    Paving Block from Residue of PS/LDPE/PP Plastic Pyrolysis Mixed with Palm Oil Bottom Ash (POBA)

    The residue of plastic mixture pyrolysis can be used for making paving blocks. Fibers in plastics can be used as an adhesive against other materials and can increase the strength of paving blocks. Palm oil bottom ash (POBA) was added to replace the role of cement, which has the same content as cement, i.e., silica (Si). This study aimed to see the effect of palm oil bottom ash and sand added to the residue of plastic mixture pyrolysis (i.e., PS/LDPE/PP) on the quality of paving blocks. The paving block as a test object was made by combining two materials. The residue of plastic mixture pyrolysis and LDPE plastic melt was mixed with a mass ratio of 70:30%, referred to as material I. Furthermore, palm oil bottom ash and sand were mixed with a mass ratio of 100%, 80%, 60%, 40%, and 20%, referred to as material II. The ratio of ingredients I and II used was 1: 1. Testing of the quality of paving blocks includes a compressive strength test and a water absorption test. The test results showed that the ratio of PS/LDPE/PP mixture (50:25:25) had a compressive strength of 104.1 kg/cm2 or 102.08 MPa and the water absorption of 2.4% at the ratio of palm oil bottom ash and sand at 80:20%. Adding palm oil bottom ash can reduce the paving block’s compressive strength, and the residue’s LDPE content can affect water absorption because of its chemical structure properties. Therefore, a reduction in the amount of LDPE residue is needed to obtain optimal water absorption values in paving blocks and add palm oil bottom ash.     Assembly and Functional Architecture of Bacterial Chemoreceptor Nanoarrays

    Transmembrane chemotaxis receptors are found in bacteria in extended hexagonal arrays stabilized by the membrane and by cytosolic binding partners, the kinase CheA and coupling protein CheW. Models of array architecture and assembly propose receptors cluster into trimers-of-dimers that associate with one CheA dimer and two CheW monomers to form the minimal core unit necessary for signal transduction. Reconstructing in vitro chemoreceptor ternary complexes that are homogenous, functional, and exhibit native architecture remains a challenge. Here we report that His-tag mediated receptor dimerization with divalent metals is sufficient to drive assembly of native-like functional arrays of a receptor cytoplasmic fragment. Our results indicate receptor dimerization initiates assembly and precedes formation of ternary complexes with partial kinase activity. Restoration of maximal kinase activity coincides with a shift to larger complexes, suggesting that kinase activity depends on interactions beyond the core unit. We hypothesize that achieving maximal activity requires building core units into hexagons and/or coalescing hexagons into the extended lattice. This discovery may also address a previously observed density-dependent transition between signaling states. To further test this, we implemented a paramagnetic relaxation enhancement (PRE) based solid-state NMR approach to obtain long-range (≥ 20 Å) distance constraints across the trimer of dimers interface. Overall, the work presented here shows that minimally perturbing His-tag mediated dimerization promotes assembly of chemoreceptor arrays with native architecture, and thus enabled us to gain insights into the mode of array assembly and the role of the core functional unit

    Use of agro-climatic zones to upscale simulated crop yield potential

    Yield gap analysis, which evaluates magnitude and variability of difference between crop yield potential (Yp) or water limited yield potential (Yw) and actual farm yields, provides a measure of untapped food production capacity. Reliable location-specific estimates of yield gaps, either derived from research plots or simulation models, are available only for a limited number of locations and crops due to cost and time required for field studies or for obtaining data on long-term weather, crop rotations and management practices, and soil properties. Given these constraints, we compare global agro-climatic zonation schemes for suitability to up-scale location-specific estimates of Yp and Yw, which are the basis for estimating yield gaps at regional, national, and global scales. Six global climate zonation schemes were evaluated for climatic homogeneity within delineated climate zones (CZs) and coverage of crop area. An efficient CZ scheme should strike an effective balance between zone size and number of zones required to cover a large portion of harvested area of major food crops. Climate heterogeneity was very large in CZ schemes with less than 100 zones. Of the other four schemes, the Global Yield Gap Atlas Extrapolation Domain (GYGA-ED) approach, based on a matrix of three categorical variables (growing degree days, aridity index, temperature seasonality) to delineate CZs for harvested area of all major food crops, achieved reasonable balance between number of CZs to cover 80% of global crop area and climate homogeneity within zones. While CZ schemes derived from two climate-related categorical variables require a similar number of zones to cover 80% of crop area, within-zone heterogeneity is substantially greater than for the GYGA-ED for most weather variables that are sensitive drivers of crop production. Some CZ schemes are cropspecific, which limits utility for up-scaling location-specific evaluation of yield gaps in regions with crop rotations rather than single crop species

