    Cancer germline antigens and tumor-agnostic CD8+ T cell evasion

    Cancer germline antigens (CGAs) are expressed in immune-privileged germline tissues, while epigenetically silenced in somatic tissues. CGAs become re-expressed in tumors and can promote oncogenesis. Tumors prominently exploit mechanisms similar to those in germline tissues to shield from immunosurveillance. We hypothesize that CGAs contribute towards tumor escape from immune effector CD8+ T cells. For illustrative purposes, we assessed the co-presence or -absence of CGAs with these cells in multiple tumor types. Considering a broad array of CD8+ T cell evasive mechanisms, we exemplify the co-occurrence of gene transcripts of eight CGAs with those of adhesion molecules, endothelial cells, and/or the Wnt pathway. We present a novel concept of CGAs and their association with CD8+ T cell evasion, which may be relevant for future immunotherapeutic interventions

    Minimal residual disease (MRD) detection in acute lymphoblastic leukaemia based on fusion genes and genomic deletions

    Minimal residual disease (MRD) diagnostics are implemented in most clinical protocols for patients with acute lymphoblastic leukaemia (ALL) and are mostly performed using rearranged immunoglobulin (IG) and/or T-cell receptor (TR) gene rearrangements as molecular polymerase chain reaction targets. Unfortunately, in 5–10% of patients no or no sensitive IG/TR targets are available, and patients therefore cannot be stratified appropriately. In the present study, we used fusion genes and genomic deletions as alternative MRD targets in these patients, which retrospectively revealed appropriate MDR stratification in 79% of patients with no (sensitive) IG/TR target, and a different risk group stratification in more than half of the cases.</p

    This article draws on the author’s experience of teaching African Studies to undergraduate South African students in order to reflect on some of the key challenges facing teachers of African Studies, both in South Africa and beyond. In particular, it discusses challenges relating to teaching a field as contested as African Studies, looking at whether teaching African alternatives to mainstream African politics is helpful and at whether and how one can teach Africa in a way that encourages and develops critical thinking. The article also explores how the racial politics of the context in which one teaches African Studies inevitably affects the way in which students engage with the content of the course. While the article discusses these issues in relation to the South African higher education context in particular, implications for other contexts are also highlighted

    Blood, timber, and the state in West Kalimantan, Indonesia

    West Kalimantan (West Borneo) has a history of violent communal conflict.1 It also has extensive forests that have been looted for decades. The argument will be that these two are linked, but not by the grievances of the forest dwellers. Except in its first few days, the two main episodes of 1997 and 1999 were not driven mainly by grievances among marginal groups. Rather, explanations based on the ‘resource curse’ carry more weight. These focus attention on the contested nature of the state, rather than on rebellious activities of marginal groups. When state institutions were thrown into disarray by the sudden resignation of President Suharto in 1998, Dayak militants already close to state power rewrote the rules of local politics by demonstratively ‘cleansing’ certain areas of an unpopular immigrant minority. This theatrical manoeuvre impressed political rivals sufficiently to allow Dayaks to gain control over several timber-rich districts, which had a thriving black economy. Malays later imitated these techniques to stem the tide
