    Existence of smooth stable manifolds for a class of parabolic SPDEs with fractional noise

    Little seems to be known about the invariant manifolds for stochastic partial differential equations (SPDEs) driven by nonlinear multiplicative noise. Here we contribute to this aspect and analyze the Lu-Schmalfu{\ss} conjecture [Garrido-Atienza, et al., J. Differential Equations, 248(7):1637--1667, 2010] on the existence of stable manifolds for a class of parabolic SPDEs driven by nonlinear mutiplicative fractional noise. We emphasize that stable manifolds for SPDEs are infinite-dimensional objects, and the classical Lyapunov-Perron method cannot be applied, since the Lyapunov-Perron operator does not give any information about the backward orbit. However, by means of interpolation theory, we construct a suitable function space in which the discretized Lyapunov-Perron-type operator has a unique fixed point. Based on this we further prove the existence and smoothness of local stable manifolds for such SPDEs.Comment: To appear in Journal of Functional Analysi

    A re-evaluation of the tidal sorting hypothesis of mangrove zonation: propagule specific gravity matters

    There is much controversy surrounding factors that affect the distribution of mangrove plants across the intertidal gradient. It was previously hypothesized that mangrove zonation was attributed to tidal sorting (TSH) of its propagules according to size (weight) or differential ability of propagules to establish in deep water. However, observational and experimental evidence have provided little support for the actual mechanism(s) of mangrove zonation. In general, species distribution pattern is the consequence of propagule dispersal. The specific gravity of water-borne mangrove propagules may affect their buoyancy, with inherent links to dispersal, thereby potentially influencing tree zonation. Propagule specific gravity can influence the distribution of mangroves in the context of global change, particularly in response to changes in seawater salinity. In this study, we measured the specific gravity and weight of 35 mangrove species propagules. There was no correlation between the weight of the propagule and its specific gravity. The specific gravity of propagules of true mangrove species was significantly greater than that of semi-mangrove. The results of the correlation between propagule specific gravity and the relative surface elevation of field distribution support the hypothesis that propagules are subject to tidal sorting and are not related to weight but to specific gravity. This newfound understanding of mangrove dispersal and distribution is critical in the context of mangrove protection and restoration, especially in projecting the effects of anthropogenic activities and global change on mangrove communities

    PRL-3 promotes the motility, invasion, and metastasis of LoVo colon cancer cells through PRL-3-integrin β1-ERK1/2 and-MMP2 signaling

    <p>Abstract</p> <p>Background</p> <p>Phosphatase of regenerating liver-3 (PRL-3) plays a causative role in tumor metastasis, but the underlying mechanisms are not well understood. In our previous study, we observed that PRL-3 could decrease tyrosine phosphorylation of integrin β1 and enhance activation of ERK1/2 in HEK293 cells. Herein we aim to explore the association of PRL-3 with integrin β1 signaling and its functional implications in motility, invasion, and metastasis of colon cancer cell LoVo.</p> <p>Methods</p> <p>Transwell chamber assay and nude mouse model were used to study motility and invasion, and metastsis of LoVo colon cancer cells, respectively. Knockdown of integrin β1 by siRNA or lentivirus were detected with Western blot and RT-PCR. The effect of PRL-3 on integrin β1, ERK1/2, and MMPs that mediate motility, invasion, and metastasis were measured by Western blot, immunofluorencence, co-immunoprecipitation and zymographic assays.</p> <p>Results</p> <p>We demonstrated that PRL-3 associated with integrin β1 and its expression was positively correlated with ERK1/2 phosphorylation in colon cancer tissues. Depletion of integrin β1 with siRNA, not only abrogated the activation of ERK1/2 stimulated by PRL-3, but also abolished PRL-3-induced motility and invasion of LoVo cells in vitro. Similarly, inhibition of ERK1/2 phosphorylation with U0126 or MMP activity with GM6001 also impaired PRL-3-induced invasion. In addition, PRL-3 promoted gelatinolytic activity of MMP2, and this stimulation correlated with decreased TIMP2 expression. Moreover, PRL-3-stimulated lung metastasis of LoVo cells in a nude mouse model was inhibited when integrin β1 expression was interfered with shRNA.</p> <p>Conclusion</p> <p>Our results suggest that PRL-3's roles in motility, invasion, and metastasis in colon cancer are critically controlled by the integrin β1-ERK1/2-MMP2 signaling.</p

    Effects of Lactic Acid Stress on the Solid-state Fermentation of Pichia kudriavzevii

    Nongxiangxing Baijiu distiller’s grains are rich in lactic acid. Pichia kudriavzevii is the dominant yeast. The effects of lactic acid on the solid-state fermentation characteristics of Nongxiangxing Baijiu of the dominant yeast were explored under lactic acid stress of different concentrations. The growth and metabolism changes of Pichia kudriavzevii (PY1) were investigated from the aspects of carbon source consumption, growth factor utilization, high temperature tolerance and main metabolite production. Results showed that, the lactic acid stress had no significant effect on the colony and cell morphology of P. kudriavzevii, but had a certain inhibition on the growth of cell. This reversible inhibition could be weakened by amino acids and vitamins. When the culture temperature exceeds 44 ℃, the effect of lactic acid on the growth of the strain was gradually replaced by temperature. In metabolism, lactic acid could improve the utilization rate of glucose, galactose, maltose, fructose and sucrose, increase the production of ethanol and esters, and reduce the production of higher alcohols. In summary, the lactic acid could affect the flavor formation of Nongxiangxing Baijiu by affecting the growth and metabolism of yeast, and could be used as the fermentation regulation factor of Nongxiangxing Baijiu

