Collisional losses, decoherence, and frequency shifts in optical lattice clocks with bosons
We have quantified collisional losses, decoherence and the collision shift in
a one-dimensional optical lattice clock with bosonic 88Sr. The lattice clock is
referenced to the highly forbidden transition 1S0 - 3P0 at 698 nm, which
becomes weakly allowed due to state mixing in a homogeneous magnetic field. We
were able to quantify three decoherence coefficients, which are due to
dephasing collisions, inelastic collisions between atoms in the upper and lower
clock state, and atoms in the upper clock state only. Based on the measured
coefficients, we determine the operation parameters at which a 1D-lattice clock
with 88Sr shows no degradation due to collisions on the relative accuracy level

Cytochrom c-basierter Superoxidsensor in organischen Medien
Das Superoxid-Radikal O2- ist wegen seiner toxischen Wirkung in der Medizin von grossem Interesse. Es hat sich ausserdem als geeignetes Modellsystem zur Untersuchung von Radikal-Scavengern bewÀhrt.
Der untersuchte Sensor besteht aus an einer Goldelektrode kovalent immobilisiertem Cytochrom c und kann amperometrisch zur Bestimmung der Konzentration von Superoxid-Radikalen verwendet werden. Dies ermöglicht, die antioxidative Wirkung von Substanzen nachzuweisen, welche Superoxid-Ionen abfangen und somit die Superoxid-Konzentration im Medium verringern.
Die Methode ist in rein wÀssrigen Systemen etabliert.
Dies limitiert allerdings die Anwendung auf hydrophile Substanzen. Der Sensor kann somit nicht fĂŒr die Charakterisierung der Vielzahl von antioxidativ wirksamen hydrophoben Naturprodukten, wie etwa Vitaminen, und der Bestimmung der EffektivitĂ€t von kosmetischen Produkten auf Creme-Basis mit verschiedenen antioxidativen Beigaben angewendet werden.
Insbesondere die Bestimmung der Gesamt-EffektivitÀt von kosmetischen Produkten mit verschiedenen antioxidativen ZusÀtzen ist hier von grossem Interesse.
Um diese BeschrÀnkung aufzuheben und somit die Anwendungsbreite des Sensors erheblich zu erweitern, werden in der vorliegenden Arbeit die Elektrochemie von kovalent immobilisiertem Cytochrom c und das Detektionsverhalten des Sensors in nicht rein wÀssrigen Lösungen, d.h. in verschiedenen organischen Lösungsmitteln sowie Gemischen dieser Lösungsmittel mit wÀssrigem Puffer, untersucht.
Im etablierten Sensor wird das Cytochrom c mittels des Linkers MercaptoundecansÀure kovalent an einer Goldelektrode immobilisiert. Dessen Schwefel-Gruppe bindet an die OberflÀche der Goldelektrode; die SÀurefunktion wird an Lysin-Reste des Cytochrom c kovalent gebunden
Wavelength dependent ac-Stark shift of the 1S0 - 3P1 transition at 657 nm in Ca
We have measured the ac-Stark shift of the 4s2 1S0 - 4s4p 3P1 line in 40Ca
for perturbing laser wavelengths between 780 nm and 1064 nm with a time domain
Ramsey-Borde atom interferometer. We found a zero crossing of the shift for the
mS = 0 - mP = 0 transition and \sigma polarized perturbation at 800.8(22) nm.
The data was analyzed by a model deriving the energy shift from known
transition wavelengths and strengths. To fit our data, we adjusted the Einstein
A coefficients of the 4s3d 3D - 4s4p 3P and 4s5s 3S - 4s4p 3P fine structure
multiplets. With these we can predict vanishing ac-Stark shifts for the 1S0 m =
0 - 3P1 m = 1 transition and \sigma- light at 983(12) nm and at 735.5(20) nm
for the transition to the 3P0 level.Comment: 8 pages, 5 figures, 2 table
Hyper-Ramsey Spectroscopy of Optical Clock Transitions
We present non-standard optical Ramsey schemes that use pulses individually
tailored in duration, phase, and frequency to cancel spurious frequency shifts
related to the excitation itself. In particular, the field shifts and their
uncertainties of Ramsey fringes can be radically suppressed (by 2-4 orders of
magnitude) in comparison with the usual Ramsey method (using two equal pulses)
as well as with single-pulse Rabi spectroscopy. Atom interferometers and
optical clocks based on two-photon transitions, heavily forbidden transitions,
or magnetically induced spectroscopy could significantly benefit from this
method. In the latter case these frequency shifts can be suppressed
considerably below a fractional level of 10^{-17}. Moreover, our approach opens
the door for the high-precision optical clocks based on direct frequency comb