    Attachment theory, developed by child psychiatrist John Bowlby, is considered a major theory in developmental psychology. Attachment theory can be seen as resulting from Bowlby's personal experiences, his psychoanalytic education, his subsequent study of ethology, and societal developments during the 1930s and 1940s. One of those developments was the outbreak of World War II and its effects on children's psychological wellbeing. In 1950, Bowlby was appointed WHO consultant to study the needs of children who were orphaned or separated from their families for other reasons and needed care in foster homes or institutions. The resulting report is generally considered a landmark publication in psychology, although it subsequently met with methodological criticism. In this paper, by reconstructing Bowlby's visit to several European countries, on the basis of notebooks and letters, the authors shed light on the background of this report and the way Bowlby used or neglected the findings he gathered

    Adaptive Evolution of a Stress Response Protein

    Some cancers are mediated by an interplay between tissue damage, pathogens and localised innate immune responses, but the mechanisms that underlie these linkages are only beginning to be unravelled.Here we identify a strong signature of adaptive evolution on the DNA sequence of the mammalian stress response gene SEP53, a member of the epidermal differentiation complex fused-gene family known for its role in suppressing cancers. The SEP53 gene appears to have been subject to adaptive evolution of a type that is commonly (though not exclusively) associated with coevolutionary arms races. A similar pattern of molecular evolution was not evident in the p53 cancer-suppressing gene.Our data thus raises the possibility that SEP53 is a component of the mucosal/epithelial innate immune response engaged in an ongoing interaction with a pathogen. Although the pathogenic stress mediating adaptive evolution of SEP53 is not known, there are a number of well-known candidates, in particular viruses with established links to carcinoma

    An animal model to evaluate the function and regulation of the adaptively evolving stress protein SEP53 in oesophageal bile damage responses

    Squamous epithelium in mammals has evolved an atypical stress response involving down-regulation of the classic HSP70 protein and induction of sets of proteins including one named SEP53. This atypical stress response might be due to the unusual environmental pressures placed on squamous tissue. In fact, SEP53 plays a role as an anti-apoptotic factor in response to DNA damage induced by deoxycholic acid stresses implicated in oesophageal reflux disease. SEP53 also has a genetic signature characteristic of an adaptively and rapidly evolving gene, and this observation has been used to imply a role for SEP53 in immunity. Physiological models of squamous tissue are required to further define the regulation and function of SEP53. We examined whether porcine squamous epithelium would be a good model to study SEP53, since this animal suffers from a bile-reflux disease in squamous oesophageal tissue. We have (1) cloned and sequenced the porcine SEP53 locus from porcine bacterial artificial chromosome genomic DNA, (2) confirmed the strikingly divergent nature of the C-terminal portion of the SEP53 gene amongst mammals, (3) discovered that a function of the conserved N-terminal domain of the gene is to maintain cytoplasmic localisation, and (4) examined SEP53 expression in normal and diseased porcine pars oesophagea. SEP53 expression in porcine tissue was relatively confined to gastric squamous epithelium, consistent with its expression in normal human squamous epithelium. Immunohistochemical staining for SEP53 protein in normal and damaged pars oesophagea demonstrated significant stabilisation of SEP53 protein in the injured tissue. These results suggest that porcine squamous epithelium would be a robust physiological model to examine the evolution and function of the SEP53 stress pathway in modulating stress-induced responses in squamous tissue

    Mortality from gastrointestinal congenital anomalies at 264 hospitals in 74 low-income, middle-income, and high-income countries: a multicentre, international, prospective cohort study

