    Strategies for Systemic Change:Youth Community Organizing to Disruptthe School-to-Prison Nexus

    The school disciplinary landscape across the United States changed significantly through the enactment of policies that criminalize students’ behaviors during the 1990s and 2000s. Schools began to involve the police and criminal legal system in school disciplinary issues that used to be handled by school administrators. This shift led youth of Color1 to increasingly come into contact with the juvenile legal system through school suspensions, expulsions, and referrals to alternative schools—what we characterize as the school-toprison nexus. Conceptualizing the school-to-prison pipeline as a nexus, or interlocking system of power over youth, allows us to understand how the criminalization of youth is a systemic problem that demands structural change and interventions across multiple levels of analysis and settings, including local schools, school districts, police departments, and state policies. Although important research has documented the ways that Black and Latino youth are referred to the juvenile legal system through punitive school policies, there has been less attention to the actions youth are taking to critique and dismantle these policies. Youth community organizing (YCO) against the school-to-prison nexus represents an arena of youth activism that deserves further attention and analysis. In this chapter, we define YCO as groups that create spaces for young people to think critically about their everyday social conditions, identify root causes of social problems, and build political power and voice to create policy solutions and change in their communities (Ginwright, Noguera, & Cammarota, 2006; Kirshner, 2015; Watts, Griffith, & Abdul- Adil, 1999)

    Where our Girls at? The Misrecognition of Black and Brown girls in schools

    Black and brown girls remain too often at the margins not only in society at large and in our schools but also in our research and writing about schools. Herein we argue for careful consideration of the specific ways that their raced and gendered identities render these girls vulnerable and put them in jeopardy so that educators and scholars do not become complicit in their marginalization. We focus on dynamics of invisibility and hypervisibility. While these dynamics may seem to be diametrically opposite, both involve the process of what scholar Nancy Fraser (2000) calls “misrecognition” (p. 113)

    Genome-Wide Identification of Transcriptional Start Sites in the Plant Pathogen Pseudomonas syringae pv. tomato str. DC3000

    RNA-Seq has provided valuable insights into global gene expression in a wide variety of organisms. Using a modified RNA-Seq approach and Illumina's high-throughput sequencing technology, we globally identified 5â€Č-ends of transcripts for the plant pathogen Pseudomonas syringae pv. tomato str. DC3000. A substantial fraction of 5â€Č-ends obtained by this method were consistent with results obtained using global RNA-Seq and 5â€ČRACE. As expected, many 5â€Č-ends were positioned a short distance upstream of annotated genes. We also captured 5â€Č-ends within intergenic regions, providing evidence for the expression of un-annotated genes and non-coding RNAs, and detected numerous examples of antisense transcription, suggesting additional levels of complexity in gene regulation in DC3000. Importantly, targeted searches for sequence patterns in the vicinity of 5â€Č-ends revealed over 1200 putative promoters and other regulatory motifs, establishing a broad foundation for future investigations of regulation at the genomic and single gene levels

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∌99% of the euchromatic genome and is accurate to an error rate of ∌1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead
