Capacidade combinatĂłria de hĂbridos de tomateiro de crescimento determinado, resistentes a Begomovirus e Tospovirus
O objetivo deste trabalho foi avaliar a capacidade combinatĂłria de linhagens de tomateiro com hĂĄbito de crescimento determinado, com resistĂȘncia mĂșltipla a espĂ©cies dos gĂȘneros Begomovirus e Tospovirus, e identificar combinaçÔes hĂbridas superiores. O experimento foi realizado em casa de vegetação, com 14 hĂbridos obtidos do cruzamento de sete linhagens femininas (grupo I) com duas linhagens masculinas (grupo II), em um dialelo parcial. As seguintes caracterĂsticas agronĂŽmicas foram avaliadas: produção total, produção precoce, massa mĂ©dia de frutos, formato, firmeza inicial e firmeza na meia-vida. As linhagens genitoras TOM-680 e TOM-682, do grupo I, se destacaram por exibir as maiores estimativas de capacidade geral de combinação (CGC) quanto Ă s caracterĂsticas produção total, produção precoce e massa mĂ©dia de frutos, enquanto a linhagem TOM-585 se destacou quanto aos maiores valores de produção total e massa mĂ©dia de frutos. No grupo II, a linhagem TOM-698 apresentou estimativas superiores de CGC para as caracterĂsticas de produção total, produção precoce, massa mĂ©dia dos frutos e firmeza inicial dos frutos. O hĂbrido TOM-682xTOM-698 apresenta as maiores estimativas de capacidade geral e especĂfica de combinação para produção total, produção precoce e meia-vida da firmeza, e Ă© o genĂłtipo mais promissor entre os materiais testados
Whole-genome sequencing reveals host factors underlying critical COVID-19
Critical COVID-19 is caused by immune-mediated inflammatory lung injury. Host genetic variation influences the development of illness requiring critical care1 or hospitalization2,3,4 after infection with SARS-CoV-2. The GenOMICC (Genetics of Mortality in Critical Care) study enables the comparison of genomes from individuals who are critically ill with those of population controls to find underlying disease mechanisms. Here we use whole-genome sequencing in 7,491 critically ill individuals compared with 48,400 controls to discover and replicate 23 independent variants that significantly predispose to critical COVID-19. We identify 16 new independent associations, including variants within genes that are involved in interferon signalling (IL10RB and PLSCR1), leucocyte differentiation (BCL11A) and blood-type antigen secretor status (FUT2). Using transcriptome-wide association and colocalization to infer the effect of gene expression on disease severity, we find evidence that implicates multiple genesâincluding reduced expression of a membrane flippase (ATP11A), and increased expression of a mucin (MUC1)âin critical disease. Mendelian randomization provides evidence in support of causal roles for myeloid cell adhesion molecules (SELE, ICAM5 and CD209) and the coagulation factor F8, all of which are potentially druggable targets. Our results are broadly consistent with a multi-component model of COVID-19 pathophysiology, in which at least two distinct mechanisms can predispose to life-threatening disease: failure to control viral replication; or an enhanced tendency towards pulmonary inflammation and intravascular coagulation. We show that comparison between cases of critical illness and population controls is highly efficient for the detection of therapeutically relevant mechanisms of disease
Implementation of immobilization accessories for positioning of small animals for radiation therapy
ABSTRACT Radiation therapy is a modality that is presenting great advances in veterinary medicine worldwide. In Brazil, this therapeutic option is underachieved. The success of this method depends on several factors, including the use of appropriate accessories for protection and immobilization of patients. For the immobilization of small animals during treatment, in addition to sedation and anesthesia, immobilizing accessories, similar to those used in human radiotherapy, are used. This study aimed to present proposals for immobilizing accessories adapted to the positioning of small animals in order to be used in radiotherapy planning. In order to achieve results, accessories were made and tested in a living animal simulating a radiotherapy planning, which proved to be favorable to use in positioning small animals undergoing radiotherapy and for implementation processes

Supplementary Material for: Proteomic Mapping of Dental Enamel Matrix from Inbred Mouse Strains: Unraveling Potential New Players in Enamel
<p>Enamel formation is a complex 2-step process by which proteins are
secreted to form an extracellular matrix, followed by massive protein
degradation and subsequent mineralization. Excessive systemic exposure
to fluoride can disrupt this process and lead to a condition known as
dental fluorosis. The genetic background influences the responses of
mineralized tissues to fluoride, such as dental fluorosis, observed in
A/J and 129P3/J mice. The aim of the present study was to map the
protein profile of enamel matrix from A/J and 129P3/J strains. Enamel
matrix samples were obtained from A/J and 129P3/J mice and analyzed by
2-dimensional electrophoresis and liquid chromatography coupled with
mass spectrometry. A total of 120 proteins were identified, and 7 of
them were classified as putative uncharacterized proteins and analyzed
in silico for structural and functional characterization. An interesting
finding was the possibility of the uncharacterized sequence Q8BIS2
being an enzyme involved in the degradation of matrix proteins. Thus,
the results provide a comprehensive view of the structure and function
for putative uncharacterized proteins found in the enamel matrix that
could help to elucidate the mechanisms involved in enamel
biomineralization and genetic susceptibility to dental fluorosis.</p
Characterization and anti-staphylococcal activity of the essential oil from Turnera subulata Sm.
The present study is pioneer in analyzing the chemical composition of the essential oil from aerial parts of Turnera subulata Sm. and evaluates their antibacterial activity against a panel of drug-resistant strains of Staphylococcus aureus. The aerial parts were kiln-dried and then powdered in mechanical mill, and was subjected to hydrodistillation in a Clevenger-type apparatus. The components were analyzed using a mass spectrometrycoupled gas chromatography (GC-MS), and the identification of substances was performed by comparison of the mass spectra obtained with the mass spectra of the database of the GC-MS and retention indices . The essential oil was evaluated using the method of broth dilution at concentrations ranging from 3.125”g/mL to 3200”g/mL. It was possible to identify 45 substances (92.1%) of the essential oils, and the major components were trans-caryophyllene (6.7%), citronellol (5.6%), sphatulenol (5.3%), α-cadinol (4.3%), n-tricosano (4.3%), geraniol (4.1%) and trans-geranilacetone (3.7%), n-pentacosano (3.5%), globulol (3.4%), caryophyllene oxide (3.2%). The essential oil of T. subulata Sm. showed effective antibacterial activity for the various strains of S. aureus tested, with the MIC values between 25 ”g/mL and 1600 ”g/mL. The study of the essential oil of Turnera subulata Sm. showed that it consists of a complex mixture of several classes of compounds having the sesquiterpenes as major constituents followed by monoterpenes, and showed an antibacterial activity significant front the strains tested
