Evaluation of biofungicides for control of clubroot on canola
Non-Peer ReviewedClubroot of canola, caused by the protist pathogen Plasmodiophora brassicae (Pb), is an
emerging threat to canola production in western Canada. Effective/practical control options
are currently lacking. This study was initiated to assess registered microbial fungicides for
control of clubroot on canola. Selected biofungicides were initially applied as a soil drench
and the fungicides Allegro and Ranman were also included for comparisons. Selected
products were further evaluated at varying concentrations, soil drench volumes, and for seed
treatment. At moderate disease pressure, the biofungicides Serenade and Prestop, as well as
synthetic fungicides Allegro and Ranman were highly effective as a soil-drench treatment,
reducing clubroot severity by 85–100% in controlled conditions. Biofungicide concentration
appeared to be important while soil-drench volumes may be reduced. All products, however,
were significantly less effective or ineffective under extremely high disease pressure. All
products were less efficacious in trials using infested field soils, a circumstance that may be
related to treatment timing. Results from seed-treatment trials were too variable to draw a
conclusion but there was a strong indication that this approach be successful though more
research is required on microbial formulations. Serenade, Prestop, Allegro, and Ranman
should be further evaluated under field conditions for clubroot control
Rapid methods of landslide hazard mapping : Fiji case study
A landslide hazard probability map can help planners (1) prepare for, and/or mitigate against,
the effects of landsliding on communities and infrastructure, and (2) avoid or minimise the
risks associated with new developments. The aims of the project were to establish, by means
of studies in a few test areas, a generic method by which remote sensing and data analysis
using a geographic information system (GIS) could provide a provisional landslide hazard
zonation map. The provision of basic hazard information is an underpinning theme of the
UN’s International Decade for Natural Disaster Reduction (IDNDR). It is an essential
requirement for disaster preparedness and mitigation planning. This report forms part of BGS
project 92/7 (R5554) ‘Rapid assessment of landslip hazards’ Carried out under the ODA/BGS
Technology Development and Research Programme as part of the British Government’s
provision of aid to developing countries. It provides a detailed technical account of work
undertaken in a test area in Viti Levu in collaboration with Fiji Mineral Resources
Department. The study represents a demonstration of a methodology that is applicable to
many developing countries.
The underlying principle is that relationships between past landsliding events, interpreted
from remote sensing, and factors such as the geology, relief, soils etc provide the basis for
modelling where future landslides are most likely to occur. This is achieved using a GIS by
‘weighting’ each class of each variable (e.g. each lithology ‘class’ of the variable ‘geology’)
according to the proportion of landslides occurring within it compared to the regional
average. Combinations of variables, produced by summing the weights in individual classes,
provide ‘models’ of landslide probability. The approach is empirical but has the advantage
of potentially being able to provide regional scale hazard maps over large areas quickly and
cheaply; this is unlikely to be achieved using conventional ground-based geotechnical
methods.
In Fiji, landslides are usually triggered by intense rain storms commonly associated with
tropical cyclones. However, the regional distribution of landslides has not been mapped nor
is it known how far geology and landscape influence the location and severity of landsliding
events. The report discusses the remote sensing and GIS methodology, and describes the
results of the pilot study over an area of 713 km2 in south east Viti Levu. The landslide
model uses geology, elevation, slope angle, slope aspect, soil type, and forest cover as
inputs. The resulting provisional landslide hazard zonation map, divided into high, medium
and low zones of landslide hazard probability, suggests that whilst rainfall is the immediate
cause, others controls do exert a significant influence. It is recommended that consideration
be given in Fiji to implementing the techniques as part of a national strategic plan for
landslide hazard zonation mapping
Longitudinal metabolic and gut bacterial profiling of pregnant women with previous bariatric surgery
Objective Due to the global increase in obesity rates and success of bariatric surgery in weight reduction, an increasing number of women now present pregnant with a previous bariatric procedure. This study investigates the extent of bariatric-associated metabolic and gut microbial alterations during pregnancy and their impact on fetal development.
