Therapeutic Activity of Superoxide Dismutase-Containing Enzymosomes on Rat Liver Ischaemia-Reperfusion Injury Followed by Magnetic Resonance Microscopy
Liver ischaemia-reperfusion injury (IRI) may occur during hepatic surgery and is unavoidable in liver transplantation. Superoxide dismutase enzymosomes (SOD-enzymosomes), liposomes where SOD is at the liposomal surface expressing enzymatic activity in intact form without the need of liposomal disruption, were developed with the aim of having a better insight into its antioxidant therapeutic outcome in IRI. We also aimed at validating magnetic resonance microscopy (MRM) at 7T as a tool to follow IRI. SOD-enzymosomes were characterized and tested in a rat ischaemia-reperfusion model and the therapeutic outcome was compared with conventional long circulating SOD liposomes and free SOD using biochemical liver injury biomarkers, histology and MRM. MRM results correlated with those obtained using classical biochemical biomarkers of liver injury and liver histology. Moreover, MRM images suggested that the therapeutic efficacy of both SOD liposomal formulations used was related to prevention of peripheral biliary ductular damage and disrupted vascular architecture. Therefore, MRM at 7T is a useful technique to follow IRI. SOD-enzymosomes were more effective than conventional liposomes in reducing liver ischaemia-reperfusion injury and this may be due to a short therapeutic window.info:eu-repo/semantics/publishedVersio
Elaboração e avaliação de um website sobre o desenvolvimento da linguagem infantil: portal dos bebês - desenvolvimento da linguagem
OBJETIVO: criar e desenvolver um website na área de fonoaudiologia com enfoque no desenvolvimento da linguagem de bebês de 0 a 48 meses, além de avaliar a qualidade do conteúdo apresentado e os recursos tecnológicos utilizados. MÉTODOS: na primeira fase do projeto foi realizado um levantamento de dados por meio de revisão de literatura sobre o desenvolvimento e aquisição da linguagem infantil. Em seguida esse conteúdo foi inserido no website e avaliado por 20 pais e/ou cuidadores. RESULTADO: o website Portal dos Bebês - Desenvolvimento da Linguagem foi desenvolvido com linguagem voltada a pais e/ou cuidadores, e seu conteúdo foi dividido em três temas: "Desenvolvimento da Linguagem", "Fala e Linguagem" e "Linguagem e o Ambiente". No que se refere à qualidade do conteúdo e da apresentação, o website foi avaliado como "excelente" por todos os participantes da amostra. Quanto à avaliação sobre o vocabulário utilizado e a forma de apresentação do conteúdo, 70% e 75% respectivamente, consideraram o website como "excelente". Em relação à apresentação visual, 85% dos participantes classificaram como "excelentes", já o tipo de navegação foi classificado como "satisfatório" por 50% dos participantes. Quanto à importância que os pais e/ou cuidadores julgam sobre o site a maioria classificou como "excelente" o auxilio que o mesmo dá no desenvolvimento da linguagem da criança. CONCLUSÃO: o website Portal dos Bebês - Desenvolvimento da Linguagem pode ser acessado pelo endereço online http://portaldosbebes.fob.usp.br, e seu conteúdo e recursos tecnológicos utilizados foram avaliados como satisfatório a excelente pelos pais e/ou cuidadores participantes
The methodological quality of systematic reviews comparing temporomandibular joint disorder surgical and non-surgical treatment
<p>Abstract</p> <p>Background</p> <p>Temporomandibular joint disorders (TMJD) are multifactor, complex clinical problems affecting approximately 60–70% of the general population, with considerable controversy about the most effective treatment. For example, reports claim success rates of 70% and 83% for non-surgical and surgical treatment, whereas other reports claim success rates of 40% to 70% for self-improvement without treatment. Therefore, the purpose of this study was to (1) identify systematic reviews comparing temporomandibular joint disorder surgical and non-surgical treatment, (2) evaluate their methodological quality, and (3) evaluate the evidence grade within the systematic reviews.</p> <p>Methods</p> <p>A search strategy was developed and implemented for MEDLINE, Cochrane Library, LILACS, and Brazilian Dentistry Bibliography databases. Inclusion criteria were: systematic reviews (± meta-analysis) comparing surgical and non-surgical TMJD treatment, published in English, Spanish, Portuguese, Italian, or German between the years 1966 and 2007(up to July). Exclusion criteria were: <it>in vitro </it>or animal studies; narrative reviews or editorials or editorial letters; and articles published in other languages. Two investigators independently selected and evaluated systematic reviews. Three different instruments (AMSTAR, OQAQ and CASP) were used to evaluate methodological quality, and the results averaged. The GRADE instrument was used to evaluate the evidence grade within the reviews.