    Numerical analysis of yield properties of closed-cell aluminum foam under multiaxial loads by 3D voronoi model

    Metallic foam is a typical porous material whose yield surface is related to not only von Mises equivalent stress but also the hydrostatic pressure. It is essential to study the yield properties of closed-cell aluminum foam under complex loading conditions. However, because the current experimental technique may support only a few proportions of multiaxial loading, it is hard to learn the yield surface well especially for the tensile hydrostatic pressure. In this article, we explored a numerical method to learn the yield properties of aluminum foam, in which the meso structures of aluminum foam were simulated by 3D Voronoi method. In addition, the yield surface of aluminum foam was drawn successfully with the numerical method. The main works included: (1) In our numerical simulation, we tested the calculating parameters such as mass scaling, elements number, and loading velocity on simulation results and verified the homogeneity of the 3D Voronoi model firstly. Furthermore, the optimized calculating parameters were determined by considering both reliability and feasibility of the calculation. Homogeneity of 3D Voronoi model was checked by the compression behaviors of aluminum in different directions. (2) In order to overcome the difficulty to determine critical yield state of metallic foams under complex loads, we recommended criterion by setting a dimensionless plastic dissipation value to determine the onset yield state of the foam under multiaxial loads. The critical value of dimensionless plastic dissipation was deduced from the common criterion—0.2% plastic strain in uniaxial loading, and the effect of relative densities on critical values was analyzed. (3) Three normal stresses were applied on cubic aluminum foam proportionally to implement the proportional loading. The different loading proportional factors of the three normal stresses were set widely to insure the yield surface including enough data, such as the hydrostatic loads cover from minimum negative to maximum positive values; each proportion has three loading proportional factors.     Sphingosine kinase 1–mediated inhibition of Fas death signaling in rheumatoid arthritis B lymphoblastoid cells

    Objective It is becoming increasingly apparent that B cells play an important role in the pathogenesis of rheumatoid arthritis (RA). Due to the scarcity of B cells in RA, it has been technically difficult to functionally characterize B cell apoptosis in this disease. As a necessary first step to identify candidate aberrations, we investigated Fas-mediated signaling events in immortalized peripheral blood B lymphoblastoid cell lines (LCLs) from patients with RA and controls. Methods Cell death was determined by the MTS assay, and apoptosis was detected by the TUNEL assay and DNA laddering. Proteolytic activation of caspase 3 was determined by immunoblotting, and its enzymatic activity was determined by a fluorometric technique. Messenger RNA (mRNA) expression was quantified by real-time polymerase chain reaction (PCR) analysis. The functional role of sphingosine kinase (SPHK) was determined by measuring its enzymatic activity, by quantifying the levels of its product, sphingosine 1-phosphate (S1P), and by investigating the ability of the SPHK inhibitor N , N -dimethylsphingosine and isozyme-specific small interfering RNA (siRNA) oligonucleotides to reverse signaling aberrations. Results LCLs from patients with RA displayed disease-specific Fas-mediated signal transduction impairment with consequent resistance to cell death. RA LCLs displayed high constitutive SPHK activity and increased levels of S1P. Real-time PCR analysis showed higher SPHK-1 mRNA expression levels in RA patients compared with paired controls. Increased SPHK-1 (but not SPHK-2) mRNA levels were observed in synovial tissue from RA patients. Competitive inhibitors of SPHK reversed the resistance of RA LCLs to Fas-induced apoptosis. Additionally, resistance to Fas-mediated signaling was reversed by siRNA oligonucleotides specific for SPHK-1 but not by oligonucleotides specific for SPHK-2. Conclusion These findings demonstrate disease-specific resistance to Fas-mediated death signaling in patients with RA and implicate increased SPHK-1 activity as the cause of this aberration.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/49513/1/21635_ftp.pd

    OWL: A Large Language Model for IT Operations

    With the rapid development of IT operations, it has become increasingly crucial to efficiently manage and analyze large volumes of data for practical applications. The techniques of Natural Language Processing (NLP) have shown remarkable capabilities for various tasks, including named entity recognition, machine translation and dialogue systems. Recently, Large Language Models (LLMs) have achieved significant improvements across various NLP downstream tasks. However, there is a lack of specialized LLMs for IT operations. In this paper, we introduce the OWL, a large language model trained on our collected OWL-Instruct dataset with a wide range of IT-related information, where the mixture-of-adapter strategy is proposed to improve the parameter-efficient tuning across different domains or tasks. Furthermore, we evaluate the performance of our OWL on the OWL-Bench established by us and open IT-related benchmarks. OWL demonstrates superior performance results on IT tasks, which outperforms existing models by significant margins. Moreover, we hope that the findings of our work will provide more insights to revolutionize the techniques of IT operations with specialized LLMs.Comment: 31 page

