neural stem cells (iNSCs) are an efficacious therapeutic strategy for brain cancer. We find that iNSCs genetically engineered with optical reporters and tumouricidal gene products retain the capacity to differentiate and induced apoptosis in co-cultured human glioblastoma cells. Time-lapse imaging shows that iNSCs are tumouritropic, homing rapidly to co-cultured glioblastoma cells and migrating extensively to distant tumour foci in the murine brain. Multimodality imaging reveals that iNSC delivery of the anticancer molecule TRAIL decreases the growth of established solid and diffuse patient-derived orthotopic glioblastoma xenografts 230- and 20-fold, respectively, while significantly prolonging the median mouse survival. These findings establish a strategy for creating autologous cell-based therapies to treat patients with aggressive forms of brain cancer

We collected images from NASA satellite-based Earth observations to visualize land cover succession at roughly five-year intervals following a severe, mid-1990's beetle infestation to the present. We classified these data by vegetation cover type to quantify grassland encroachment patterns over time. Raster band math provided a change detection analysis on the land cover classifications. Results indicate the highest wildfire risk is linked to herbaceous and black spruce land cover types, The resulting land cover change image will give the Kenai National Wildlife Refuge (KENWR) ecologists a better understanding of where forests have converted to grassland since the 1990s. These classifications provided a foundation for us to integrate digital elevation models (DEMs), temperature, and historical fire data into a model using Python for assessing and mapping changes in wildfire risk. Spatial representations of this risk will contribute to a better understanding of ecological trajectories of beetle-affected landscapes, thereby informing management decisions at KENWR
SOX9 Maintains Reserve Stem Cells and Preserves Radioresistance in Mouse Small Intestine
Reserve intestinal stem cells (rISCs) are quiescent/slowly cycling under homeostatic conditions, allowing for their identification with label-retention assays. rISCs mediate epithelial regeneration after tissue damage by converting to actively proliferating stem cells (aISCs) that self renew and demonstrate multipotency, which are defining properties of stem cells. Little is known about the genetic mechanisms that regulate the production and maintenance of rISCs. High expression levels of the transcription factor Sox9 (Sox9high) are associated with rISCs. Analysis of the Macaca mulatta transcriptome and the sequence divergence between Macaca and human
We report the initial sequencing and comparative analysis of the Macaca mulatta transcriptome. Cloned sequences from 11 tissues, nine animals, and three species (M. mulatta, M. fascicularis, and M. nemestrina) were sampled, resulting in the generation of 48,642 sequence reads. These data represent an initial sampling of the putative rhesus orthologs for 6,216 human genes. Mean nucleotide diversity within M. mulatta and sequence divergence among M. fascicularis, M. nemestrina, and M. mulatta are also reported
