Optimised procedures for the cryopreservation of different species of Heterorhabditis
Il est démontré que les techniques mises au point pour la cryoconservation d'#Heterorhabditis bacterophora pouvaient être appliquées avec succès à différentes souches du complexe d'espèces #H. bacteriophora et aux espèces d'#Heterorhabditis présentes dans le Nord-Ouest de l'Europe. Par contre, ces techniques ne conviennent pas pour la cryoconservation des espèces irlandaises ou tropicales d'#Heterorhabditis. A donc été déterminée la survie après cryoconservation de différentes espèces d'#Heterorhabditis en fonction de la nature du traitement préalable à l'application du cryoconservateur (méthanol), de son intensité et de sa durée. Des différences inter- et intraspécifiques sont observées parmi les souches d'#Heterorhabditis et les conditions optimales pour la cryoconservation des souches d'Irlande, d'Europe du Nord-Ouest et tropicales ont été déterminées et sont décrites dans le présent article. Sediment routing and basin evolution in Proterozoic to Mesozoic east Gondwana: A case study from southern Australia
Sedimentary rocks along the southern margin of Australia host an important record of the break-up history of east Gondwana, as well as fragments of a deeper geological history, which collectively help inform the geological evolution of a vast and largely underexplored region. New drilling through Cenozoic cover has allowed examination of the Cretaceous rift-related Madura Shelf sequence (Bight Basin), and identification of two new stratigraphic units beneath the shelf; the possibly Proterozoic Shanes Dam Conglomerate and the interpreted Palaeozoic southern Officer Basin unit, the Decoration Sandstone. Recognition of these new units indicates an earlier basinal history than previously known. Lithostratigraphy of the new drillcore has been integrated with that published from onshore and offshore cores to present isopach maps of sedimentary cover on the Madura Shelf. New palynological data demonstrate progression from more localised freshwater-brackish fluvio-lacustrine clastics in the early Cretaceous (Foraminisporis wonthaggiensis – Valanginian to Barremian) to widespread topography-blanketing, fully marine, glauconitic mudrocks in the mid Cretaceous (Endoceratium ludbrookiae – Albian). Geochronology and Hf-isotope geochemistry show detrital zircon populations from the Madura Shelf are comparable to those from the southern Officer Basin, as well as Cenozoic shoreline and palaeovalley sediments in the region. The detrital zircon population from the Shanes Dam Conglomerate is defined by a unimodal ~1400 Ma peak, which correlates with directly underlying crystalline basement of the Madura Province. Peak ages of ~1150 Ma and ~1650 Ma dominate the age spectra of all other samples, indicating a stable sediment reservoir through much of the Phanerozoic, with sediments largely sourced from the Albany-Fraser Orogen and Musgrave Province (directly and via multiple recycling events). The Madura Shelf detrital zircon population differs from published data for the Upper CretaceousCeduna Delta to the east, indicating significant differences in sediment provenance and routing between the Ceduna Sub-basin and central Bight Basin
Doing race: how secondary school pupils in mainly white schools construct 'race'
This article examines how ‘race’ impacts upon the lives of young people who attend secondary schools in a mainly white British area of the United Kingdom. Schools Stand up 2 Racism (SSu2R), a Big Lottery research project, brought together a community partner – the Cheshire, Halton and Warrington Race and Equality Centre – and a team from Manchester Metropolitan University to investigate racism in Cheshire secondary schools. In an area where the population is over 93% ‘white British’, the sense that ‘there's nothing to be racist about in this school’ (Year 8 pupil) was found to be common. The three-year SSu2R study used a multimethod approach to study how students ‘do race’ in these schools, where the silent advocacy of a ‘colour-blind’ approach is promulgated through the popular rhetoric of ‘everyone is unique’ and ‘we should treat everyone the same’. Race is tackled only tangentially through the curriculum, accompanied by silences that close down discussion. Perhaps unsurprisingly given this context, forms of everyday racism are endemic and yet mostly unnoticed by staff or students. The article uses Bourdieu's concepts of ‘doxa’ and ‘the game’ to examine the nature of this silence