</p> <p>Results</p> <p>The search strategy identified 211 reports; of which 2 were systematic reviews meeting inclusion criteria. The first review met 23.5 ± 6.0% and the second met 77.5 ± 12.8% of the methodological quality criteria (mean ± sd). In these systematic reviews between 9 and 15% of the trials were graded as high quality, and 2 and 8% of the total number of patients were involved in these studies.</p> <p>Conclusion</p> <p>The results indicate that in spite of the widespread impact of TMJD, and the multitude of potential interventions, clinicians have expended sparse attention to systematically implementing clinical trial methodology that would improve validity and reliability of outcome measures. With some 20 years of knowledge of evidence-based healthcare, the meager attention to these issues begins to raise ethical issues about TMJD trial conduct and clinical care.</p
Human predisposition to cognitive impairment and its relation with environmental exposure to potentially toxic elements
New lines of evidence suggest that less than 10% of neurodegenerative diseases have a strict genetic aetiology and other factors may be prevalent. Environmental exposures to potentially toxic elements appear to be a risk factor for Parkinson’s, Alzheimer’s and sclerosis diseases. This study proposes a multidisciplinary approach combining neurosciences, psychology and environmental sciences while integrating socio-economic, neuropsychological, environmental and health data. We present the preliminary results of a neuropsychological assessment carried out in elderly residents of the industrial city of Estarreja. A battery of cognitive tests and a personal questionnaire were administered to the participants. Multivariate analysis and multiple linear regression analysis were used to identify potential relationships between the cognitive status of the participants and environmental exposure to potentially toxic elements. The results suggest a relationship between urinary PTEs levels and the incidence of cognitive disorders. They also point towards water consumption habits and profession as relevant factors of exposure. Linear regression models show that aluminium (R2 = 38%), cadmium (R2 = 11%) and zinc (R2 = 6%) are good predictors of the scores of the Mini-Mental State Examination cognitive test. Median contents (µg/l) in groundwater are above admissible levels for drinking water for aluminium (371), iron (860), manganese (250), and zinc (305). While the World Health Organization does not provide health-based reference values for aluminium, results obtained from this study suggest that it may have an important role in the cognitive status of the elderly. Urine proved to be a suitable biomarker of exposure both to elements with low and high excretion rates
Individual-environment interactions in swimming: The smallest unit for analysing the emergence of coordination dynamics in performance?
Displacement in competitive swimming is highly dependent on fluid characteristics,
since athletes use these properties to propel themselves. It is essential for sport
scientists and practitioners to clearly identify the interactions that emerge between
each individual swimmer and properties of an aquatic environment. Traditionally, the
two protagonists in these interactions have been studied separately. Determining the
impact of each swimmer’s movements on fluid flow, and vice versa, is a major
challenge. Classic biomechanical research approaches have focused on swimmers’
actions, decomposing stroke characteristics for analysis, without exploring
perturbations to fluid flows. Conversely, fluid mechanics research has sought to
record fluid behaviours, isolated from the constraints of competitive swimming
environments (e.g. analyses in two-dimensions, fluid flows passively studied on
mannequins or robot effectors). With improvements in technology, however, recent
investigations have focused on the emergent circular couplings between swimmers’
movements and fluid dynamics. Here, we provide insights into concepts and tools that
can explain these on-going dynamical interactions in competitive swimming within
the theoretical framework of ecological dynamics
