Biomimetic and Bioactive Small Diameter Tubular Scaffolds for Vascular Tissue Engineering
The present work aimed at the production and characterization of small caliber biomimetic and bioactive tubular scaffolds, which are able to favor the endothelialization process, and therefore potentially be suitable for vascular tissue engineering. The tubular scaffolds were produced using a specially designed mold, starting from a gelatin/gellan/elastin (GGE) blend, selected to mimic the composition of the extracellular matrix of native blood vessels. GGE scaffolds were obtained through freeze-drying and subsequent cross-linking. To obtain systems capable of promoting endothelization, the scaffolds were functionalized using two different bioactive peptides, Gly-Arg-Gly-Asp-Ser-Pro (GRGSDP) and Arg-Glu-Asp-Val (REDV). A complete physicochemical, mechanical, functional, and biological characterization of the developed scaffolds was performed. GGE scaffolds showed a good porosity, which could promote cell infiltration and proliferation and a dense external surface, which could avoid bleeding. Moreover, developed scaffolds showed good hydrophilicity, an elastic behavior similar to natural vessels, suitability for sterilization by an ISO accepted treatment, and an adequate suture retention strength. In vitro cell culture tests showed no cytotoxic activity against 3T3 fibroblasts. The functionalization with the REDV peptide favored the adhesion and growth of endothelial cells, while GRGDSP-modified scaffolds represented a better substrate for fibroblasts
Spatiotemporal Overlap between the European Brown Hare and Its Potential Predators and Competitors
Simple SummaryPredator-prey relationships and competition shape interspecific coexistence in wildlife communities. So far, most published studies have focused on large carnivores and their prey, whereas little is known about medium and small-sized mammal communities. The European brown hare Lepus europaeus is a widespread species in Europe and is part of the diet of many birds of prey and mammalian carnivores of all sizes. Furthermore, competition with other herbivorous mammals at feeding sites has also been suggested. In an area in Central Italy, we have assessed spatiotemporal overlap among brown hare and its potential predators (red fox Vulpes vulpes, pine marten Martes martes, domestic cat Felis catus, and domestic dog Canis familiaris) and a competitor (roe deer Capreolus capreolus). We showed that, outside a fenced area excluding predators and competitors, brown hares become more nocturnal and more active on dark nights to limit encounters with predators, and that they adopt spatial partitioning to avoid competitors, as expected by ecological theory.Analysis of spatiotemporal partitioning is pivotal to shed light on interspecific coexistence. Most research effort has involved large-sized carnivores and their prey, whereas little attention has been devoted to lagomorphs. We assessed spatiotemporal overlap among the European brown hare Lepus europaeus and its potential competitors and predators through camera-trapping in an area in Central Italy. We estimated the interspecific patterns of the spatiotemporal activity rhythms of brown hares, its potential predators (the red fox Vulpes vulpes, the pine marten Martes martes, the domestic cat Felis catus, and the domestic dog Canis familiaris), and a competitor, the roe deer Capreolus capreolus. Brown hare activity was studied in natural conditions as well as in a fenced area that excluded terrestrial predators and competitors. Free-ranging hares developed a more nocturnal behavior to avoid diurnal predators (i.e., domestic carnivores and martens). Although high temporal overlap was observed between free-ranging brown hares and both red foxes (82%) and roe deer (81%), hares avoided fox by being more active on darkest nights, as well as avoided roe deer through spatial partitioning. We suggest that hares may adapt their spatiotemporal behavior to avoid potential predators and competitors
There is a correlation between nutritional status, Self-Rated Health and Life Satisfaction? Evidence from 2018 Health Behaviour in School-aged Children cross sectional study in a sample of Italian Adolescents living in Tuscany Region
Background: Overweight has been associated with several social and phycological problems and is perceived as one of the major health care challenges to focus on in the future. The purpose of the study is to investigate the correlations among nutritional status, assessed by the Body Mass Index, the perception of one's own health status and Life Satisfaction, detected in Italian adolescents living in Tuscany Region, and to investigate the influence of gender on them; Methods: A statistically representative sample of 2760 Tuscan adolescents aged 11, 13 and 15 was involved in the 2018 Health Behaviours at School-aged Children survey. The participants were divided into three nutritional status class: underweight, normal weight and overweight (overweight + obese); Results: The results show that there is a statistically significant difference in all categories between boys and girls aged 13 and 15 years; in girls aged 11 and 13 years, the Life Satisfaction of the overweight group is statistically lower than that of normal and underweight groups; Self-Rated Health is statistically lower in all age groups for overweight individuals compared to normal weight children, except for 11-year-old females; Conclusions: Viewing the psychosocial problems related to overweight, more attention and care must be placed on adolescents to ensure their healthier developmen
