Tolerance degree of some maize (Zea mays) hybrids to rimsulfuron1
The degree of tolerance to the sulfonylurea herbicide rimsulfuron (R) was investigated
in five different commercial maize hybrids: Sirena (Si), Sara (Sa), Eleonora (El),
Gitana (Gi) and Lucia (Lu). The susceptibility of the hybrids to R treatment was
investigated by checking their protein content and evaluating R persistence in the
maize hybrids as well as the activity and kinetic parameters of the target enzyme
they contain, acetolactate syntase (ALS). 72 hours after treatment, decreases in
the protein content were considerable in Gi and Lu, moderate in El, and not significant
in Si and Sa. At the same time El, Lu and Gi exhibited significant R residues, while
the herbicide was completely metabolized in Si and Sa. The values of extractable ALS
activity in the untreated hybrids ("native" ALS) - with and without R addition in the
reaction mixture - and in the R-treated hybrids, together with the ALS V and K
parameters of the untreated and R-treated hybrids, evidenced varying degrees of
tolerance to R among the hybrids. Despite the susceptibility of "native" ALS in all
of the hybrids to R, Si and Sa appeared to overcome the action of R through a rapid
detoxifying capability, and El through an ability to produce more ALS enzymes in
response to R treatment. Gi and Lu exhibited a lower tolerance to R since they were
unable to activate any of the above mechanisms.Degré de tolérance au rimsulfuron de certains hybrides de maïs. Le degré de tolérance à
l'herbicide sulfonyl-urée rimsulfuron (R) a été étudié chez cinq hybrides différents
de maïs Sirena (Si), Sara (Sa), Eleonora (El), Gitana (Gi) et Lucia (Lu). Pour étudier
la sensibilité des hybrides au traitement avec R, on a contrôlé leur teneur en protéines
et évalué les paramètres cinétiques et d'activité de l'enzyme-cible qu'ils contiennent,
l'acétolactate synthétase (ALS). Soixante douze heures après le traitement, on a relevé
une diminution considérable de la teneur en protéines chez Gi et Lu, modérée chez El et
non significative chez Si et Sa. Dans le même temps, El, Lu et Gi ont montré des résidus
significatifs de R, tandis que l'herbicide avait été complètement métabolisé chez Si
et Sa. Les valeurs de l'activité d'ALS que l'on a pu relever dans les hybrides non
traités (ALS "à l'état natif " ) - avec ou sans adjonction de R dans le mélange de
réaction - et dans les hybrides traités avec R, ainsi que les valeurs des paramètres
V et K d'ALS des hybrides non traités et traités avec R ont permis de relever
des degrés variables de tolérance à R parmi les hybrides. Malgré la sensibilité
envers R de l'ALS "à l'état natif " de tous les hybrides, on a pu voir que Si et
Sa surmontaient l'action de R par le biais d'une capacité de métabolisation rapide,
et El grâce à sa capacité de produire davantage d'enzymes ALS en réponse au traitement
avec R. Par contre, Gi et Lu, incapables d'activer les mécanismes ci-dessus, ont donc
fait apparaître une tolérance moindre à R
Factors of tolerance to rimsulfuron in four pepper (Capsicum annum L.) lines
The degree of tolerance to the sulfonylurea herbicide rimsulfuron (R) was investigated in
four F pepper lines: Argo (Ar), Heldor (He), Ideal (Id) and Indalo (In), by evaluating
the activity and kinetic parameters of the target site they contain, acetolactate synthase
(ALS), as well as herbicide persistence. After 48 hours from treatment the herbicide was
completely metabolised in He, Id and In, while Ar had a 0.4 ppm residue. As shown by I
values, ALS sensitivity to R proved to be different in the four lines according to the
following increasing order: In, He, Id and Ar. Enzyme kinetic studies revealed a distinct
expression of natural ALS endowment in all lines. In particular, V in the untreated
samples was greatest in He followed by, in decreasing order, Id, Ar and In. Compared to
untreated samples V values were greater in treated In and lower in the other treated
as follows He Id Ar. K values were lower in In and He than in Ar and Id, and were
unaffected by R treatment. The present study indicates that In and He are more tolerant
to R than Ar and Id, given their lower ALS sensitivity to R together with their higher
enzyme activity, which is naturally occurring in He and R-induced in In. Moreover,
a greater R persistence resulted in Ar than in the other lines. These findings were
confirmed by a field trial which showed growth damage and a reduction of productivity
in the Id and Ar lines.Facteurs de tolérance au rimsulfuron dans quatre lignées de poivron (Capsicum annum L.).
On a étudié le degré de tolérance à l'herbicide sulfonyluré rimsulfuron (R) dans quatre
lignées de poivrons F : Argo (Ar), Heldor (He), Ideal (Id) et Indalo (In), en évaluant
l'activité et les paramètres cinétiques du site-cible qu'ils contiennent - acétolactate
synthase (ALS) - ainsi que la persistance de l'herbicide. Quarante huit heures après le
traitement, l'herbicide avait subi une métabolisation complète dans He, Id et In, tandis
que Ar présentait un résidu de 0,4 ppm. Comme l'ont montré les valeurs I, les quatre
lignées ont témoigné d'une sensibilité différente de ALS à R, dans l'ordre croissant
suivant : In, He, Id et Ar. Les études sur la cinétique des enzymes ont révélé que la
dotation naturelle en ALS s'exprime différemment dans toutes les lignées. Notamment,
dans les échantillons non traités, la V la plus élevée était celle de He, suivie,
par ordre décroissant, de Id, Ar et In. Par rapport aux échantillons non traités, on
a relevé des valeurs de V plus élevées dans In traité et plus basses dans les autres
lignées traitées avec décréments dans l'ordre : He Id Ar. Les valeurs K étaient
plus basses dans In et He que dans Ar et Id et n'ont pas été touchées par le traitement
au R. Ces données indiquent que In et He ont une tolérance plus élevée à R que Ar et Id,
grâce à une sensibilité moindre d'ALS à l'égard de R ainsi qu'à leur activité enzymatique
plus élevée, naturelle dans He et provoquée par R dans In. En outre, dans Ar il y a une
plus grande persistance de R que dans les autres lignées. Ces résultats ont été confirmés
par un essai sur le terrain qui a mis en évidence un dégât durant la croissance et une
réduction de productivité dans les lignées Id et Ar
Festuca arundinacea grass and herbicide safeners to prevent herbicide pollution
Buffer strips are uncultivated zones left along the boundaries of crops.
