    Highly-stable, flexible delivery of microjoule-level ultrafast pulses in vacuumized anti-resonant hollow-core fibers for active synchronization

    We demonstrate the stable and flexible light delivery of multi-{\mu}J, sub-200-fs pulses over a ~10-m-long vacuumized anti-resonant hollow-core fiber (AR-HCF), which was successfully used for high-performance pulse synchronization. Compared with the pulse train launched into the AR-HCF, the transmitted pulse train out of the fiber exhibits excellent stabilities in pulse power and spectrum, with pointing stability largely improved. The walk-off between the fiber-delivery and the other free-space-propagation pulse trains, in an open loop, was measured to be <6 fs root-mean-square (RMS) over 90 minutes, corresponding to a relative optical-path variation of <2x10-7. This walk-off can be further suppressed to ~2 fs RMS simply using an active control loop, highlighting the great application potentials of this AR-HCF set-up in large-scale laser and accelerator facilities

    Ubiquitin-Proteasome-Collagen (CUP) Pathway in Preterm Premature Rupture of Fetal Membranes

    Spontaneous preterm birth (sPTB) occurs before 37 gestational weeks, with preterm premature rupture of the membranes (PPROM) and spontaneous preterm labor (sPTL) as the predominant adverse outcomes. Previously, we identified altered expression of long non-coding RNAs (lncRNAs) and message RNAs (mRNAs) related to the ubiquitin proteasome system (UPS) in human placentas following pregnancy loss and PTB. We therefore hypothesized that similar mechanisms might underlie PPROM. In the current study, nine pairs of ubiquitin-proteasome-collagen (CUP) pathway–related mRNAs and associated lncRNAs were found to be differentially expressed in PPROM and sPTL. Pathway analysis showed that the functions of their protein products were inter-connected by ring finger protein. Twenty variants including five mutations were identified in CUP-related genes in sPTL samples. Copy number variations were found in COL19A1, COL28A1, COL5A1, and UBAP2 of sPTL samples. The results reinforced our previous findings and indicated the association of the CUP pathway with the development of sPTL and PPROM. This association was due not only to the genetic variation, but also to the epigenetic regulatory function of lncRNAs. Furthermore, the findings suggested that the loss of collagen content in PPROM could result from degradation and/or suppressed expression of collagens

    Efficient generation of human primordial germ cell-like cells from pluripotent stem cells in a methylcellulose-based 3D system at large scale

    Background The mechanisms underlying human germ cell development and infertility remain largely unknown due to bioethical issues and the shortage of experimental materials. Therefore, an effective in vitro induction system of human primordial germ-like cells (hPGCLCs) from human pluripotent stem cells (hPSC) is in high demand. The current strategies used for the generation of hPGCLCs are not only costly but also difficult to perform at a large scale, thereby posing barriers to further research. In this study, we attempted to solve these problems by providing a new 3D culture system for hPGCLC differentiation. Methods The efficiency and relative yield of a methylcellulose (MC)-based 3D hPGCLC induction system were first compared with that of a conventional U96 system. Then, we examined the gene expression of germ cell marker genes and the key epigenetic modifications of the EpCAM-/INTEGRINα6-high cells from the 3D MC induction system and the U96 system via quantitative PCR and immunofluorescence. Finally, the reliability of the MC-based 3D hPGCLC induction system was evaluated via the generation of induced pluripotent stem cells (iPSCs) from the testicular cells of one patient with obstructive azoospermia (OA) and followed by the subsequent differentiation of iPSCs into the germ cell lineage. Results In the present study, we demonstrated that the 3D MC induction system combined with low-cell attachment plates facilitated the generation of hPGCLCs at a large scale. We found that the hPGCLCs generated via the MC system shared similar characteristics to that via the U96 system in terms of the gene expression profiles, germ cell-specific markers, epigenetic modification states and cellular states. In addition, hPGCLCs from iPSCs derived from one OA patient were generated with high efficiency via the present 3D MC induction system. Discussion The in vitro induction of hPGCLCs from human embryonic stem cells (hESCs)/human induced pluripotent stem cells (hiPSCs) has significant implications in exploring the underlying mechanisms of the origin and specification of hPGCs and the epigenetic programming of the human germ line as well as treating male infertility. Here, we developed a simple and efficient 3D induction system to generate hPGCLCs from hESCs/hiPSCs at a large scale, which facilitated the study of human germ cell development and stem cell-based reproductive medicine

    Redefining cardiac biomarkers in predicting mortality and adverse outcomes of inpatients with COVID-19

    The prognostic power of circulating cardiac biomarkers, their utility and pattern of release in coronavirus disease 2019 (COVID-19) patients have not been clearly defined. In this multi-centered retrospective study, we enrolled 3,219 patients with diagnosed COVID-19 admitted to 9 hospitals from December 31, 2019 to March 4, 2020, to estimate the associations and prognostic power of circulating cardiac injury markers with the poor outcomes of COVID-19. In the mixed-effect Cox model, after adjusting for age, gender and comorbidities, the adjusted hazard ratios of 28-day mortality for high-sensitivity cardiac troponin I (hs-cTnI) was 7.12 (95%CI, 4.60-11.03; P&lt;0.001), NT-proB-type natriuretic peptide (NT-proBNP) was 5.11 (95%CI, 3.50-7.47; P&lt;0.001), CK-MB was 4.86 (95%CI, 3.33-7.09; P&lt;0.001), myoglobin was 4.50 (95%CI, 3.18-6.36; P &lt; 0.001), and CK was 3.56 (95%CI, 2.53-5.02; P &lt; 0.001). The cutoffs of those cardiac biomarkers for effective prognosis of 28-day mortality of COVID-19 were found to be much lower than for regular heart disease at about 49% of the currently recommended thresholds. Patients with elevated cardiac injury markers above the newly established cutoffs were associated with significantly increased risk of COVID-19 death. In conclusion, cardiac biomarker elevations are significantly associated with 28-day death in patients with COVID-19. The prognostic cutoffs for of these values might be much lower than the current reference standards. These findings can assist better management of COVID-19 patients to improve outcomes. Importantly, the newly established cutoff levels of COVID-19 associated cardiac biomarkers may serve as useful criteria for the future prospective studies and clinical trials

    Changes in the potential multiple cropping system in response to climate change in China from 1960-2010.

    The multiple cropping practice is essential to agriculture because it has been shown to significantly increase the grain yield and promote agricultural economic development. In this study, potential multiple cropping systems in China are calculated based on meteorological observation data by using the Agricultural Ecology Zone (AEZ) model. Following this, the changes in the potential cropping systems in response to climate change between the 1960s and the 2010s were subsequently analyzed. The results indicate that the changes of potential multiple cropping systems show tremendous heterogeneity in respect to the spatial pattern in China. A key finding is that the magnitude of change of the potential cropping systems showed a pattern of increase both from northern China to southern China and from western China to eastern China. Furthermore, the area found to be suitable only for single cropping decreased, while the area suitable for triple cropping increased significantly from the 1960s to the 2000s. During the studied period, the potential multiple cropping index (PMCI) gap between rain-fed and irrigated scenarios increased from 18% to 24%, which indicated noticeable growth of water supply limitations under the rain-fed scenario. The most significant finding of this research was that from the 1960s to the 2000s climate change had led to a significant increase of PMCI by 13% under irrigated scenario and 7% under rain-fed scenario across the whole of China. Furthermore, the growth of the annual mean temperature is identified as the main reason underlying the increase of PMCI. It has also been noticed that across China the changes of potential multiple cropping systems under climate change were different from region to region

    Mechanism of Spatiotemporal Air Quality Response to Meteorological Parameters: A National-Scale Analysis in China

    The air quality over China exhibits seasonal and regional variation, resulting from heterogeneity in industrialization, and is highly affected by variability in meteorological conditions. We performed the first national-scale exploration of the relationship between the Air Pollution Index (API) and multiple meteorological parameters in China, using partial correlation and hierarchical cluster analyses. Relative humidity, wind speed, and temperature were the dominant factors influencing air quality year-round, due to their significant effects on pollutant dispersion and/or transformation of pollutants. The response of the API to single or multiple meteorological factors varied among cities and seasons, and a regional clustering of response mechanisms was observed, particularly in winter. Clear north&ndash;south differentiation was detected in the mechanisms of API response to relative humidity and wind speed. These findings provide insight into the spatiotemporal variation in air quality sensitivity to meteorological conditions, which will be useful for implementing regional air pollution control strategies

    Assessment of Urban Heat Risk in Mountain Environments: A Case Study of Chongqing Metropolitan Area, China

    For urban climatic environments, the urban heat island (UHI) effect resulting from land use and land cover change (LUCC) caused by human activities is rapidly becoming one of the most notable characteristics of urban climate change due to urban expansion. UHI effects have become a significant barrier to the process of urbanization and sustainable development of the urban ecological environment. Predicting the spatial and temporal patterns of the urban heat environment from the spatial relationship between land use and land surface temperature (LST) is key to predicting urban heat environment risk. This study established an Urban Heat Environment Risk Model (UHERM) as follows. First, the urban LST was normalized and classified during three different periods. Second, a Markov model was constructed based on spatio-temporal change in the urban heat environment between the initial year (2005) and middle year (2010), and then a cellular automata (CA) model was used to reveal spatial relationships between the urban heat environments of the two periods and land use in the initial year. The spatio-temporal pattern in a future year (2015) was predicted and the accuracy of the simulation was verified. Finally, the spatio-temporal pattern of urban heat environment risk was quantitatively forecasted based on the decision rule for the urban heat environment risk considering both the present and future status of the spatial characteristics of the urban heat environment. The MODIS LST product and LUCC dataset retrieved from remote sensing images were used to verify the accuracy of UHERM and to forecast the spatio-temporal pattern of urban heat environment risk during the period of 2015&ndash;2020. The results showed that the risk of urban heat environment is increasing in the Chongqing metropolitan area. This method for quantitatively evaluating the spatio-temporal pattern of urban heat environment risk could guide sustainable growth and provide effective theoretical and technical support for the regulation of urban spatial structure to minimize urban heat environment risk