Genetic or pharmacological blockade of noradrenaline synthesis enhances the neurochemical, behavioral, and neurotoxic effects of methamphetamine
N-(2-chloroethyl)-N-ethyl-2-bromobenzylamine (DSP-4) lesions of the locus coeruleus, the major brain noradrenergic nucleus, exacerbate the damage to nigrostriatal dopamine (DA) terminals caused by the psychostimulant methamphetamine (METH). However, because noradrenergic terminals contain other neuromodulators and the noradrenaline (NA) transporter, which may act as a neuroprotective buffer, it was unclear whether this enhancement of METH neurotoxicity was caused by the loss of noradrenergic innervation or the loss of NA itself. We addressed the specific role of NA by comparing the effects of METH in mice with noradrenergic lesions (DSP-4) and those with intact noradrenergic terminals but specifically lacking NA (genetic or acute pharmacological blockade of the NA biosynthetic enzyme dopamine beta-hydroxylase; DBH). We found that genetic deletion of DBH (DBH-/- mice) and acute treatment of wild-type mice with a DBH inhibitor (fusaric acid) recapitulated the effects of DSP-4 lesions on METH responses. All three methods of NA depletion enhanced striatal DA release, extracellular oxidative stress (as measured by in vivo microdialysis of DA and 2,3-dihydroxybenzoic acid), and behavioral stereotypies following repeated METH administration. These effects accompanied a worsening of the striatal DA neuron terminal damage and ultrastructural changes to medium spiny neurons. We conclude that NA itself is neuroprotective and plays a fundamental role in the sensitivity of striatal DA terminals to the neurochemical, behavioral, and neurotoxic effects of METH
IGF-1 loaded injectable microspheres for potential repair of the infarcted myocardium
The use of injectable scaffolds to repair the infarcted heart is receiving great interest. Thermosensitive polymers, in situ
polymerization, in situ cross-linking, and self-assembling peptides are the most investigated approaches to obtain
injectability.
Aim of the present work was the preparation and characterization of a novel bioactive scaffold, in form of injectable
microspheres, for cardiac repair. Gellan/gelatin microspheres were prepared by a water-in-oil emulsion and loaded by
adsorption with Insulin-like growth factor 1 to promote tissue regeneration. Obtained microspheres underwent morphological,
physicochemical and biological characterization, including cell culture tests in static and dynamic conditions
and in vivo tests. Morphological analysis of the microspheres showed a spherical shape, a microporous surface and an
average diameter of 66 ± 17mm (under dry conditions) and 123 ± 24 mm (under wet conditions). Chemical Imaging
analysis pointed out a homogeneous distribution of gellan, gelatin and Insulin-like growth factor-1 within the microsphere
matrix. In vitro cell culture tests showed that the microspheres promoted rat cardiac progenitor cells adhesion,
and cluster formation. After dynamic suspension culture within an impeller-free bioreactor, cells still adhered to microspheres,
spreading their cytoplasm over microsphere surface. Intramyocardial administration of microspheres in a
cryoinjury rat model attenuated chamber dilatation, myocardial damage and fibrosis and improved cell homing.
Overall, the findings of this study confirm that the produced microspheres display morphological, physicochemical,
functional and biological properties potentially adequate for future applications as injectable scaffold for cardiac tissue
engineering
On farm agronomic and first environmental evaluation of oil crops for sustainable bioenergy chains
Energy crops, and in particular oil crops, could be an important occasion for developing new non food production
rows for a new multi-functional agriculture in Italy. In this view, the use of local biomass is a fundamental starting
point for the development of a virtuous energy chain that should pursue not only agricultural profitability, but also
chain sustainability and that is less dependent on the global market, characterized by instability in terms of biomass
availability and price. From this perspective, particular attention must be paid to crop choice on the basis of
its rusticity and of its adaptability to local growing conditions and to low input cropping systems. In this context,
alike woody and herbaceous biomasses, oil crops such as sunflower and rapeseed should be able to support local
agricultural bioenergy chain in Italy.