Design A parallel metabonomic (molecular phenotyping based on proton nuclear magnetic resonance spectroscopy) and gut bacterial (16S ribosomal RNA gene amplicon sequencing) profiling approach was used to determine maternal longitudinal phenotypes associated with malabsorptive/mixed (n=25) or restrictive (n=16) procedures, compared with women with similar early pregnancy body mass index but without bariatric surgery (n=70). Metabolic profiles of offspring at birth were also analysed.
Results Previous malabsorptive, but not restrictive, procedures induced significant changes in maternal metabolic pathways involving branched-chain and aromatic amino acids with decreased circulation of leucine, isoleucine and isobutyrate, increased excretion of microbial-associated metabolites of protein putrefaction (phenylacetlyglutamine, p-cresol sulfate, indoxyl sulfate and p-hydroxyphenylacetate), and a shift in the gut microbiota. The urinary concentration of phenylacetylglutamine was significantly elevated in malabsorptive patients relative to controls (p=0.001) and was also elevated in urine of neonates born from these mothers (p=0.021). Furthermore, the maternal metabolic changes induced by malabsorptive surgery were associated with reduced maternal insulin resistance and fetal/birth weight.
Conclusion Metabolism is altered in pregnant women with a previous malabsorptive bariatric surgery. These alterations may be beneficial for maternal outcomes, but the effect of elevated levels of phenolic and indolic compounds on fetal and infant health should be investigated further
Predictors of care-giver stress in families of preschool-aged children with developmental disabilities
Background This study examined the predictors, mediators and moderators of parent stress in families of preschool-aged children with developmental disability. Method One hundred and five mothers of preschool-aged children with developmental disability completed assessment measures addressing the key variables. Results Analyses demonstrated that the difficulty parents experienced in completing specific caregiving tasks, behaviour problems during these caregiving tasks, and level of child disability, respectively, were significant predictors of level of parent stress. In addition, parents’ cognitive appraisal of care-giving responsibilities had a mediating effect on the relationship between the child’s level of disability and parent stress. Mothers’ level of social support had a moderating effect on the relationship between key independent variables and level of parent stress. Conclusions Difficulty of care-giving tasks, difficult child behaviour during care-giving tasks, and level of child disability are the primary factors which contribute to parent stress. Implications of these findings for future research and clinical practice are outlined
A Hedged Monte Carlo Approach to Real Option Pricing
In this work we are concerned with valuing optionalities associated to invest
or to delay investment in a project when the available information provided to
the manager comes from simulated data of cash flows under historical (or
subjective) measure in a possibly incomplete market. Our approach is suitable
also to incorporating subjective views from management or market experts and to
stochastic investment costs. It is based on the Hedged Monte Carlo strategy
proposed by Potters et al (2001) where options are priced simultaneously with
the determination of the corresponding hedging. The approach is particularly
well-suited to the evaluation of commodity related projects whereby the
availability of pricing formulae is very rare, the scenario simulations are
usually available only in the historical measure, and the cash flows can be
highly nonlinear functions of the prices.Comment: 25 pages, 14 figure
Thermal annealing response following irradiation of a CMOS imager for the JUICE JANUS instrument
ESA's JUICE (JUpiter ICy moon Explorer) spacecraft is an L-class mission destined for the Jovian system in 2030. Its primary goals are to investigate the conditions for planetary formation and the emergence of life, and how does the solar system work. The JANUS camera, an instrument on JUICE, uses a 4T back illuminated CMOS image sensor, the CIS115 designed by Teledyne e2v. JANUS imager test campaigns are studying the CIS115 following exposure to gammas, protons, electrons and heavy ions, simulating the harsh radiation environment present in the Jovian system. The degradation of 4T CMOS device performance following proton fluences is being studied, as well as the effectiveness of thermal annealing to reverse radiation damage. One key parameter for the JANUS mission is the Dark current of the CIS115, which has been shown to degrade in previous radiation campaigns. A thermal anneal of the CIS115 has been used to accelerate any annealing following the irradiation as well as to study the evolution of any performance characteristics. CIS115s have been irradiated to double the expected End of Life (EOL) levels for displacement damage radiation (2×1010 protons, 10 MeV equivalent). Following this, devices have undergone a thermal anneal cycle at 100°C for 168 hours to reveal the extent to which CIS115 recovers pre-irradiation performance. Dark current activation energy analysis following proton fluence gives information on trap species present in the device and how effective anneal is at removing these trap species. Thermal anneal shows no quantifiable change in the activation energy of the dark current following irradiation
Use of cultivar resistance and crop rotation with Bacillus subtilis for clubroot control in canola
Non-Peer ReviewedThis study was conducted to assess additional strategies potentially complimentary to cultivar
resistance or biocontrol in control of clubroot. New granular Bacillus subtilis formulations
and a seed dressing method were developed to facilitate biofungicide delivery in field trials.
