Ideas for the Provision of Ontology Access in Grid Environments
Ontologies are the backbone of the Semantic Web. Current grid architectures do not consider their usage, and there are no protocols nor standards in the Grid community for dealing with them. Therefore, the provision of appropriate means for accessing, querying and using ontologies effectively is a key factor if we want to enrich the current grid with semantic technologies and to support progress towards the next generation Grid, that is, the Semantic Grid
Diagnostic concordance of DSM-IV and DSM-5 Posttraumatic Stress Disorder (PTSD) in a clinical sample
The present study aims to analyze diagnostic concordance between the DSM-IV and the DSM-5 for posttraumatic stress disorder (PTSD) diagnostic criteria and their different groups of symptoms.
Furthermore, analyses are conducted to establish the features of participants with no concordant diagnoses. Method: The study assessed 166 people over 18 who had experienced at least one traumatic event. PTSD diagnosis was established using the Global Scale for Posttraumatic Stress (EGEP), a self-report measure to assess PTSD.
Results: The presence of cognitive avoidance was a determinant in the PTSD DSM-5 diagnosis (86% positive predictive value). The analysis of the non-concordant individuals revealed that individuals who were diagnosed according to the DSM-IV criteria but not the DSM-5 criteria were primarily indirect victims. Conversely, individuals who were diagnosed with the DSM-5 criteria and not with the DSM-IV criteria presented cognitive avoidance and alterations in cognition not included in the DSM-IV criteria.
Conclusions: A withinsubjects concordance analysis showed high agreement for PTSD diagnosis between the two classifi cations. Differences between the diagnoses are due to the new defi nition of C (avoidance) and D (negative alterations in cognitions and mood) in the DSM-
Blockade of the Interaction of Calcineurin with FOXO in Astrocytes Protects Against Amyloid-βInduced Neuronal Death
Astrocytes actively participate in neuro-inflammatory processes associated to Alzheimer’s disease (AD), and other brain pathologies. We recently showed that an astrocyte-specific intracellular signaling pathway involving an interaction of the phosphatase calcineurin with the transcription factor FOXO3 is a major driver in AD associated pathological inflammation, suggesting a potential new druggable target for this devastating disease. We have now developed decoy molecules to interfere with calcineurin/FOXO3 interactions, and tested them in astrocytes and neuronal co-cultures exposed to amyloid-β (Aβ) toxicity.
We observed that interference of calcineurin/FOXO3 interactions exerts a protective action against A-induced neuronal death and favors the production of a set of growth factors that we hypothesize form part of a cytoprotective pathway to resolve inflammation. Furthermore, interference of the A-induced interaction of calcineurin with FOXO3 by decoy compounds significantly decreased amyloid-β protein precursor (AβPP) synthesis, reduced the AβPP amyloidogenic pathway, resulting in lower Alevels, and blocked the expression of pro-inflammatory cytokines TNFα and IL-6 in astrocytes. Collectively, these data indicate that interrupting pro-inflammatory calcineurin/FOXO3 interactions in astrocytes triggered by Aβ accumulation in brain may constitute an effective new therapeutic approach in AD. Future studies with intranasal delivery, or brain barrier
permeable decoy compounds, are warranted
Executing Evaluations over Semantic Technologies using the SEALS Platform
The SEALS European project aims to develop an infrastructure for the evaluation of semantic technologies. This paper presents in detail the approach followed to automate the execution of these evaluations in the infrastructure. To materialize this approach, we have dened the entities managed by the infrastructure and their life cycle, the process followed to execute evaluations, the management of the computing resources that form the execution infrastructure, and how tools can be integrated with the infrastructure
Towards an Infrastructure for the Evaluation of Semantic Technologies
This paper presents and discusses the current development status of the SEALS Platform, a lasting reference infrastructure for semantic technology evaluation. It describes the different entities managed by the platform and the ontology-based model that has been defined to represent them; it also provides an overview of the platform architecture. In addition, it presents the different challenges faced during the development of the SEALS Platform and a use scenario of the platform that supports the execution of evaluation campaigns over semantic technologies