    A two-sample Mendelian randomization analysis: causal association between chemokines and pan-carcinoma

    Objective: According to the 2020 data from the World Health Organization (WHO), cancers stand as one of the foremost contributors to global mortality. Revealing novel cancer risk factors and protective factors is of paramount importance in the prevention of disease occurrence. Studies on the relationship between chemokines and cancer are ongoing; however, due to the coordination of multiple potential mechanisms, the specific causal association remains unclear.Methods: We performed a bidirectional Mendelian randomization analysis to explore the causal association between serum chemokines and pan-carcinoma. All data is from the GWAS catalog and IEU Open GWAS database. The inverse-variance weighted (IVW) method is primarily employed for assessing the statistical significance of the findings. In addition, the significance threshold after the multiple hypothesis test (Bonferroni) was 0.0013, and the evidence of a potential association was considered if the p-value < 0.05, but remained greater than Bonferroni’s threshold.Results: The results indicate that CCL1 (odds ratio, OR = 1.18), CCL2 (OR = 1.04), CCL8 (OR = 1.36), CCL14 (Colorectal, OR = 1.08, Small intestine, OR = 0.77, Lung, OR = 1.11), CCL15 (OR = 0.85), CCL18 (Breast, OR = 0.95, Prostate, OR = 0.96), CCL19 (Lung, OR = 0.66, Prostate, OR = 0.92), CCL20 (Lung, OR = 0.53, Thyroid, OR = 0.76), CCL21 (OR = 0.62), CCL22 (OR = 2.05), CCL23 (OR = 1.31), CCL24 (OR = 1.06), CCL27 (OR = 1.49), CCL28 (OR = 0.74), CXCL5 (OR = 0.95), CXCL9 (OR = 3.60), CXCL12 (Breast, OR = 0.87, Small intestine, OR = 0.58), CXCL13 (Breast, OR = 0.93, Lung, OR = 1.29), CXCL14 (Colon, OR = 1.40) and CXCL17 (OR = 1.07) are potential risk factors for cancers. In addition, there was a reverse causal association between CCL1 (OR = 0.94) and CCL18 (OR = 0.94) and breast cancer. Sensitivity analysis results were similar. The results of the other four MR Methods were consistent with the main results, and the leave-one-out method showed that the results were not driven by a Single nucleotide polymorphism (SNP). Moreover, there was no heterogeneity and pleiotropy in our analysis.Conclusion: Based on the two-sample MR Analysis method, we found that chemokines might be upstream factors of cancer pathogenesis. These results might provide new insights into the future use of chemokines as potential targets for cancer prevention and treatment. Our results also provide important clues for tumor prevention, and changes of serum chemokine concentration may be recognized as one of the features of precancerous lesions in future clinical trials

    The prevalence of food allergy in cesarean-born children aged 0–3 years: A systematic review and meta-analysis of cohort studies

    PurposePrevious studies reported a higher risk of food allergy for cesarean-born children than vaginal-born children. This study aims to systematically compare the prevalence of food allergy among cesarean-born and vaginal-born children aged 0–3 years.MethodsThree English and two Chinese databases were searched using terms related to food allergies and cesarean sections. Cohort studies that reported the prevalence of food allergy in cesarean-born and vaginal-born children aged 0–3 years were included. Two reviewers performed study selection, quality assessment, and data extraction. The pooled prevalence of food allergy in cesarean-born and vaginal-born children was compared by meta-analysis.ResultsNine eligible studies, with 9,650 cesarean-born children and 20,418 vaginal-born children aged 0–3 years, were included. Of them, 645 cesarean-born children and 991 vaginal-born children were identified as having food allergies. The pooled prevalence of food allergy was higher in cesarean-born children (7.8%) than in vaginal-born children (5.9%). Cesarean section was associated with an increased risk of food allergy [odds ratio (OR): 1.45; 95% confidence interval (CI): 1.03–2.05] and cow's milk allergy (OR: 3.31; 95% CI: 1.98–5.53). Additionally, cesarean-born children with a parental history of allergy had an increased risk of food allergy (OR: 2.60; 95% CI: 1.28–5.27).ConclusionThis study suggests that cesarean sections was associated with an increased risk of food and cow's milk allergies in children aged 0–3 years. Cesarean-born children with a parental history of allergy demonstrated a higher risk for food allergy than did vaginal-born children. These results indicate that caregivers should be aware of the risks of food allergies in cesarean-born children, reducing the risk of potentially fatal allergic events.
