Ecologia e extrativismo de plantas utilizadas como fixadoras de corantes no artesanato Baniwa, alto rio Negro, Amazonas
Neste trabalho descrevi modos de uso e a estrutura populacional das espécies Inga spp., Miconia dispar e Miconia minutiflora em cronosseqüências de capoeiras Baniwa. A estrecasca destas árvores é utilizada para dar brilho e fixar corantes naturais nos talos de Arumã (Ischnosiphon spp.) utilizados no artesanato Baniwa. Comparei padrões de demanda e disponibilidade do recurso entrecasca em comunidades Baniwa, grupo que está envolvido em projeto de comercialização de sua arte tradicional desde o ano de 1998. Assim, o objetivo maior desta pesquisa foi produzir informações sobre a sustentabilidade da exploração destas espécies fornecedoras de fixadores de tintas. Amostrei 38 transectos em capoeiras com idades que variaram entre 5 a 60 anos em 3 comunidades distribuídas ao longo da bacia do rio Içana, Território Indígena do Alto Rio Negro. Eu caracterizei a idade do local através de entrevistas com os Baniwa, a estrutura das populações das espécies ao longo dos transectos e quantifiquei a disponibilidade e o consumo atual de entrecasca pelos artesãos nas comunidades Baniwa estudadas. Também observei evidências de extração e manejo. As densidades de M. minutiflora, M. dispar e Inga spp. tenderam a declinar com o avanço da idade das capoeiras e populações mostraram diferenças em suas distribuições e estrutura nos diferentes sítios amostrados. O consumo atual de entrecasca nas 2 comunidades estudadas foi extremamente baixo quando comparado com os estoques disponíveis. A não ser que as taxas de exploração aumentem substancialmente, não existe a necessidade imediata de iniciar um programa de monitoramento para estas espécies. Entretanto, diferenças na distribuição das espécies de plantas fixadoras ao longo do território Baniwa e as diferentes densidades humanas e produtividade dos artesãos de cada comunidade determinam a necessidade de adequar eventuais medidas de manejo para as realidades encontradas em cada comunidade Baniwa
Serological Survey of Antibodies to Mannheimia haemolytica and Pasteurella multocida in Camelids from Argentina
South American camelids are a source of livestock wealth in Andean countries. In Argentina,there is little information about camelid pathogens, and most of the literature data available areseroprevalence works against virus. Besides, little is known about the immunological status againstbacterial agents affecting these animals. In an effort to explore the serological status of Argentineancamelids, we evaluated the presence of serum antibodies against bacterial pathogens involved inpneumonic diseases (Pasteurella multocida and Mannheimia haemolytica) in llamas from differentregions of the country. By ELISA, a high seroprevalence for both pathogens was found in the serumsamples; higher optical density (OD) values were obtained when the sera were incubated with heatkilledP. multocida as coating antigen compared to M. haemolytica. In addition, a large number ofsera analyzed presented high OD values for both microorganisms independently of their originregion. Serum avidity was also evaluated, by means of an assay based on antibody desorption byurea. No correlation was found between the high ODs obtained for P. multocida and the serumavidity. On the other hand, samples reacting with M. haemolytica had lower OD values but higheravidity index. The antigenic recognition pattern for both microorganisms was determined bywestern blot. Unlike P. multocida, the antigenic recognition pattern of M. haemolytica did not differamong serum samples obtained from animals living in different areas. In summary, we found thatcamelids can synthetize antibodies that recognize M. haemolytica with high avidity for differentantigens of the bacterium, suggesting that Argentinean camelids are in contact with M. haemolyticawhich is probably a causative agent of subclinical infections. Immune System Modulation Produced by Ultraviolet Radiation
