    Localization of BDNF mRNA with the Huntington's disease protein in rat brain

    <p>Abstract</p> <p>Background</p> <p>Studies have implicated reduced levels of brain-derived neurotrophic factor (BDNF) in the pathogenesis of Huntington's disease. Mutant huntingtin (Htt) protein was previously reported to decrease BDNF gene transcription and axonal transport of BDNF. We recently showed that wild-type Htt is associated with the Argonaute 2 microRNA-processing enzyme involved in gene silencing. In dendrites, Htt co-localizes with components of neuronal granules and mRNAs, indicating that it might play a role in post-transcriptional processing/transport of dendritic mRNAs.</p> <p>Results</p> <p>We conducted imaging experiments in cultured cortical neurons to demonstrate the co-localization of endogenous Htt and BDNF mRNA in fixed cells, and co-trafficking of BDNF 3'UTR mRNA with endogenous and fluorescently tagged Htt in live neurons. We used an enhanced technique that combines FISH and immunofluorescent staining to co-localize BDNF mRNA with Htt, Ago2, CPEB and dynein in thick vibratome sections of the rat cortex.</p> <p>Conclusions</p> <p>In cultured neurons and sections of the rat cortex, we found BDNF mRNA associated with Htt and components of neuronal RNA granules, which are centers for regulating RNA transport and local translation. Htt may play a role in post-transcriptional transport/targeting of mRNA for BDNF, thus contributing to neurotrophic support and neuron survival.</p

    Deceleration and Dispersion of Large-scale Coronal Bright Fronts

    One of the most dramatic manifestations of solar activity are large-scale coronal bright fronts (CBFs) observed in extreme ultraviolet (EUV) images of the solar atmosphere. To date, the energetics and kinematics of CBFs remain poorly understood, due to the low image cadence and sensitivity of previous EUV imagers and the limited methods used to extract the features. In this paper, the trajectory and morphology of CBFs was determined in order to investigate the varying properties of a sample of CBFs, including their kinematics and pulse shape, dispersion, and dissipation. We have developed a semi-automatic intensity profiling technique to extract the morphology and accurate positions of CBFs in 2.5-10 min cadence images from STEREO/EUVI. The technique was applied to sequences of 171A and 195A images from STEREO/EUVI in order to measure the wave properties of four separate CBF events. Following launch at velocities of ~240-450kms^{-1} each of the four events studied showed significant negative acceleration ranging from ~ -290 to -60ms^{-2}. The CBF spatial and temporal widths were found to increase from ~50 Mm to ~200 Mm and ~100 s to ~1500 s respectively, suggesting that they are dispersive in nature. The variation in position-angle averaged pulse-integrated intensity with propagation shows no clear trend across the four events studied. These results are most consistent with CBFs being dispersive magnetoacoustic waves.Comment: 15 pages, 18 figure

    Towards an Improved Health Service Quality Delivery and Patient Loyalty: Does Satisfaction Really Matter?

    The healthcare industry in developing nations has chronicled high development rate in our current dispensation. This study sought to identify the most critical factors in hospitals related service delivery quality that will drive loyalty and hence ensure the survival and success of the hospital in the future; it ideally tested if the satisfaction of a patient had any role to play in such relationship. This study was conducted using data from (562) patients who received services from (4) four major but different private hospitals in Ghana. The five Service Quality dimensions; Tangibility, reliability, responsiveness, assurance and empathy, patient satisfaction and loyalty to the hospital were the variables well-thought-out for this study. A path analysis was done utilizing SmartPLS V3.2.8; a second-generation multivariate data analysis method (PLS-SEM) in order to compute path coefficients, direct and indirect effects of the variables on patient's satisfaction and also loyalty to the hospital. The study prompted that empathy, responsiveness and tangibility (not assurance and reliability) impact patients’ satisfaction and patient's satisfaction is directly related to patients’ loyalty to the hospital/clinic

    The Antecedes of Service Quality and Patients’ Satisfaction Equally Drive Patients’ Loyalty in Private Healthcare Delivery: Thinking the future

    The healthcare industry in developing nations has recorded high progress rate in recent years, leading an initiative varied studies into the examination of patients’ comparative and varied views with regards to private healthcare providers service delivery. This study objectively focused on the hospital’s service quality, and patients’ satisfaction as a predictor and collective impact or on patient’s loyalty; it attempted to draw a distinctive border amongst quality of service and satisfaction of the patient, one with more effect size and predictive relevance to drive greater loyalty amongst the patients in a private healthcare delivery setting. A total of 562 patients recruited, participated in a cross-sectional survey with a questionnaire as the main and only data collection tool from four major private hospitals in Ghana. A path and linear regression analysis of the data was performed through SPSS 23 and Smart PLS version 3 in order to compute path coefficients, direct and indirect impacts of the factors; service quality and patient's satisfaction on patient’s loyalty to the hospital.  The study suggested that both clientele perceived service quality and patient satisfaction significantly influence patient loyalty. However, patient satisfaction with service delivery had a better predictive relevance and effect size than the quality of the service on patient loyalty in this study

    Evidence for electron-electron interaction in topological insulator thin films

    We consider in our work high quality single crystal thin films of Bi2Se3, grown by molecular beam epitaxy, both with and without Pb doping. Our ARPES data demonstrate topological surface states with a Fermi level lying inside the bulk band gap in the Pb doped filims. Transport data show weak localization behavior, as expected for a 2D system, but a detailed analysis within the standard theoretical framework of diffusive transport shows that the temperature and magnetic field dependences of resistance cannot be reconciled in a theory that neglects inter-electron interactions. We demonstrate that an excellent account of quantum corrections to conductivity is achieved when both disorder and interaction are taken into account. These results clearly demonstrate that it is crucial to include electron electron interaction for a comprehensive understanding of diffusive transport in topological insulators.Comment: Submitted to Phys.
