Identification of the chromosome complement and the spontaneous 1R/1V translocations in allotetraploid Secale cereale × Dasypyrum villosum hybrids through cytogenetic approaches
Genome modifications that occur at the initial interspecific hybridization event are dynamic and can be consolidated during the process of stabilization in successive generations of allopolyploids. This study identifies the number and chromosomal location of ribosomal DNA (rDNA) sites between Secale cereale, Dasypyrum villosum, and their allotetraploid S. cereale × D. villosum hybrids. For the first time, we show the advantages of FISH to reveal chromosome rearrangements in the tetraploid Secale × Dasypyrum hybrids. Based on the specific hybridization patterns of ribosomal 5S, 35S DNA and rye species-specific pSc200 DNA probes, a set of genotypes with numerous Secale/Dasypyrum translocations of 1R/1V chromosomes were identified in successive generations of allotetraploid S. cereale × D. villosum hybrids. In addition we analyse rye chromosome pairs using FISH with chromosome-specific DNA sequences on S. cereale × D. villosum hybrids
Zawartość wybranych składników odżywczych w ziarnie pszenicy, jęczmienia i owsa z uprawy ekologicznej i konwencjonalnej
The success of organic farming results from the need to produce top-quality food and,
additionally, to protect the natural environment by applying eco-friendly agricultural practices
and abandoning synthetic fertilizers and chemical pesticides. The aim of this study
was to assess the impact of organic and conventional production systems on the content
of minerals and fatty acids in grain of oat, wheat and hulled and naked barley.
Wheat grain from organic farming was characterized by a higher content of Mn and
by significantly higher content of Fe, Zn, Ca and Mg when compared to grain originating
from conventional farming. The increased availability of potassium in soil, caused by the
applied potassium salt fertilization, was reflected in a higher content of this macronutrient
in grain of all cereals from conventional cultivation. The tillage system was found not to
exert the same effect on the content of Ca and Mg in all the cereals examined. Lower
content of iron and zinc was determined in organic grains of barley and oat. Grain from
the conventional cropping system (except naked barley) proved to be richer in lipids. Crude
oil of organic cereals was richer in the most valuable polyunsaturated fatty acids than
that of the conventionally-grown crops The study demonstrated a stronger dependency between
the level of the elements examined and cereal species than between the level of elements
and cultivation system.Popularność rolnictwa ekologicznego wynika z poszukiwania żywności o najwyżej jakości
oraz z potrzeby ochrony środowiska naturalnego, przez stosowanie zabiegów agrotechnicznych
przyjaznych ekosystemowi. Celem pracy była ocena wpływu ekologicznego
i konwencjonalnego systemu uprawy na zawartość składników mineralnych i kwasów tłuszczowych
w ziarnie owsa, pszenicy oraz jęczmienia oplewionego i nagiego.
Ziarniaki pszenicy z uprawy ekologicznej zawierały więcej Mn oraz istotnie więcej Fe,
Zn, Ca i Mg w porównaniu z pochodzącymi z uprawy konwencjonalnej. Zwiększona dostępność
potasu w glebie, spowodowana zastosowanym nawożeniem w postaci soli potasowej,
odzwierciedla się wyższą zawartością tego makroskładnika w ziarniakach wszystkich
zbóż z uprawy konwencjonalnej. Nie stwierdzono jednokierunkowego wpływu systemu
uprawy na zawartość Ca i Mg w przypadku wszystkich badanych zbóż, natomiast niższą
zawartość żelaza i cynku stwierdzono w ziarniakach jęczmienia i owsa z upraw ekologicznych.