    Synuclein gamma predicts poor clinical outcome in colon cancer with normal levels of carcinoembryonic antigen

    <p>Abstract</p> <p>Background</p> <p>Synuclein gamma (SNCG), initially identified as a breast cancer specific gene, is aberrantly expressed in many different malignant tumors but rarely expressed in matched nonneoplastic adjacent tissues. In this study, we investigated the prognostic potential of SNCG in colon cancer particularly in the patients with normal carcinoembryonic antigen (CEA) levels.</p> <p>Methods</p> <p>SNCG levels were assessed immunohistochemically in cancer tissues from 229 colon adenocarcinoma patients with a mean follow-up of 44 months. Correlations between SNCG levels and clinicopathologic features, preoperative serum CEA level, and clinical outcome were analyzed statistically using SPSS.</p> <p>Results</p> <p>SNCG levels in colon adenocarcinoma were closely associated with intravascular embolus and tumor recurrence but independent of preoperative serum CEA levels. SNCG expression was an independent prognostic factor of a shorter disease-free survival (DFS) and overall survival (OS) (<it>P </it>< 0.0001). Multivariate analysis revealed that both tissue SNCG and serum CEA were independent prognostic factors of DFS (<it>P </it>= 0.001, <0.0001, respectively) for 170 patients with colon adenocarcinomas. Importantly, SNCG remained a prognostic determinant of DFS and OS (<it>P </it>= 0.001, 0.002) for 97 patients with normal preoperative serum CEA level.</p> <p>Conclusions</p> <p>Our results suggest for the first time that SNCG is a new independent predicator for poor prognosis in patients with colon adenocarcinoma, including those with normal CEA levels. Combination of CEA with SNCG improves prognostic evaluation for patients with colon adenocarcinoma.</p

    The difference in mean arterial pressure induced by remimazolam compared to etomidate in the presence of fentanyl at tracheal intubation: A randomized controlled trial

    Background: Combined use of hypnotic and opioids during anesthesia inductions decreases blood pressure. Post-induction hypotension (PIHO) is the most common side effect of anesthesia induction. We aimed to compare the difference in mean arterial pressure (MAP) induced by remimazolam with that induced by etomidate in the presence of fentanyl at tracheal intubation.Methods: We assessed 138 adult patients with American Society of Anesthesiologists physical status I–II who underwent elective urological surgery. Patients were randomly allocated to receive either remimazolam or etomidate as alterative hypnotic in the presence of fentanyl during anesthesia induction. Comparable BIS values were achieved in both groups. The primary outcome was the difference in the MAP at tracheal intubation. The secondary outcomes included the characteristics of anesthesia, surgery, and adverse effects.Results: The MAP was higher in the etomidate group than in the remimazolam group at tracheal intubation (108 [22] mmHg vs. 83 [16] mmHg; mean difference, −26; 95% confidence interval [CI], −33 to −19; p &lt; 0.0001). Heart rate was significantly higher in the etomidate group than in the remimazolam group at tracheal intubation. The patients’ condition warranted the administration of ephedrine more frequently in the remimazolam group (22%) than in the etomidate group (5%) (p = 0.0042) during anesthesia induction. The remimazolam group had a lower incidence of hypertension (0% vs. 9%, p = 0.0133), myoclonus (0% vs. 47%, p &lt; 0.001), and tachycardia (16% vs. 35%, p = 0.0148), and a higher incidence of PIHO (42% vs. 5%, p = 0.001) than the etomidate group during anesthesia induction.Conclusion: Remimazolam was associated with lower MAP and lower heart rate compared to etomidate in the presence of fentanyl at tracheal intubation. Patients in the remimazolam group had a higher incidence of PIHO, and their condition warranted the administration of ephedrine more frequently than in the etomidate group during anesthesia induction

    Proteomic analysis of PBMCs: characterization of potential HIV-associated proteins

    <p>Abstract</p> <p>Background</p> <p>The human immunodeficiency virus type 1 (HIV-1) pandemic has continued unabated for nearly 30 years. To better understand the influence of virus on host cells, we performed the differential proteome research of peripheral blood mononuclear cells (PBMCs) from HIV-positive patients and healthy controls.</p> <p>Results</p> <p>26 protein spots with more than 1.5-fold difference were detected in two dimensional electrophoresis (2DE) gels. 12 unique up-regulated and one down-regulated proteins were identified in HIV-positive patients compared with healthy donors. The mRNA expression of 10 genes was analyzed by real time RT-PCR. It shows that the mRNA expression of talin-1, vinculin and coronin-1C were up-regulated in HIV positive patients and consistent with protein expression. Western blotting analysis confirmed the induction of fragments of vinculin, talin-1 and filamin-A in pooled and most part of individual HIV-positive clinical samples. Bioinformatic analysis showed that a wide host protein network was disrupted in HIV-positive patients.</p> <p>Conclusions</p> <p>Together, this work provided useful information to facilitate further investigation of the underlying mechanism of HIV and host cell protein interactions, and discovered novel potential biomarkers such as fragment of vinculin, filamin-A and talin-1 for anti-HIV research.</p
