The goals of the present study were, on the one hand, to determine the anthropometric, strength (measured by jump and hand-grip tests) compared to other racquet sports, and on the other hand, to analyze the time-motion characteristics during the under-23Spanish Paddle championships. Thus, 14 men (20,38 ± 1,92 yr) and 12 women(19.73 ± 2,37 yr) participated in this study. Firstly, participants were assessed foranthropometric measurements; secondly, strength was measured by jump andhand-grip tests before and immediately after the match; finally, the time-motion ofmatches were quantified during the under-23 Spanish Championship in 2012. Theanthropometric characteristics of paddle players were slightly different in comparisonto players of other racket sports. In respect of strength parameters, there were notsignificant differences between pre-match and post-match measurements, indicatingno fatigue component after competition. In respect to time motion analyses, restingperiods were significantly longer compared to the effective playing time. Paddle seemsto have less time active than table-tennis and badminton, but more time active thantennis.

Genomic and functional regulation of TRIB1 contributes to prostate cancer pathogenesis

Prostate cancer is the most frequent malignancy in European men and the second worldwide. One of the major oncogenic events in this disease includes amplification of the transcription factor cMYC. Amplification of this oncogene in chromosome 8q24 occurs concomitantly with the copy number increase in a subset of neighboring genes and regulatory elements, but their contribution to disease pathogenesis is poorly understood. Here we show that TRIB1 is among the most robustly upregulated coding genes within the 8q24 amplicon in prostate cancer. Moreover, we demonstrate that TRIB1 amplification and overexpression are frequent in this tumor type. Importantly, we find that, parallel to its amplification, TRIB1 transcription is controlled by cMYC. Mouse modeling and functional analysis revealed that aberrant TRIB1 expression is causal to prostate cancer pathogenesis. In sum, we provide unprecedented evidence for the regulation and function of TRIB1 in prostate cancer

Extracellular enzyme production in the coastal upwelling system off Peru during different upwelling scenarios: a mesocosm experiment

The Peruvian upwelling system is a highly productive ecosystem that could be altered by ongoing global changes. We carried out a mesocosm experiment off Peru, with the addition of water masses from the regional oxygen minimum zone (OMZ) collected at two different sites simulating two different upwelling scenarios. Here we focus on pelagic remineralization of organic matter by extracellular enzyme production of leucine aminopeptidase (LAP) and alkaline phosphatase activity (APA). After addition of the OMZ water, dissolved inorganic nitrogen (N) was depleted, but the standing stock of phytoplankton was relatively high even after nutrient depletion (mostly >4 µg chlorophyll a L-1). During the initial phase of the experiment, APA was 0.6 nmol L-1 h-1 even though the PO43- concentration was >0.5 µmol L-1. Initially, the dissolved organic phosphorus (DOP) decreased, coinciding with an increase in PO43- concentration probably linked to the APA. The LAP activity was very high with most of the measurements in the range 200–800 nmol L-1 h-1. This enzyme degrades amino acids and these high values are probably linked to the highly productive, but N-limited coastal ecosystem. Also, the experiment took place during a rare coastal El Niño event with higher-than-normal surface temperatures, which could have affected the enzyme production. Using a non-parametric multidimensional scaling analysis (NMDS) with a generalized additive model (GAM), we found that biogeochemical variables (e.g. nutrient and chlorophyll a concentrations), phytoplankton and bacterial communities explained up to 64 % of the variability in APA. The bacterial community explained best the variability (34 %) in LAP. The high hydrolysis rates for this enzyme suggests that pelagic N remineralization supported the high standing stock of primary producers in the mesocosms after N depletion.</p

    Angiocrine polyamine production regulates adiposity.

Reciprocal interactions between endothelial cells (ECs) and adipocytes are fundamental to maintain white adipose tissue (WAT) homeostasis, as illustrated by the activation of angiogenesis upon WAT expansion, a process that is impaired in obesity. However, the molecular mechanisms underlying the crosstalk between ECs and adipocytes remain poorly understood. Here, we show that local production of polyamines in ECs stimulates adipocyte lipolysis and regulates WAT homeostasis in mice. We promote enhanced cell-autonomous angiogenesis by deleting Pten in the murine endothelium. Endothelial Pten loss leads to a WAT-selective phenotype, characterized by reduced body weight and adiposity in pathophysiological conditions. This phenotype stems from enhanced fatty acid β-oxidation in ECs concomitant with a paracrine lipolytic action on adipocytes, accounting for reduced adiposity. Combined analysis of murine models, isolated ECs and human specimens reveals that WAT lipolysis is mediated by mTORC1-dependent production of polyamines by ECs. Our results indicate that angiocrine metabolic signals are important for WAT homeostasis and organismal metabolism. We thank members of the Endothelial Pathobiology and Microenvironment Group for helpful discussions. METTL1 promotes tumorigenesis through tRNA-derived fragment biogenesis in prostate cancer

Newly growing evidence highlights the essential role that epitranscriptomic marks play in the development of many cancers; however, little is known about the role and implications of altered epitranscriptome deposition in prostate cancer. Here, we show that the transfer RNA N-7-methylguanosine (m(7)G) transferase METTL1 is highly expressed in primary and advanced prostate tumours. Mechanistically, we find that METTL1 depletion causes the loss of m(7)G tRNA methylation and promotes the biogenesis of a novel class of small non-coding RNAs derived from 5'tRNA fragments. 5'tRNA-derived small RNAs steer translation control to favour the synthesis of key regulators of tumour growth suppression, interferon pathway, and immune effectors. Knockdown of Mettl1 in prostate cancer preclinical models increases intratumoural infiltration of pro-inflammatory immune cells and enhances responses to immunotherapy. Collectively, our findings reveal a therapeutically actionable role of METTL1-directed m(7)G tRNA methylation in cancer cell translation control and tumour biology

Nitrogen removal in domestic wastewater. Effect of nitrate recycling and COD/N ratio

A denitrification/nitrification pilot plant was designed, built and put into operation, treating the effluent of an anaerobic reactor. The operation of the plant examined the effect of the nitrate recycling and the COD/N ratio on the nitrogen and the remaining organic matter removal at 18 °C. The system consisted of a two-stage treatment process: anoxic and aerobic. The hydraulic retention time (HRT) of the system was 1 h for the anoxic bioreactor and 2 h for the aerobic one. The increase in the nitrate recycling ratio did not cause a significant improvement in the nitrogen removal due to the insufficient carbon source. The wastewater to be treated had a C/N ratio of 1.1 showing a lack of organic carbon. The addition of methanol was a key point in the denitrification process used as a model for the traditional wastewater by-pass in the WWTP. The maximum nitrogen and organic matter removal (87.1% and 96%, respectively) was achieved with a nitrate recycling ratio of 600% and a C/N of 8.25, adjusted by methanol addition.Cadagua S.A. company, the European Regional Development Fund, the IPT-2011-1078-310000 project, and the INNPACTO 2011 program of the Ministry of Economy and Competitivenes