    Summary Background Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. Methods We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung’s disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. Findings We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung’s disease) from 264 hospitals (89 in high-income countries, 166 in middleincome countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). Mortality among all patients was 37 (39·8%) of 93 in low-income countries, 583 (20·4%) of 2860 in middle-income countries, and 50 (5·6%) of 896 in high-income countries (p<0·0001 between all country income groups). Gastroschisis had the greatest difference in mortality between country income strata (nine [90·0%] of ten in lowincome countries, 97 [31·9%] of 304 in middle-income countries, and two [1·4%] of 139 in high-income countries; p≤0·0001 between all country income groups). Factors significantly associated with higher mortality for all patients combined included country income status (low-income vs high-income countries, risk ratio 2·78 [95% CI 1·88–4·11], p<0·0001; middle-income vs high-income countries, 2·11 [1·59–2·79], p<0·0001), sepsis at presentation (1·20 [1·04–1·40], p=0·016), higher American Society of Anesthesiologists (ASA) score at primary intervention (ASA 4–5 vs ASA 1–2, 1·82 [1·40–2·35], p<0·0001; ASA 3 vs ASA 1–2, 1·58, [1·30–1·92], p<0·0001]), surgical safety checklist not used (1·39 [1·02–1·90], p=0·035), and ventilation or parenteral nutrition unavailable when needed (ventilation 1·96, [1·41–2·71], p=0·0001; parenteral nutrition 1·35, [1·05–1·74], p=0·018). Administration of parenteral nutrition (0·61, [0·47–0·79], p=0·0002) and use of a peripherally inserted central catheter (0·65 [0·50–0·86], p=0·0024) or percutaneous central line (0·69 [0·48–1·00], p=0·049) were associated with lower mortality. Interpretation Unacceptable differences in mortality exist for gastrointestinal congenital anomalies between lowincome, middle-income, and high-income countries. Improving access to quality neonatal surgical care in LMICs will be vital to achieve Sustainable Development Goal 3.2 of ending preventable deaths in neonates and children younger than 5 years by 2030

    La obesidad es una enfermedad crónica, caracterizada por el aumento de la grasa corporal, asociada a un mayor riesgo para la salud. La cirugía bariátrica es actualmente la estrategia más eficaz para conseguir una pérdida de peso significativa y sostenible, resolviendo comorbilidades y reduciendo la mortalidad a largo plazo. Se desconoce el impacto que puede tener los pacientes operados de cirugía bariátrica con relación a su calidad de vida. Objetivos: Evaluar la calidad de vida de los pacientes operados de cirugía bariátrica en el Servicio de Cirugía General del CMDLT. Metodología: estudio retrospectivo, observacional y analítico de cohorte. Aplicado en personas con obesidad, donde se aplicaron escalas de calidad de vida SF 36 y escala BAROS. Estas nos permiten evaluar la pérdida de peso, mejoría de las condiciones médicas, y calidad de vida. Los datos fueron recolectados mediante llamadas telefónicas y revisión de historias clínicas. Se sustrae toda la información en una hoja de Excel 2019 y se analiza en Epi-info 3.5.1. Resultados: La muestra estuvo constituida por 19 pacientes, 14 del sexo femenino (73,7%) y 5 del sexo masculino (26.3%) todos ellos fueron sometidos a una cirugía bariátrica y atendidos en el servicio de Cirugía General del CMDLT en los últimos 10 años. Conclusión: la cirugía bariátrica ayuda al paciente a mantener un buen estado de actividad física, salud emocional y sexual, conclusión que podemos ver gracias a nuestros resultados obtenidos mediante la aplicación de las escalas de calidad de vida SF-36 y BAROS

    The calcium-binding domain of the stress protein SEP53 is required for survival in response to deoxycholic acid-mediated injury

    Stress protein responses have evolved in part as a mechanism to protect cells from the toxic effects of environmental damaging agents. Oesophageal squamous epithelial cells have evolved an atypical stress response that results in the synthesis of a 53 kDa protein of undefined function named squamous epithelial-induced stress protein of 53 kDa (SEP53). Given the role of deoxycholic acid (DCA) as a potential damaging agent in squamous epithelium, we developed assays measuring the effects of DCA on SEP53-mediated responses to damage. To achieve this, we cloned the human SEP53 gene, developed a panel of monoclonal antibodies to the protein, and showed that SEP53 expression is predominantly confined to squamous epithelium. Clonogenic assays were used to show that SEP53 can function as a survival factor in mammalian cell lines, can attenuate DCA-induced apoptotic cell death, and can attenuate DCA-mediated increases in intracellular free calcium. Deletion of the highly conserved EF-hand calcium-binding domain in SEP53 neutralizes the colony survival activity of the protein, neutralizes the protective effects of SEP53 after DCA exposure, and permits calcium elevation in response to DCA challenge. These data indicate that the squamous cell-stress protein SEP53 can function as a modifier of the DCA-mediated calcium influx and identify a novel survival pathway whose study may shed light on mechanisms relating to squamous cell injury and associated cancer development
