    β-Catenin mutations and aberrant nuclear expression during endometrial tumorigenesis

    To clarify the possible role of aberrant β-catenin expression during endometrial tumorigenesis, a total of 199 cases of endometrial carcinomas (endometrioid type), as well as 37 cases of simple/complex and 32 of atypical hyperplasias, was consecutively investigated for immunohistochemistry, along with 141 normal endometrial samples distant from carcinomas. Of 199 carcinoma cases, 73 tumours as well as 44 normal samples were also analysed using a combination of RT-PCR and Southern blot hybridization, Western blot, and mutation gene assays. Cell membrane β-catenin immunoreactivity showed a stepwise decrease from normal, through atypical hyperplasia, to grade 3 carcinomas. In contrast, the nuclear accumulation in atypical hyperplasias and grade 1 or 2 tumours was higher than in simple/complex hyperplasias. Mutations in exon 3 of the β-catenin gene involving codons 33, 34, 37, 41, and 45 were observed in 16 (22.9%) of 70 endometrial carcinomas, as well as 3 (12.5%) of 24 atypical hyperplasias, the results being significantly related to low membrane and high nuclear immunoreactivity but not relative mRNA expression levels, suggesting that the gene mutations may be closely associated with changes in subcellular distribution. In addition to significant association between β-catenin mutation and low grade histological malignancy (P = 0.048), the mutations were detected in none of 15 and 13 (26%) of 50 tumours with or without lymph node metastasis, the difference being significant (P = 0.027). These findings suggest that β-catenin abnormalities may play an important role in a relatively early event during the endometrial hyperplasia-carcinoma sequence. © 2001 Cancer Research Campaign http://www.bjcancer.co

    Loss of DCC gene expression during ovarian tumorigenesis: relation to tumour differentiation and progression

    To clarify the possible role of DCC gene alteration in ovarian neoplasias, we immunohistochemically investigated 124 carcinomas, as well as 55 cystadenomas and 41 low malignant potential (LMP) tumours and compared the results with those for p53 protein expression, clinicopathological factors and survival. A combination of the reverse transcription polymerase chain reaction (RT-PCR) and Southern blot hybridization (SBH) for DCC mRNA levels was also carried out on 26 malignant, five LMP, eight benign and seven normal ovarian samples. Significantly decreased levels of overall DCC values in carcinomas compared with benign and LMP lesions were revealed by both immunohistochemical and RT-PCR/SBH assays. Similar findings were also noted when subdivision was into serous and mucinous categories. In carcinomas, reduction or loss of DCC expression was significantly related to the serous phenotype (serous vs non-serous, P< 0.0001), a high histological grade (grade 1 vs 2 or 3, P< 0.02) and a more advanced stage (FIGO stage I vs II/III/IV, P = 0.0083), while no association was noted with survival. Although p53 immunopositivity demonstrated significant stepwise increase from benign through to malignant lesions, there was no clear association with DCC score values. The results indicated that impaired DCC expression may play an important role in ovarian tumorigenesis.     Vortex pinning in Au-irradiated FeSe0.4Te0.6 crystals from the static limit to gigahertz frequencies

    Fe(Se,Te) is one of the simplest compounds of iron-based superconductors, but it shows a variety of vortex pinning phenomena both in thin-film and single-crystal forms. These properties are particularly important in light of its potential for applications ranging from the development of coated conductors for high-field magnets to topological quantum computation exploiting the Majorana particles found in the superconducting vortex cores. In this paper, we characterize the pinning properties of FeSe 0.4 Te 0.6 single crystals, both pristine and Au-irradiated, with a set of characterization techniques ranging from the static limit to the GHz frequency range by using dc magnetometry, ac susceptibility measurements of both the fundamental and the third harmonic signals, and by microwave coplanar waveguide resonator measurements of London and Campbell penetration depths. We observed signatures of single vortex pinning that can be modeled by a parabolic pinning potential, dissipation caused by flux creep, and a general enhancement of the critical current density after 320 MeV Au ion irradiation

    Josephson Plasma Resonance in Bi2Sr2CaCu2O8+yBi_2 Sr_2 Ca Cu_2 O_{8+y} with Spatially Dependent Interlayer-Phase Coherence

    We study the Josephson plasma resonance (JPR) in Bi2_2Sr2_2CaCu2_2O8+y_{8+y} (BSCCO) with spatially dependent interlayer-phase coherence (IPC). The half-irradiated BSCCO (HI-BSCCO), in which columnar defects are introduced only in a half of the sample, shows several resonance peaks, which are not simple superposition of the peaks in irradiated- and pristine-parts. JPR in HI-BSCCO changes its character from irradiated- to pristine-type at a crossover frequency (ωcr\omega_{cr}). We demonstrate that the one-dimensional \LSGE, which takes into account the spatial dependence of IPC, can reproduce most of the experimental findings including the presence of ωcr\omega_{cr}.Comment: 4 figure

    Sub-Arcsecond Imaging of 3C123:108-GHz Continuum Observations of the Radio Hotspots

    We present the results of sub-arcsecond 108 GHz continuum interferometric observations toward the radio luminous galaxy 3C123. Using multi-array observations, we utilize the high u,v dynamic range of the BIMA millimeter array to sample fully spatial scales ranging from 0.5" to 50". This allows us to make one-to-one comparisons of millimeter-wavelength emission in the radio lobes and hotspots to VLA centimeter observations at 1.4, 4.9, 8.4, and 15 GHz. At 108 GHz, the bright, eastern double hotspot in the southern lobe is resolved. This is only the second time that a multiple hotspot region has been resolved in the millimeter regime. We model the synchrotron spectra of the hotspots and radio lobes using simple broken power-law models with high energy cutoffs, and discuss the hotspot spectra and their implications for models of multiple hotspot formation.Comment: 16 pages, 3 Figures, ApJ Accepte

    Population sensitivity of acute flaccid paralysis and environmental surveillance for serotype 1 poliovirus in Pakistan: an observational study.

    BACKGROUND: To support poliomyelitis eradication in Pakistan, environmental surveillance (ES) of wastewater has been expanded alongside surveillance for acute flaccid paralysis (AFP). ES is a relatively new method of surveillance, and the population sensitivity of detecting poliovirus within endemic settings requires estimation. METHODS: Data for wild serotype 1 poliovirus from AFP and ES from January 2011 to September 2015 from 14 districts in Pakistan were analysed using a multi-state model framework. This framework was used to estimate the sensitivity of poliovirus detection from each surveillance source and parameters such as the duration of infection within a community. RESULTS: The location and timing of poliomyelitis cases showed spatial and temporal variability. The sensitivity of AFP surveillance to detect serotype 1 poliovirus infection in a district and its neighbours per month was on average 30.0% (95% CI 24.8-35.8) and increased with the incidence of poliomyelitis cases. The average population sensitivity of a single environmental sample was 59.4% (95% CI 55.4-63.0), with significant variation in site-specific estimates (median varied from 33.3-79.2%). The combined population sensitivity of environmental and AFP surveillance in a given month was on average 98.1% (95% CI 97.2-98.7), assuming four samples per month for each site. CONCLUSIONS: ES can be a highly sensitive supplement to AFP surveillance in areas with converging sewage systems. As ES for poliovirus is expanded, it will be important to identify factors associated with variation in site sensitivity, leading to improved site selection and surveillance system performance