Buffer strips are used to eliminate or reduce the environmental impact of
herbicides. As the efficiency of buffer strips is improved by the presence
of non-crop vegetation, we studied the possible role of growing the
perennial grass Festuca arundinacea. We investigated the activity in festuca
of glutathione S-transferase (GST), which is an enzyme very active in metabolising
herbicides. Our results evidence GST activity, which is enhanced by the
following compounds: benoxacor, cloquintocet-mexyl, fenchlorazol-ethyl,
fenclorim, fluxofenim and oxabetrinil. These synthetic compounds are named
herbicide safeners because they protect crop plants against injury from some
herbicides without reducing the action of herbicides against the target
weeds. The increases in GST activity were found to be concomitant with
changes in V and K, that are kinetic constants related
directly to the enzyme concentration in the protein “pull” and inversely
to the substrate–enzyme affinity, respectively. In particular, V increase with
K decrease was observed in response to benoxacor, V increases
were found in response to fenchlorazol-ethyl, fenclorim, fluxofenim and
oxabetrinil, and K decrease was observed in response to
cloquintocet-mexyl. The GST activity was also found to be enhanced by the
safeners when it was tested toward the herbicides terbuthylazine and
butachlor as substrates. In particular, the increases in GST toward
terbuthylazine ranged in the following decreasing order: 154.6%,
91.7%, 89.2%, 88.3%, 82.5% and 30.8% in response to
fluxofenim, fenchlorazol-ethyl, fenclorim, oxabetrinil, benoxacor and
cloquintocet-mexyl, respectively. The increases in GST toward butachlor
ranged in the following decreasing order: 77.0%, 71.2% 59.0%,
41.0% and 33.1% in response to oxabetrinil, benoxacor, fenclorim,
fluxofenim and fenchlorazol-ethyl, respectively. A further test, performed
to evaluate the relevance of the above effects on a macro-scale level,
evidenced 10.1% and 32.7% increased amounts of metabolised
terbuthylazine and butachlor, respectively, in response to the addition of
benoxacor safener to the herbicide treatments. These results indicate that
herbicide diffusion following the runoff of surface waters can be prevented
or significantly reduced by vegetating buffer strips with festuca and by the
combination of herbicide and a suitable safener
Ability of three pesticide leaching models to simulate summer crops in a Mediterranean scenario
The capability to predict the fate of pesticides in a southern Italian
scenario was evaluated for three Pesticide Leaching Models (PLMs)
(VARLEACH, PELMO and PESTLA), using a data set previously achieved
for this purpose. The measurement of actual evapotranspiration in
field experiments allowed a calibration of the three PLMs for the
hydrological component in the scenario studied, where evapotranspiration
was very high in the hotter months. While PELMO and PESTLA were easily
calibrated, VARLEACH did not give satisfactory results, maybe due to its
intrinsic deficiency. After calibration, the three PLMs simulated well
the movement of the two herbicides imazethapyr and bentazone along the
soil profile. Agreement could be casual for VARLEACH, since it did not
simulate well the movement of water and bromide (non-interfering ion).
PESTLA simulated well the total herbicide amount leached below 80 cm depth,
while VARLEACH and PELMO failed at this. The three calibrated PLMs could be
used to perform simulations in the same climatic conditions as the experiment.Capacité de trois modèles de lessivage des pesticides à simuler
une culture d'été dans un scénario méditerranéen. La capacité de
prévoir le devenir de pesticides dans un scénario du sud de l'Italie
a été testée pour trois modèles de lessivage des pesticides (PLM),
VARLEACH, PELMO et PESTLA, en utilisant un jeu de données préalablement
obtenu dans une expérience au champ. Les mesures de l'évapotranspiration
réelle dans une expérience au champ a nécessité le calage des trois
modèles (PLM) pour la composante hydrologique dans le scénario étudié,
dans lequel l'évapotranspiration était très élevée durant les mois chauds.
Alors que PELMO et PESTLA ont été calés facilement, VARLEACH n'a pas donné
de résultats satisfaisants, peut-être à cause de ses déficiences intrinsèques.
Après calage, les trois modèles PLM ont bien simulé les mouvements de deux
herbicides imazethapyr et bentazone selon le profil du sol. L'accord pourrait
être fortuit pour VARLEACH, puisqu'il ne simule pas correctement le mouvement
d'eau et d'ion bromure (ion non interférent). PESTLA simule bien la quantité
totale d'herbicide lessivée au-dessous de 80 cm de profondeur, domaine dans
lequel VARLEACH et PELMO ont échoué. Les trois modèles PLM calés pourraient
être utilisés pour effectuer des simulations dans des conditions climatiques
semblables à celles de l'expérience