Exposure to ultraviolet radiation (UVr) contained in sunlight is a major cause of skin illness such as sunburn, aging and cancer. UVr triggers local effects on the skin, which involve local inflammation, tissue remodeling, regulatory cytokines release and migration of dendritic cells (DCs). However, these localized effects on the exposed area are not the only ones that take place after sun or UVr exposure. A less known effect of UVr is the modulation of systemic immunity, through the generation of specific regulatory cells. These cells are induced, at least in part, by skin-migrating tolerogenic DCs. Moreover, bone marrow cell precursors can also be biased to a tolerogenic or suppressor phenotype. The sunlight- or UVr-induced immune system modulation can cause skin disorders like skin cancer and cutaneous photosensitivity in Lupus, but it also may be useful to treat cutaneous pathologies such as psoriasis and vitiligo. Moreover, the systemic immunosuppressor effect of UVr exposure may also be useful to treat autoimmunity of internal organs. Finally, as an inducer of cutaneous inflammatory response, UV phototherapy may also be useful in the treatment of cutaneous infections. Overall, UVr has profound immunomodulatory capacity that can be beneficial or deleterious for human health
Lipoteichoic Acid from Lacticaseibacillus rhamnosus GG Modulates Dendritic Cells and T Cells in the Gut
Lipoteichoic acid (LTA) from Gram-positive bacteria exerts different immune effects depending on the bacterial source from which it is isolated. Lacticaseibacillus rhamnosus GG LTA (LGG-LTA) oral administration reduces UVB-induced immunosuppression and skin tumor development in mice. In the present work, we evaluate the immunomodulatory effect exerted by LGG-LTA in dendritic cells (DC) and T cells, both in vitro and in the gut-associated lymphoid tissue (GALT). During cell culture, LTA-stimulated BMDC increased CD86 and MHC-II expression and secreted low levels of pro and anti-inflammatory cytokines. Moreover, LTA-treated BMDC increased T cell priming capacity, promoting the secretion of IL-17A. On the other hand, in orally LTA-treated mice, a decrease in mature DC (lamina propria and Peyer’s patches) was observed. Concomitantly, an increase in IL-12p35 and IFN-γ transcription was presented (lamina propria and Peyer’s Patches). Finally, an increase in the number of CD103+ DC was observed in Peyer’s patches. Together, our data demonstrate that LGG-LTA activates DC and T cells. Moreover, we show that a Th1-biased immune response is triggered in vivo after oral LTA administration. Caracterización de IgM, IgG Total, IgG1 y anticuerpos de cadena pesada en calostro de llamas ("Lama glama") mediante Elisa
Objectives: to determine the levels of IgM, total IgG, IgG1 and HCAbs (IgG2 and IgG3) in colostra of llamas and to evaluate the concentration of HCAbs in relation to total IgG and the conventional isotype IgG1. Methods: Fifteen pregnant llamas were used in this study, which were milked within the first 24 hours after delivery. Sandwich ELISAs were designed for the quantification of IgM, total IgG and IgG1. The HCAbs concentration was calculated by difference between total IgG and IgG1 (HCAbs = total IgG – IgG1). Results: The levels of Igs were found to be: IgM= 17.02 mg/ml (SD= 9.85), total IgG= 42.54mg/ml (SD=27.79), IgG1=24.34mg/ml (SD=13.96) and HCAbs (IgGtotal-IgG1) = 18.19 mg/ml (SD=15.49). Results regarding total IgG are consistent with those described in other species with similar type of placentation. HCAbs represented 43% of colostral IgG and IgM concentration presented values that were much higher (two and three times respectively) than those described for species with similar type of placentation. Conclusions: This study is the first which evaluate amounts of each immunoglobulin isotype in colostra of llamas. We described a higher value of IgM in in comparison with species with similar placentation type. The IgG1 isotype, which represents the conventional isotype, is the more abundant antibody in colostra, being the HCAbs the second important fraction. Two Approaches to the Study of a Controversial Relationship: Cutaneous Photosensitivity and Anti-Ro/SS-A Autoantibodies