DLG4-related synaptopathy: a new rare brain disorder
PURPOSE: Postsynaptic density protein-95 (PSD-95), encoded by DLG4, regulates excitatory synaptic function in the brain. Here we present the clinical and genetic features of 53 patients (42 previously unpublished) with DLG4 variants.METHODS: The clinical and genetic information were collected through GeneMatcher collaboration. All the individuals were investigated by local clinicians and the gene variants were identified by clinical exome/genome sequencing.RESULTS: The clinical picture was predominated by early onset global developmental delay, intellectual disability, autism spectrum disorder, and attention deficit-hyperactivity disorder, all of which point to a brain disorder. Marfanoid habitus, which was previously suggested to be a characteristic feature of DLG4-related phenotypes, was found in only nine individuals and despite some overlapping features, a distinct facial dysmorphism could not be established. Of the 45 different DLG4 variants, 39 were predicted to lead to loss of protein function and the majority occurred de novo (four with unknown origin). The six missense variants identified were suggested to lead to structural or functional changes by protein modeling studies.CONCLUSION: The present study shows that clinical manifestations associated with DLG4 overlap with those found in other neurodevelopmental disorders of synaptic dysfunction; thus, we designate this group of disorders as DLG4-related synaptopathy.Genetics of disease, diagnosis and treatmen
Genetics of coronary artery calcification among African Americans, a meta-analysis
Background: Coronary heart disease (CHD) is the major cause of death in the United States. Coronary artery calcification (CAC) scores are independent predictors of CHD. African Americans (AA) have higher rates of CHD but are less well-studied in genomic studies. We assembled the largest AA data resource currently available with measured CAC to identify associated genetic variants.Methods: We analyzed log transformed CAC quantity (ln(CAC + 1)), for association with ~2.5 million single nucleotide polymorphisms (SNPs) and performed an inverse-variance weighted meta-analysis on results for 5,823 AA from 8 studies. Heritability was calculated using family studies. The most significant SNPs among AAs were evaluated in European Ancestry (EA) CAC data; conversely, the significance of published SNPs for CAC/CHD in EA was queried within our AA meta-analysis.Results: Heritability of CAC was lower in AA (~30%) than previously reported for EA (~50%). No SNP reached genome wide significance (p < 5E-08). Of 67 SNPs with p < 1E-05 in AA there was no evidence of association in EA CAC data. Geomorphic evidence of major sea-level fluctuations during marine isotope substage-5e, Cape Cuvier, Western Australia
A detailed geomorphologic and morphostratigraphic investigation of raised marine terraces at Cape Cuvier, Western Australia, reveals two morphologically distinct units. A lower, well-developed accretional reef terrace between 3 and 5.5 m above MLWS (mean low-water springs; hereafter denoted as "+") represents an extended interval of stable sea level. An upper erosional terrace and incipient coralgal rim between + 8.5 to 10.5 m represents a brief sea-level stillstand at this higher elevation. These features suggest the lower and upper terraces developed during discrete sea-level events. In an attempt to better define the timing of emplacement of each marine unit, 20 coral samples collected along vertical and lateral reef growth axis from both terraces were analysed with U-series dating. Unfortunately, all coral samples exhibited elevated δ234Uinitial values, suggesting that pervasive uptake of 234U-enriched uranium and 230Th thorium had occurred. Despite the shortcomings of absolute dating, a succession of events can be resolved though morphostratigraphic relationships. Comparison of the facies relationships, coral growth, and morphostratigraphic features between the lower and upper terraces indicates that an early to mid MIS 5e stillstand at + 3 to 5 m was followed by a late rise to + 8.5 to 10.5 m. This agrees with an emerging global view of MIS 5e sea-level history derived from stable carbonate platforms, rejecting the hypothesis that these higher sea-level benchmarks are an artefact of localized tectonic processes
