Plasmid segregation: spatial awareness at the molecular level
In bacteria, low-copy number plasmids ensure their stable inheritance by partition loci (par), which actively distribute plasmid replicates to each side of the cell division plane. Using time-lapse fluorescence microscopic tracking of segregating plasmid molecules, a new study provides novel insight into the workings of the par system from Escherichia coli plasmid R1. Despite its relative simplicity, the plasmid partition spindle shares characteristics with the mitotic machinery of eukaryotic cells
Functional Compost
The aim of the research program Functional Compost is to develop and test compost, which have been enriched with chitin, for plant growth promoting properties and to recognise specific mechanisms. Two types of compost were included in the program: source separated biodegradable municipal solid waste compost (DM = 62 %) and garden and park waste compost (DM = 66 %). Chitin was added in trace amounts during the maturity phase, combined with two levels of trace amounts immediately before adding the compost to the growth medium. The research program includes several parallel experiments. In experiment I, compost (20 vol. %) was added to soil (no plants) and incubated at 15 C for 5 month, under regular determination of microbial respiration and gross and net N mineralization. There was a significant increase in respiration due to chitin enrichment, which could not be explained by the amount of C derived from the chitin, which therefore suggest a priming effect. The N analyses are still being processed in the laboratory, but data are expected to be available at the conference. In experiment II, compost was mixed with sand, put into pots in a climate chamber, and spring barley seeds infected with Fusarium culmorum were sown in the pots. After 3 weeks of growth, the health of the plants was determined, and the chitinase activity in the sand was measured. The health of the plants and the chitinase activity was significantly higher in the treatments receiving municipal waste compared to the treatments receiving garden waste compost. However, there was no clear effect of the chitin enrichment. Additionally, the microbial community structure of the two types of compost, with and without early chitin, was determined by Denaturing Gradient Gel Electrophoresis (DGGE). There was a clear separation between compost types, and with or without early chitin amendment. Experiment III is a regular growth experiment, and is running right now. Compost has been incorporated into soil, put into pots in the greenhouse, and spring barley is grown for 2 month before determination for wet and dry weight and N uptake. Draft Genome Sequence of <em>Parabacteroides goldsteinii</em> with Putative Novel Metallo-β-Lactamases Isolated from a Blood Culture from a Human Patient
Parabacteroides goldsteinii was isolated from a blood culture. Genomic DNA was sequenced using a MiSeq sequencer and assembled using the SPAdes genome assembler. The draft genome sequence was 6,851,868 bp, spanning 282 contigs of 5,253 coding sequences, 66 tRNAs, and 5 rRNAs. Several putative novel metallo-β-lactamases were discovered
Impact of Chromosomal Architecture on the Function and Evolution of Bacterial Genomes
The bacterial nucleoid is highly condensed and forms compartment-like structures within the cell. Much attention has been devoted to investigating the dynamic topology and organization of the nucleoid. In contrast, the specific nucleoid organization, and the relationship between nucleoid structure and function is often neglected with regard to importance for adaption to changing environments and horizontal gene acquisition. In this review, we focus on the structure-function relationship in the bacterial nucleoid. We provide an overview of the fundamental properties that shape the chromosome as a structured yet dynamic macromolecule. These fundamental properties are then considered in the context of the living cell, with focus on how the informational flow affects the nucleoid structure, which in turn impacts on the genetic output. Subsequently, the dynamic living nucleoid will be discussed in the context of evolution. We will address how the acquisition of foreign DNA impacts nucleoid structure, and conversely, how nucleoid structure constrains the successful and sustainable chromosomal integration of novel DNA. Finally, we will discuss current challenges and directions of research in understanding the role of chromosomal architecture in bacterial survival and adaptation
A Bipolar Spindle of Antiparallel ParM Filaments Drives Bacterial Plasmid Segregation
To ensure their stable inheritance by daughter cells during cell division, bacterial low copy-number plasmids make simple DNA segregating machines that use an elongating protein filament between sister plasmids. In the ParMRC system of Escherichia coli R1 plasmid, ParM, an actin-like protein, forms the spindle between ParRC complexes on sister plasmids. Using a combination of structural work and total internal reflection fluorescence microscopy, we show that ParRC bound and could accelerate growth at only one end of polar ParM filaments, mechanistically resembling eukaryotic formins. The architecture of ParM filaments enabled two ParRC-bound filaments to associate in an antiparallel orientation, forming a bipolar spindle. The spindle elongated as a bundle of at least two antiparallel filaments, thereby pushing two plasmid clusters towards the poles

textabstractWe propose a consistent utility-based framework to jointly explain a household's decisions on purchase incidence, brand choice and purchase quantity. The approach differs from other approaches, currently available in the literature, as it is able to take into account consumption dynamics. In the model, households derive utility from consumption, and they relate their purchase behavior to consumption planning. We illustrate our model for yogurt purchases, and show that our model yields important additional insights. One such insight is that the reservation price of households is not fixed, but depends on the available inventory stock. Furthermore, we find that promotional activities increase sales through more purchases in the product category and brand switching, but the effect through larger purchase quantities is limited
