BPR Implementation: A Decision-Making Strategy

    Get PDF
To support the efficient appraisal and selection of available best practices, this paper proposes a strategy for the implementation of Business Process Redesign (BPR). Its backbone is formed by the analytical hierarchy process (AHP) multi-criteria method and our earlier research on the popularity and impact of redesign best practices. Using (AHP) we derive a classification of most suitable best practices for the process being redesigned. Criteria such as the popularity, the impact, the goals and the risks of BPR implementation are taken into account. A case study of a municipality in the Netherlands is included. It discusses which best practices should be applied to redesign the invoicing process at the municipality. © Springer-Verlag Berlin Heidelberg 2006

Combined anti-AGEs and antioxidant activities of different solvent extracts of Solanum elaeagnifolium Cav. (Solanaceae) fruits during ripening and related to their phytochemical compositions

Oxidative stress and advanced glycation end products (AGEs) are known as key factors for the development of diabetic complications such as retinopathy, cataract as well as atherosclerosis and neurodegenerative diseases, including Alzheimer’s diseases. In this context, natural products have been previously identified as promising sources for antioxidant and anti-glycation compounds. The current study focuses on the evaluation of antioxidant and glycation inhibitory activities of different solvent extracts of Solanum elaeagnifolium Cav (Solanaceae) fruits at different ripening stages. The results showed that antioxidant and anti-AGEs activities were significantly influenced by solvents polarities and ripening stages of S. elaeagnifolium Cav. With one exception, methanolic extract of overripe S. elaeagnifolium Cav fruit showed important protective effects against cellular oxidative stress. The aqueous extract showed the highest ABTS+ scavenging ability. Principal component analysis showed that total phenolic and flavonoid contents correlated well with observed antioxidants and anti-glycation activities. These results bring attention to the possible use of S. elaeagnifolium Cav as a valuable source of bioactive compounds exhibiting antioxidant effects and potentially alleviating diabetic complications

Partitioning clustering algorithms for protein sequence data sets

Abstract

Background

Genome-sequencing projects are currently producing an enormous amount of new sequences and cause the rapid increasing of protein sequence databases. The unsupervised classification of these data into functional groups or families, clustering, has become one of the principal research objectives in structural and functional genomics. Computer programs to automatically and accurately classify sequences into families become a necessity. A significant number of methods have addressed the clustering of protein sequences and most of them can be categorized in three major groups: hierarchical, graph-based and partitioning methods. Among the various sequence clustering methods in literature, hierarchical and graph-based approaches have been widely used. Although partitioning clustering techniques are extremely used in other fields, few applications have been found in the field of protein sequence clustering. It is not fully demonstrated if partitioning methods can be applied to protein sequence data and if these methods can be efficient compared to the published clustering methods.

Methods

We developed four partitioning clustering approaches using Smith-Waterman local-alignment algorithm to determine pair-wise similarities of sequences. Four different sets of protein sequences were used as evaluation data sets for the proposed methods.

Results

We show that these methods outperform several other published clustering methods in terms of correctly predicting a classifier and especially in terms of the correctness of the provided prediction. KSHV gB associated RGD interactions promote attachment of cells by inhibiting the potential migratory signals induced by the disintegrin-like domain

Background: Kaposi's sarcoma-associated herpesvirus (KSHV) glycoprotein B (gB) is not only expressed on the envelope of mature virions but also on the surfaces of cells undergoing lytic replication. Among herpesviruses, KSHV gB is the only glycoprotein known to possess the RGD (Arg-Gly-Asp) binding integrin domain critical to mediating cell attachment. Recent studies described gB to also possess a disintegrin-like domain (DLD) said to interact with non-RGD binding integrins. We wanted to decipher the roles of two individually distinct integrin binding domains (RGD versus DLD) within KSHV gB in regulating attachment of cells over cell migration

Snake Venom Disintegrins and Cell Migration

    Get PDF
Cell migration is a key process for the defense of pluricellular organisms against pathogens, and it involves a set of surface receptors acting in an ordered fashion to contribute directionality to the movement. Among these receptors are the integrins, which connect the cell cytoskeleton to the extracellular matrix components, thus playing a central role in cell migration. Integrin clustering at focal adhesions drives actin polymerization along the cell leading edge, resulting in polarity of cell movement. Therefore, small integrin-binding proteins such as the snake venom disintegrins that inhibit integrin-mediated cell adhesion are expected to inhibit cell migration. Here we review the current knowledge on disintegrin and disintegrin-like protein effects on cell migration and their potential use as pharmacological tools in anti-inflammatory therapy as well as in inhibition of metastatic invasion

Genomic insights into members of the candidate phylum Hyd24-12 common in mesophilic anaerobic digesters

    Get PDF
Members of the candidate phylum Hyd24-12 are globally distributed, but no genomic information or knowledge about their morphology, physiology or ecology is available. In this study, members of the Hyd24-12 lineage were shown to be present and abundant in full-scale mesophilic anaerobic digesters at Danish wastewater treatment facilities. In some samples, a member of the Hyd24-12 lineage was one of the most abundant genus-level bacterial taxa, accounting for up to 8% of the bacterial biomass. Three closely related and near-complete genomes were retrieved using metagenome sequencing of full-scale anaerobic digesters. Genome annotation and metabolic reconstruction showed that they are Gram-negative bacteria likely involved in acidogenesis, producing acetate and hydrogen from fermentation of sugars, and may play a role in the cycling of sulphur in the digesters. Fluorescence in situ hybridization revealed single rod-shaped cells dispersed within the flocs. The genomic information forms a foundation for a more detailed understanding of their role in anaerobic digestion and provides the first insight into a hitherto undescribed branch in the tree of life

Reliability analysis of an RC member impacted by a rockfall using a nonlinear SDOF model

International audienceThis paper presents the reliability analysis of a reinforced concrete (RC) member subjected to an impact arising from a rockfall in order to derive fragility curves dedicated to risk analysis framework. A dynamic model of a simply supported RC member accounting for material and contact nonlinearities has been proposed. In a reliability analysis, several runs of the deterministic model are needed in order to assess the failure probability of the system. Hence ad hoc assumptions are proposed in the aim of reducing the computational cost of the deterministic model, while main physical phenomena involved during the impact are taken into account. Thus Moment–Curvature approach is used and validated with Finite Element Method (FEM) calculations under quasi-static and dynamic loadings. Failure probability is assessed by coupling Monte Carlo, Important Sampling and First Order Reliability Method (FORM) with the simplified deterministic model. Finally, fragility curves are derived which allow underlining the influence of several parameters related either to the loading or the RC member
