    The Influence of Short-term Quercetin Supplementation on Peak Oxygen Uptake during Simulated Altitude Exposure in Trained Cyclists

    Endurance performance and peak oxygen uptake (VO2peak) decline as altitude increases. Some data exist suggesting that quercetin supplementation improves aerobic capacity in trained and untrained individuals at sea-level (normobaric normoxic conditions). Few studies have examined the effects of quercetin on endurance performance during simulated altitude exposure (normobaric hypoxic conditions). The purpose of this study was to determine the effects of consuming 1000 mg·day-1 of quercetin for two weeks on cycling VO2peak in healthy trained male cyclists performing under normobaric normoxic and hypoxic conditions (NP and HP, respectively). Fourteen age-matched healthy male subjects were randomized to either a placebo or quercetin group. Baseline and post supplementation VO2peak values were quantified during incremental cycling under normobaric normoxic (FIO2 = 20.9%) and normobaric hypoxic (FIO2 = 13.6 + 0.2%) conditions. Subjects consumed capsules twice daily with either 500 mg quercetin or placebo (Tang) for two weeks and were re-assessed. Test order was randomized and assessments were separated by 48-72 hours. At baseline, there were no significant differences between groups for VO2peak normobaric normoxic trials (NPbaseline Placebo vs. NPbaseline Quercetin = 58.7+8.8 and 61.5+7.9 ml . kg-1 . min-1, respectively, p = 0.541) and normobaric hypoxic trials (HPbaseline Placebo vs. HPbaseline Quercetin = 48.5+8.3 and 50.8+4.8 ml . kg-1 . min-1, respectively, p = 0.534). No significant differences were found after treatment (Placebo: NPbaseline vs. NPpost = 58.7+ 8.8 and 56.7+7.4 ml . kg-1 . min-1, respectively, p = 0.10; HPbaseline vs .HPpost = 48.5+8.3 and 47.1+8.3, respectively, p = 0.50; Quercetin: NPbaseline vs. NPpost = 61.5+7.9 and 62.4+7.3 ml . kg-1 . min-1, respectively, p = 0.558; HPbaseline vs. HPpost = 50.8+4.8 and 51.2+3.8 ml . kg-1 . min-1, respectively, p= 0.656. These data suggest that short-term quercetin supplementation at 1000 mg . day-1 does not affect VO2peak elicited via incremental maximal cycle testing under normobaric normoxic and hypoxic conditions in trained male cyclists. Furthermore, quercetin supplementation did not attenuate the decline in VO2peak that was evident in the normobaric hypoxic condition

    Gigantism in unique biogenic magnetite at the Paleocene-Eocene Thermal Maximum

    We report the discovery of exceptionally large biogenic magnetite crystals in clay-rich sediments spanning the Paleocene-Eocene Thermal Maximum (PETM) in a borehole at Ancora, New Jersey. Aside from previously-described abundant bacterial magnetofossils, electron microscopy reveals novel spearhead-like and spindle-like magnetite up to 4 μm long and hexaoctahedral prisms up to 1.4 μm long. Similar to magnetite produced by magnetotactic bacteria, these single-crystal particles exhibit chemical composition, lattice perfection, and oxygen isotopes consistent with an aquatic origin. Electron holography indicates single-domain magnetization despite their large crystal size. We suggest that the development of a thick suboxic zone with high iron bioavailability – a product of dramatic changes in weathering and sedimentation patterns driven by severe global warming – drove diversification of magnetite-forming organisms, likely including eukaryotes

    Proprietary Milk Protein Concentrate Reduces Joint Discomfort While Improving Exercise Performance in Non-Osteoarthritic Individuals

    Milk and dairy products are known to contain various bioactives with potential anti-inflammatory and immune modulating effects. Previous research has indicated that milk produced from hyperimmunized cows provided meaningful health benefits to individuals suffering from varying degrees of osteoarthritis and rheumatoid arthritis. PURPOSE: To examine the impact of a proprietary milk protein concentrate on joint discomfort and physical function, exercise performance, quality of life and various measures of affect. METHODS: Non-osteoarthritic men (42.5 ± 8.9 years, 176.7 ± 6.7 cm, 89.9 ± 11.5 kg, 28.8 ± 3.5 kg/m2, n = 30) and women (46.4 ± 9.6 years, 163.1 ± 8.2 cm, 72.2 ± 13.1 kg, 27.2 ± 5.3 kg/m2, n = 28) with mild to moderate knee pain during physical activity were randomized in a double-blind, placebo-controlled fashion to consume daily either a placebo (PLA) or a proprietary milk protein concentrate (MP) for a period of 8 weeks. Participants completed a functional capacity test pre and post-supplementation and completed visual analog scales (VAS), a 6-min walking test, WOMAC and profile of mood states (POMS) to assess changes in joint health, discomfort, physical function, exercise performance and affect. Mixed factorial ANOVA was used for all statistical analysis and significance was set a priori at p ≤ 0.05. RESULTS: Distance covered in the 6-min walking significantly improved 9% in MP versus 2% in PLA (mean difference: 110 ± 43 m, p = 0.012) in addition to 11 WOMAC components and 5 VAS reflective of MP improving joint health, discomfort and joint stability (all p \u3c 0.05 vs. PLA). Additionally, MP also improved overall perceptions of neck and back health compared to PLA. Serum and whole blood indicators of clinical safety remained within normal ranges throughout the study. CONCLUSIONS: In comparison to placebo, daily doses of proprietary milk protein concentrate yielded improvements in several components of the WOMAC, multiple visual analog scales indicative of joint health and stability, discomfort and pain, as well as significant improvements in distance covered during a 6-min walking test. Supplementation was well tolerated with no significant changes in whole-blood or serum markers of clinical safety