Interrogation laser for a strontium lattice clock
We report on the setup and characterization of a 698 nm master-slave diode
laser system to probe the 1S0-3P0 clock transition of strontium atoms confined
in a one-dimensional optical lattice. A linewidth in the order of around 100 Hz
of the laser system has been measured with respect to an ultrastable 657 nm
diode laser with 1 Hz linewidth using a femtosecond fiber comb as transfer
oscillator. The laser has been used to measure the magnetically induced 1S0-3P0
clock transition in 88Sr where a linewidth of 165 Hz has been observed. The
transfer oscillator method provides a virtual beat signal between the two diode
lasers that has been used to phase lock the 698 nm laser to the 1 Hz linewidth
laser at 657 nm, transferring its stability to the 698 nm laser system.Comment: 5 pages, 7 figures, to be published in "IEEE Transactions on

Absolute frequency measurement of the magnesium intercombination transition
We report on a frequency measurement of the clock
transition of Mg on a thermal atomic beam. The intercombination
transition has been referenced to a portable primary Cs frequency standard with
the help of a femtosecond fiber laser frequency comb. The achieved uncertainty
is which corresponds to an increase in accuracy of six
orders of magnitude compared to previous results. The measured frequency value
permits the calculation of several other optical transitions from to
the -level system for Mg, Mg and Mg. We describe in
detail the components of our optical frequency standard like the stabilized
spectroscopy laser, the atomic beam apparatus used for Ramsey-Bord\'e
interferometry and the frequency comb generator and discuss the uncertainty
contributions to our measurement including the first and second order Doppler
effect. An upper limit of in one second for the short term
instability of our optical frequency standard was determined by comparison with
a GPS disciplined quartz oscillator.Comment: 8 pages, 8 figure
Long range transport of ultra cold atoms in a far-detuned 1D optical lattice
We present a novel method to transport ultra cold atoms in a focused optical
lattice over macroscopic distances of many Rayleigh ranges. With this method
ultra cold atoms were transported over 5 cm in 250 ms without significant atom
loss or heating. By translating the interference pattern together with the beam
geometry the trap parameters are maintained over the full transport range.
Thus, the presented method is well suited for tightly focused optical lattices
that have sufficient trap depth only close to the focus. Tight focusing is
usually required for far-detuned optical traps or traps that require high laser
intensity for other reasons. The transport time is short and thus compatible
with the operation of an optical lattice clock in which atoms are probed in a
well designed environment spatially separated from the preparation and

Nutzung von NukleinsĂ€ure-Protein-Wechselwirkungen fĂŒr die Wirkanalytik von reaktiven Species
Reaktive Species wie Superoxid, Stickstoffmonoxid oder Peroxynitrit sind in eine Vielzahl pathophysiologischer Situationen wie z.B. das Reperfusionssyndrom involviert. Besonders hohe Konzentrationen treten auf, wenn das antioxidative Abwehrsystem nicht mehr in der Lage ist, den Radikalanstieg abzufangen. Die biomedizinische Forschung hat sich auf die Mechanismen der Freisetzung dieser Species unter unterschiedlichen StreĂbedingungen fokussiert. Aus diesem Grund sind verschiedene analytische MeĂmethoden fĂŒr den Nachweis entwickelt worden. Sensorische Methoden bieten den Vorteil einer rĂ€umlich und zeitlich aufgelösten Analyse der Einzelteilchen. Jedoch gibt es betrĂ€chtliche Wechselwirkungen zwischen den verschiedenen reaktiven Species, was die AussagefĂ€higkeit von Einzelmessungen in komplexen Situationen einschrĂ€nkt. Hier erscheint ein Summenparameter fĂŒr die Radikalwirkung im Sinne einer Wirkanalytik aussagekrĂ€ftiger.
Das âiron regulatory protein 1â (IRP1) kann als ein solches Markerprotein betrachtet werden, dessen Konzentration AufschluĂ ĂŒber den zellulĂ€ren StreĂlevel gibt. Das Protein wird unter der Wirkung von oxidativem StreĂ aus dem 4Fe-4S-Enzym cytosolische Aconitase gebildet. Das entstandene IRP1 zeigt im Gegensatz zur Aconitase eine ausgeprĂ€gte BindungsaffinitĂ€t zu spezifischen m-RNA-Strukturen - den sogenannten âiron responsive elementsâ - IREs. Dieses Verhalten kann als Grundlage fĂŒr die sensorische Detektion des Markerproteins genutzt werden.
In dem hier vorzustellenden experimentellen Ansatz wurde die Proteinbindung mit Hilfe der OberflĂ€chenplasmonresonanz detektiert. Hierzu wurde eine IRE-Konsensus-Sequenz in vitro transkribiert und anschlieĂend auf einem Carboxydextranâmodifizierten Biacore-Chip immobilisiert. Eine relativ hohe OberflĂ€chenbelegung (2000 RU /mm2) wurde sichergestellt
