    Electrochemical detection of the toxic dinoflagellate Alexandrium ostenfeldii with a DNA-biosensor

    The steady rise of observations of harmful or toxic algal blooms throughout the world in the past decades constitute a menace for coastal ecosystems and human interests. As a consequence, a number of programs have been launched to monitor the occurrence of harmful and toxic algae. However, the identification is currently done by microscopic examination, which requires a broad taxonomic knowledge, expensive equipment and is very time consuming. In order to facilitate the identification of toxic algae, an inexpensive and easy-to-handle DNA-biosensor has been adapted for the electrochemical detection of the toxic dinoflagellate Alexandrium ostenfeldii. The detection of the toxic algae is based on a sandwich hybridisation, which is carried out on a disposable sensor chip. A set of two probes for the species specific identification of A. ostenfeldii was developed. The specificity of the probes could be shown in dot-blot hybridisation and with the DNA-biosensor. The sensitivity of the DNA-biosensor was optimised with respect to hybridisation temperature and NaCl-concentration and a significant increase of the sensitivity of the DNA-biosensor could be obtained by a fragmentation of the rRNA prior to the hybridisation and by adding a helper oligonucleotide, which binds in close proximity to the probes to the hybridisation

    Multi-locality in rural areas – an underestimated phenomenon

    Multi-locality – a lifestyle involving living in several places – is very diverse. Involving temporary presences and absences, it can impact local communities and the environment. This paper looks at the effects of multi-local lifestyles in three rural districts in Lower Saxony, Germany, evaluating quantitative surveys and qualitative interviews with multi-local residents, company representatives and officials. The methods are supplemented by visualizations and reflexive photography. Multi-locals can be seen as both an opportunity and a risk for civic involvement: multi-locality makes it possible to avoid relocation, but the limited periods of presence make it unlikely for multi-locals to become involved locally. Likewise, multi-locality impacts the environment, leading to an increasing volume of traffic and higher land consumption, even if it less significant than the impact of daily commuting

    Emotional Self-Disclosure, Keeping Secrets and its Effect on Perceived Physical and Mental Health

    Sinds lange tijd bestaat er consensus over het bestaan van verbanden tussen emotionele disclosure en zowel de fysieke- als mentale gezondheid. Dit verband wordt tevens verondersteld met betrekking tot het hebben van geheimen. In huidig onderzoek wordt getracht met inbreng van moderatoren meer zicht te krijgen in de determinanten die van invloed zijn op de gestelde relaties. Huidig onderzoek beoogt meer inzicht te genereren in de effecten van emotionele disclosure en het hebben van geheimen op de fysieke en mentale gezondheid. Daarbij is het van belang te weten wanneer deze effecten variëren, danwel aan- of afwezig zijn. Respondenten zijn geworven middels diverse academische discussiefora en sociale netwerksites. De totale responsgroep bestond uit 151 respondenten, waarvan 72% vrouw en 28% man. De leeftijd van de respondenten lag tussen de 18 en 62 jaar, de gemiddelde leeftijd was 36,6 jaar (SD = 11.8). Huidig onderzoek betreft een survey-onderzoek. Respondenten vulden online een vragenlijst in. Voor dit onderzoek werd een vragenlijst vervaardigd die bestaat uit delen van de Symptom Checklist -90 (SCL-90; Arrindel & Ettema, 1975; 1986; 2003), delen van de Self-Disclosure Index (SDI; - Miller, Berg & Archer, (1983), en om het hebben van geheimen te meten werd een definitie overgenomen van Frijns, Finkenauer, Vermulst en Engels (2005). Een significant positief verband is gevonden voor de relatie tussen emotionele disclosure en de mentale en fysieke gezondheid. Een significant negatief verband werd gevonden voor de relatie tussen geheimen en de gezondheid, waarbij zwaarte van de geheimen dit verband modereert. Geconcludeerd is dat meer emotionele disclosure in verband staat tot een verbeterde mentale- en fysieke gezondheid. Een omgekeerd verband is gevonden voor het hebben van geheimen. Het hebben van meer minder zware geheimen wordt gerelateerd aan een betere gezondheid. Aanbevolen interventies wijzen in de richting van gespreksgroepen die emotionele disclosure ten doel stellen alsmede verder longitudinaal onderzoek