    A dynamic Archean sulfur cycle

    Many aspects of the Earth’s early sulfur cycle, from the origin of mass anomalous fractionations to the scale and degree of biological involvement, remain poorly understood. We have been studying the nature of multiple sulfur isotope (^(32)S, ^(33)S, and ^(34)S) signals using a novel combination of scanning high-resolution low-temperature superconductivity SQUID microscopy and secondary ion mass spectrometry (SIMS) techniques in a suite of samples from distal slope and basinal environments adjacent to a major Late Archean-age (~2.6-2.52 Ga) carbonate platform. Coupled with petrography, these techniques allow us to interrogate, at the same microscopic scale, the complex history of mineralization in samples containing diverse sulfide-bearing mineral components. Because of a general lack of Archean sulfate minerals, we focused our analyses on early diagenetic pyrite nodules, precipitated in surface sediments. This allows us to assay fractionations by controlling for isotope mass balance

    A single atom detector integrated on an atom chip: fabrication, characterization and application

    We describe a robust and reliable fluorescence detector for single atoms that is fully integrated into an atom chip. The detector allows spectrally and spatially selective detection of atoms, reaching a single atom detection efficiency of 66%. It consists of a tapered lensed single-mode fiber for precise delivery of excitation light and a multi-mode fiber to collect the fluorescence. The fibers are mounted in lithographically defined holding structures on the atom chip. Neutral 87Rb atoms propagating freely in a magnetic guide are detected and the noise of their fluorescence emission is analyzed. The variance of the photon distribution allows to determine the number of detected photons / atom and from there the atom detection efficiency. The second order intensity correlation function of the fluorescence shows near-perfect photon anti-bunching and signs of damped Rabi-oscillations. With simple improvements one can boost the detection efficiency to > 95%.Comment: 24 pages, 11 figure

    Effects of an Aqueous Extract of Withania somnifera on Strength Training Adaptations and Recovery: The STAR Trial

    Withania somnifera (Ashwagandha) is an Ayurvedic herb categorized as having “rasayana” (rejuvenator), longevity, and revitalizing properties. Sensoril® is a standardized aqueous extract of the roots and leaves of Withania somnifera. Purpose: To examine the impact of Sensoril® supplementation on strength training adaptations. Methods: Recreationally active men (26.5 ± 6.4 years, 181 ± 6.8 cm, 86.9 ± 12.5 kg, 24.5 ± 6.6% fat) were randomized in a double-blind fashion to placebo (PLA, n = 19) or 500 mg/d Sensoril® (S500, n = 19). Body composition (DEXA), muscular strength, power, and endurance, 7.5 km cycling time trial, and clinical blood chemistries were measured at baseline and after 12 weeks of supplementation and training. Subjects were required to maintain their normal dietary habits and to follow a specific, progressive overload resistance-training program (4-day/week, upper body/lower body split). 2 × 2 mixed factorial ANOVA was used for analysis and statistical significance was set a priori at p ≤ 0.05. Results: Gains in 1-RM squat (S500: +19.1 ± 13.0 kg vs. PLA +10.0 ± 6.2 kg, p = 0.009) and bench press (S500: +12.8 ± 8.2 kg vs. PLA: +8.0 ± 6.0 kg, p = 0.048) were significantly greater in S500. Changes in DEXA-derived android/gynoid ratio (S500: +0.0 ± 0.14 vs. PLA: +0.09 ± 0.1, p = 0.03) also favored S500. No other between-group differences were found for body composition, visual analog scales for recovery and affect, or systemic hemodynamics, however, only the S500 group experienced statistically significant improvements in average squat power, peak bench press power, 7.5 km time trial performance, and perceived recovery scores. Clinical chemistry analysis indicated a slight polycythemia effect in PLA, with no other statistical or clinically relevant changes being noted. Conclusions: A 500 mg dose of an aqueous extract of Ashwagandha improves upper and lower-body strength, supports a favorable distribution of body mass, and was well tolerated clinically in recreationally active men over a 12-week resistance training and supplementation period

    Optical coherence microscopy for the evaluation of a tissue-engineered artificial cornea

    A transparent artificial cornea derived from biological material is the ultimate goal of corneal research. Attempts at artificial corneal constructs produced from synthetic polymers have proved unsuccessful due to lack of biocompatibility and ability to integrate into the tissue. We have designed a corneal model derived from collagenous biological materials that has several advantages: it has low antigenicity and therefore small chance of eliciting an immune reaction, it can be broken down by the body’s own cells and gradually replaced over time by natural materials, and it may contain signaling information for native cells, thereby inducing normal phenotype and behavior. In addition, a transparent corneal model has the potential to be used for testing of novel ophthalmic drugs or gene therapy approaches, eliminating the need for animal testing. We have used an optical coherence microscope (OCM) to evaluate both the structure of our tissue constructs over time in culture and the optical properties of the tissue itself. This imaging technique promises to be an important diagnostic tool in our efforts to understand the influence of mechanical forces, cell phenotype, and soluble factors on the transparency of corneal tissue.     Superconductivity Induced by Bond Breaking in the Triangular Lattice of IrTe2

    IrTe2, a layered compound with a triangular iridium lattice, exhibits a structural phase transition at approximately 250 K. This transition is characterized by the formation of Ir-Ir bonds along the b-axis. We found that the breaking of Ir-Ir bonds that occurs in Ir1-xPtxTe2 results in the appearance of a structural critical point in the T = 0 limit at xc = 0.035. Although both IrTe2 and PtTe2 are paramagnetic metals, superconductivity at Tc = 3.1 K is induced by the bond breaking in a narrow range of x > xc in Ir1-xPtxTe2. This result indicates that structural fluctuations can be involved in the emergence of superconductivity.Comment: 10 pages, 4 figure