Background: Autoantibodies (Aabs) are the hallmark of numerous systemic autoimmune pathologies (SAPs), for instance anti-Ro/SS-A Aabs are usually found in Systemic Lupus Erythematosus (SLE) and Sjogren’s Syndrome. Cutaneous photosensitivity (CP) is found in most forms and subsets of LE and consists of a skin rash as a result of unusual reaction to sunlight. There are many theories which relate specifically the presence of circulating anti-Ro/SS-A Aabs with the CP phenomenon, though there are several studies which are in disagreement. Results: In this study we analyzed the relationship between CP and anti-Ro Aabs by means of two approaches. The first one included an in vitro model where we evaluated by flow cytometry the binding capacity of affinity-purified Aabs to autoantigens relocalized on apoptotic keratinocyte’s surface. We found that there was no relationship between the binding capacity of serum from 10 selected patients or their corresponding purified anti-Ro52 and anti-Ro60 Aabs, and the presence or absence of CP, neither with the SAPs. The in vivo model consisted of Hairless SKH:1 mice which were induced to produce anti-murine Ro52 and/or Ro60 Aabs and were subsequently irradiated with UVB light. We evaluated the skin histology and also the epidermal production of TNF-α. We found no differences between the groups in neither of the parameters evaluated. Conclusions: These results agree with some studies on the role of the Aabs in CP, considering anti-Ro Aabs not as the only responsible for the manifestation; and disagree with many other authors, who believe in the strong association between these two events.Fil: Paz, Mariela Laura. C3, C5a and anti-acetylcholine receptor antibody as severity biomarkers in myasthenia gravis
Background: Although the pathogenesis of myasthenia gravis (MG) is well known, prognostic markers are not yet available. We assessed the utility of anti-acetylcholine receptor (AChR) antibody (AChR-ab) titer and concentration of C3, C4, and C5a as potential severity biomarkers in MG. Methods: Levels of C3, C4, C5a, and AChR-ab were measured in 60 AChR-ab-positive patients with MG. Their relationship with clinical severity was analyzed using the activities of daily living (ADL) and MG composite (MGC) scales. Results: AChR-ab titer correlated with severity of MG according to ADL (p = 0.002) and MGC scales (p = 0.001). When patients were classified according to disease duration, a statistically significant correlation between AChR-ab titer and clinical severity was only found in the subgroup of patients with fewer than 5 years from symptoms onset. C5a levels showed a positive correlation with MG severity according to the ADL scale (p = 0.041; τb = 0.18), although C5a levels were not different from the control group. Sacolas retornáveis: uma alternativa para redução do impacto de larga escala causado pela eliminação irregular de sacolas descartáveis
This study aimed to determine the receptivity and acceptance of customers in a supermarket chain in relation to the adoption of reusable bags as a strategy for environmental marketing. Therefore, we conducted a case study by implementing a structured questionnaire. The questionnaire was administered to a non-probability sample of 150 customers of a supermarket chain, located in the town of Santa Maria, RS. The results were positive regarding customer acceptance of replacement of disposable bags for reusable bags, however, despite the acceptance is high, adherence is still low, since most customers forget to take reusable bags when they go shopping. This result demonstrates the need to increase investments in environmental education, to raise awareness about the need and importance of protecting and preserving the environment.O presente trabalho objetivou verificar a receptividade e aceitação dos clientes de uma rede de supermercados, em relação à adoção de sacolas retornáveis como estratégia de marketing ambiental. Para tanto, foi realizado um estudo de caso por meio da aplicação de um questionário estruturado. O questionário foi aplicado a uma amostra não probabilística de 150 clientes de uma rede de supermercados situada na cidade de Santa Maria, RS. Os resultados foram positivos quanto à aceitação pelos clientes da substituição das sacolas descartáveis por sacolas retornáveis, entretanto, a pesar da aceitação ser alta, a adesão ainda é baixa, uma vez que a maioria dos clientes se esquece de levar as sacolas retornáveis quando vão às compras. Esse resultado demonstra a necessidade de ampliar os investimentos em Educação Ambiental, para conscientizar a população quanto à necessidade e importância da proteção e preservação do meio ambiente