Ziarniaki z uprawy konwencjonalnej były zasobniejsze w tłuszcz (wyjątek stanowił
jęczmień nagi). Tłuszcz zbóż ekologicznych był bogatszy w większość cennych kwasów
wielonienasyconych w porównaniu ze zbożami konwencjonalnymi. Obserwowano silniejszą
zależność zawartości badanych składników od gatunku zboża niż od systemu uprawy
Macroelements and beta-glucan content in hully and hulless cultivars of barley and oats in relation to chemical protection
Badano zmiany zawartości β-glukanów i makro składników w oplewionych i nagoziarnistych odmianach jęczmienia (odmian Antek, Rastik) i owsa (odmian Cwał, Cacko). W doświadczeniu uwzględniono 2 poziomy ochrony chemicznej: minimalną - zaprawa nasienna Oxafun T 75 DS/WS i herbicyd Mustang 306 SE oraz intensywną - zaprawa nasienna, herbicyd, fungicyd Folicur Plus 375 EC i insektycyd Karate Zeon 050 CS. Stwierdzono, że zróżnicowany poziom ochrony chemicznej roślin nie wpłynął istotnie na zmiany zawartości zarówno badanych makroskładników, jak i β-glukanów. W ziarniakach badanych odmian jęczmienia, zarówno oplewionych, jak i nagoziarnistych, wykazano wyższy poziom β-glukanów w porównaniu z ziarniakami owsa.Changes in the content of β-glucans and macroelements in hully and hulless barley (Antek and Rastik cv.) and oats (Cwał and Cacko cv.) cultivars were monitored in the study. Two chemical protection levels were considered in the experiments: minimum - seed dressing with Oxafun T 75 DS/WS and herbicide Mustang 306 SE, as well as intensive - seed dressing, herbicide, fungicide Folicur Plus 375 EC and insecticide Karate Zeon 050 CS. It was found that the different levels of plant chemical protection did not significantly affect the changes in the content of the macroelements or β-glucans. Kernels of the barley cultivars, both hully and hulless, were characterized by a higher content of higher β-glucans than oats kernels
Antioxidant activity and content of resorcinol of lipids in hybrids strain of winter triticale
Materiał badawczy stanowiły ziarniaki 5 nowych rodów mieszańcowych pszenżyta ozimego oraz ich formy rodzicielskie. Techniką TLC i spektrometrią UV-VIS potwierdzono obecność alkilorezorcynoli w otrzymanych z ziarna ekstraktach i oznaczono ich zawartość. Poziom alkilorezorcynoli w 3 rodach mieszańcowych był istotnie niższy, w porównaniu z obu formami rodzicielskimi, a w 2 pozostałych przewyższał te formy. W ekstraktach lipidów rezorcynolowych określono aktywność przeciwutleniającą metodą neutralizacji wolnych rodników wobec DPPH• (2,2-difenylo-1pikrylohydrazyl). Ekstrakty alkilorezorcynoli mieszańców charakteryzowały się wyższą aktywnością przeciwutleniającą w porównaniu z ekstraktami otrzymanymi z ich form matecznych, ale niższą niż form ojcowskich.The grains of 5 new strains hybrid of winter triticale and their parental forms were the material for the study. The samples was analyzed by thin-layer chromatography and spectrometry UV -VIS. The level of alkylresorcinols in the 3 strains hybrids the greater part was significantly less in compare with the both parental forms, and in the 2 others was higher than that forms. The antioxidant properties of alkylresorcinols were examined using free radical scavenging method against stable 2,2 - diphenyl - 1- picrylhydrazyl radical (DPPH*). The hybrids displayed higher antioxidant activity as compared to the triticale maternal components, but lower than paternal forms
Structural properties of the human acidic ribosomal P proteins forming the P1-P2 heterocomplex.
The ribosome has a morphologically distinct structural feature called the stalk, recognized as a vital element for its function. The ribosomal P proteins constitute the main part of the eukaryotic ribosomal stalk, forming a pentameric structure P0-(P1-P2)(2). The group of P1/P2 proteins in eukaryotes is very diverse, and in spite of functional and structural similarities they do not fully complement one another, probably constituting an adaptive feature of the ribosome from a particular species to diverse environmental conditions. The functional differences among the P1/P2 proteins were analysed in vivo several times; however, a thorough molecular characterization was only done for the yeast P1/P2 proteins. Here, we report a biophysical analysis of the human P1 and P2 proteins, applying mass spectrometry, CD and fluorescence spectroscopy, cross-linking and size exclusion chromatography. The human P1/P2 proteins form stable heterodimer, as it is the case for P1/P2 from yeast. However, unlike the yeast complex P1A-P2B, the human P1-P2 dimer showed a three-state transition mechanism, suggesting that an intermediate species may exist in solution
