    Promoter DNA Methylation of Oncostatin M receptor-β as a Novel Diagnostic and Therapeutic Marker in Colon Cancer

    In addition to genetic changes, the occurrence of epigenetic alterations is associated with accumulation of both genetic and epigenetic events that promote the development and progression of human cancer. Previously, we reported a set of candidate genes that comprise part of the emerging “cancer methylome”. In the present study, we first tested 23 candidate genes for promoter methylation in a small number of primary colon tumor tissues and controls. Based on these results, we then examined the methylation frequency of Oncostatin M receptor-β (OSMR) in a larger number of tissue and stool DNA samples collected from colon cancer patients and controls. We found that OSMR was frequently methylated in primary colon cancer tissues (80%, 80/100), but not in normal tissues (4%, 4/100). Methylation of OSMR was also detected in stool DNA from colorectal cancer patients (38%, 26/69) (cut-off in TaqMan-MSP, 4). Detection of other methylated markers in stool DNA improved sensitivity with little effect on specificity. Promoter methylation mediated silencing of OSMR in cell lines, and CRC cells with low OSMR expression were resistant to growth inhibition by Oncostatin M. Our data provide a biologic rationale for silencing of OSMR in colon cancer progression and highlight a new therapeutic target in this disease.     The results of an investigation of the performance of a TDM FBG sensor array using a tunable laser source are reported. The system performance is found to be limited by the extinction ratio of the optical pulse modulator used for pulse amplitude modulation. Formulas that relate the crosstalk to the extinction ratio of the optical pulse modulator, the modulation parameters of the tunable laser, and the optical path differences among sensing channels are derived. Computer simulation shows that an array of 20 FBG sensors with 3 (mu) (epsilon) resolution can be realized with a commercially available single Mach-Zehnder type optical pulse modulator of -35 dB extinction ratio. Experiments with a 4-FBG sensor array demonstrated a strain resolution of about 3.98 (mu) (epsilon) with an optical pulse modulator of about -24 dB extinction ratio.Department of Electrical Engineerin

    Annealing effects on the loss and birefringence of silicon oxynitride rectangular optical waveguides

    Silicon oxynitride rectangular optical waveguides have been fabricated by plasma-enhanced chemical vapor deposition and dry etching. The propagation loss and polarization dependent loss in as-grown samples show a substantial reduction after the samples have been annealed. Birefringence measurements before and after annealing on waveguides of different widths suggest that the waveguide modal birefringence is strongly affected by both waveguide geometry and stress in the material. Hence, the modal birefringence can be minimized by designing the appropriate waveguide geometry to compensate for any stress effects. (c) 2005 American Institute of Physics