In addition, in a local bioenergy chain, the role of the farmers should not be limited just to grain production; but
also grain processing should be performed at farm or consortium level in oilseed extraction plants well proportioned
to the cropped surface. In this way, by means of a simple power generator, farmer could thus produce its
own thermal and electric energy from the oil, maximizing his profit. This objective could also be achieved through
the exploitation of the total biomass, including crop residues and defatted seed meals, that may be considered as
fundamental additional economic and/or environmental benefits of the chain. This paper reports some results of
three-years on-farm experiments on oil crop chain carried out in the framework of "Bioenergie" project, that was
focused to enhance farmers awareness of these criteria and to the feasibility at open field scale of low-input cultivation
of rapeseed, sunflower and Brassica carinata in seven Italian regions. In several on-farm experiences, these
crops produced more than 800 kg ha-1 of oil with good energy properties. Defatted seed meals could be interesting
as organic fertilizers and, in the case of B. carinata, as a biofumigant amendment that could offer a total or partial
alternative to some chemicals in agriculture. Furthermore, biomass soil incorporation could contribute to C sequestration,
catching CO2 from atmosphere and sinking a part in soil as stable humus. Finally, four different open
field experiences carried out again in the second year of the project, have been analysed in order to evaluate their
energy and greenhouse gasses balance after cultivation phase

Utilization of mechanical power and associations with clinical outcomes in brain injured patients: a secondary analysis of the extubation strategies in neuro-intensive care unit patients and associations with outcome (ENIO) trial
Background: There is insufficient evidence to guide ventilatory targets in acute brain injury (ABI). Recent studies have shown associations between mechanical power (MP) and mortality in critical care populations. We aimed to describe MP in ventilated patients with ABI, and evaluate associations between MP and clinical outcomes. Methods: In this preplanned, secondary analysis of a prospective, multi-center, observational cohort study (ENIO, NCT03400904), we included adult patients with ABI (Glasgow Coma Scale ≤ 12 before intubation) who required mechanical ventilation (MV) ≥ 24 h. Using multivariable log binomial regressions, we separately assessed associations between MP on hospital day (HD)1, HD3, HD7 and clinical outcomes: hospital mortality, need for reintubation, tracheostomy placement, and development of acute respiratory distress syndrome (ARDS). Results: We included 1217 patients (mean age 51.2 years [SD 18.1], 66% male, mean body mass index [BMI] 26.3 [SD 5.18]) hospitalized at 62 intensive care units in 18 countries. Hospital mortality was 11% (n = 139), 44% (n = 536) were extubated by HD7 of which 20% (107/536) required reintubation, 28% (n = 340) underwent tracheostomy placement, and 9% (n = 114) developed ARDS. The median MP on HD1, HD3, and HD7 was 11.9 J/min [IQR 9.2-15.1], 13 J/min [IQR 10-17], and 14 J/min [IQR 11-20], respectively. MP was overall higher in patients with ARDS, especially those with higher ARDS severity. After controlling for same-day pressure of arterial oxygen/fraction of inspired oxygen (P/F ratio), BMI, and neurological severity, MP at HD1, HD3, and HD7 was independently associated with hospital mortality, reintubation and tracheostomy placement. The adjusted relative risk (aRR) was greater at higher MP, and strongest for: mortality on HD1 (compared to the HD1 median MP 11.9 J/min, aRR at 17 J/min was 1.22, 95% CI 1.14-1.30) and HD3 (1.38, 95% CI 1.23-1.53), reintubation on HD1 (1.64; 95% CI 1.57-1.72), and tracheostomy on HD7 (1.53; 95%CI 1.18-1.99). MP was associated with the development of moderate-severe ARDS on HD1 (2.07; 95% CI 1.56-2.78) and HD3 (1.76; 95% CI 1.41-2.22). Conclusions: Exposure to high MP during the first week of MV is associated with poor clinical outcomes in ABI, independent of P/F ratio and neurological severity. Potential benefits of optimizing ventilator settings to limit MP warrant further investigation