The granular formulations were applied in furrow during seeding at 50 kg/ha to a clubroot
resistant (CR) and susceptible (CS) canola cultivar, respectively, in three field trials. The seed
dressing applied approximately 1×105 to 5×106 cfu/seed doses of the biocontrol agent, and
was evaluated on the CS cultivar seeded to different crop-rotation scenarios where the plots
had a 1-year, 3-year, or 11-year break from last canola crop. Clubroot disease pressure was
high at all trial sites with disease severity indexes (DSI) ranging from 69% to 98% on the CS
cultivar. None of the granular formulations reduced clubroot substantially, whereas the CR
cultivar showed a high effect, reducing DSI to below 15% and doubling the yield over that of
CS cultivar. Plots of varying rotation showed a pattern of clubroot pathogen pressure, with
those of 1-year break from canola being the highest. The DSI for all rotational scenarios was
high, reaching 100% in short-rotation plots. Biofungicide seed dressing did not reduce DSI,
but longer crop rotation often reduced gall size slightly, showed much milder above-ground
damage, and increased the yield significantly relative to short rotation in two separate trials.
Even a 3-year break from canola was highly beneficial, with the yield doubled as opposed to
that with only 1-year break from canola
A comparison of precipitation and filtration-based SARS-CoV-2 recovery methods and the influence of temperature, turbidity, and surfactant load in urban wastewater
Wastewater-based epidemiology (WBE) has become a complimentary surveillance tool during the SARS-CoV-2 pandemic. Viral concentration methods from wastewater are still being optimised and compared, whilst viral recovery under different wastewater characteristics and storage temperatures remains poorly understood. Using urban wastewater samples, we tested three viral concentration methods; polyethylene glycol precipitation (PEG), ammonium sulphate precipitation (AS), and CP select™ InnovaPrep® (IP) ultrafiltration. We found no major difference in SARS-CoV-2 and faecal indicator virus (crAssphage) recovery from wastewater samples (n = 46) using these methods, PEG slightly (albeit non-significantly), outperformed AS and IP for SARS-CoV-2 detection, as a higher genome copies per litre (gc/l) was recorded for a larger proportion of samples. Next generation sequencing of 8 paired samples revealed non-significant differences in the quality of data between AS and IP, though IP data quality was slightly better and less variable. A controlled experiment assessed the impact of wastewater suspended solids (turbidity; 0–400 NTU), surfactant load (0–200 mg/l), and storage temperature (5–20 °C) on viral recovery using the AS and IP methods. SARS-CoV-2 recoveries were >20% with AS and  0.05), whilst surfactant and storage temperature combined were significant negative correlates (p < 0.001 and p < 0.05, respectively). In conclusion, our results show that choice of methodology had small effect on viral recovery of SARS-CoV-2 and crAssphage in wastewater samples within this study. In contrast, sample turbidity, storage temperature, and surfactant load did affect viral recovery, highlighting the need for careful consideration of the viral concentration methodology used when working with wastewater samples
Cytogenetic characterization of telomeres in the holocentric chromosomes of the lepidopteran Mamestra brassicae
Telomeres of the Mamestra brassica holocentric chromosomes were studied by Southern blotting, in-situ hybridization and Bal31 assay evidencing the presence of the telomeric (TTAGG)(n) repeat. Successively, molecular analysis of telomeres showed that TRAS1 transposable elements were present at the subtelomeric regions of autosomes but not in the NOR-bearing telomeres of the Z and W sex chromosomes. TRAS1 appeared to be transcriptionally active and non-methylated, as evaluated by RT-PCR and digestion with MspI and HpaII.
