    Archaeozoological analysis of the animal remains from the Slavkov gallows

    V doplnění kontextu informací o šibenici ze Slavkova byla provedena archeozoologická analýza, s cílem popsat výskyt živočichů na této lokalitě a rozšířit tak možnosti interpretace archeologické situace. Analyzovaný osteologický materiál byl na základě podrobného zhodnocení tafonomických projevů na kostech rozčleněn do dvou chronologických úseků – první náležející původnímu kontextu provozu šibenice, druhý spíše recentního charakteru. Z prvního období pochází šest druhů domácí i volně žijící fauny, v mladší části souboru bylo identifikováno několik druhů malých či drobných savců, a také pozůstatky ptactva. Lidské zásahy, které byly pozorovány na domestikované fauně ve starším období, svědčí o využití těchto zvířat v rámci potravní strategie lidí na popravišti; stopy natrávení žaludečními šťávami a charakteristické lomy na některých kostech volně žijící fauny v obou etapách odkazují na možné nahromadění vývržků dravým ptactvem.With the aim to broaden the context of the Slavkov gallows archaeological situation, an archaeozoological analysis was carried out in order to describe the occurrence of animal species at this site. Based on the detailed evaluation of the bones taphonomic characteristics, the analysed osteological material was divided into two historical periods. The first one belongs to the gallows original context, the second exhibits more recent character. Six species of domestic and wild fauna originate from the former archaeological era; several species of small mammals, as well as bird remains, have been identified in the second part of the collection. Evidence of human manipulations observed on domesticated fauna from the earlier period indicate the use of these animals as a part of the people’s subsistence strategy at the execution site; traces of digestion by gastric acids and characteristic fractures on wild fauna bones during both eras refer to the possible accumulation of pellets by birds of prey at this site

    Towards cross-linguistic assessment of associative memory

    Associative memory (AM) is conceptualized as the ability to form links between two previously unrelated pieces of information so that the subsequent presentation of one activates the memory of the other. Unlike other types of memory for which standardized assessment tools exist, AM is assessed mostly by ad hoc tasks designed to tackle specific research questions that are not meant to capture AM as a universal cognitive ability. Typically, AM is assessed using paired-associate paradigms with unimodal (e.g., word pairs) or multimodal (e.g., face-word) stimuli sets. In culturally diverse and multilingual societies, the application of these paradigms can lead to an unreliable and biased assessment of memory abilities. To address this issue, we developed an AM paradigm that combines key aspects of AM assessment – associative encoding, associative recognition, and cued recall, as well as implicit AM effect. The stimuli for the task - pictures of common objects and natural scenes - have been selected to minimize language and culture effects. The task has been developed using free software (OpenSesame) and stimuli, in both online and offline mode of administration, thus enabling wide and free use for research purposes across different settings. The large-scale international collaboration is set to adapt the task into 25 languages so far, including Arabic, Bosnian, Czech, Catalan, Chinese, Croatian, Danish, Dutch, English, Estonian, French, German, Greek, Hebrew, Hungarian, Italian, Lithuanian, Malay, Polish, Portuguese, Romanian, Serbian, Slovak, Slovene, and Spanish. Data is collected across 26 countries with a total of 34 samples (150-300 participants each) to assess the psychometric properties of the task and crosslinguistic (in)variance of the memory performance. The collaboration is expected to result in a comprehensive multilingual AM assessment tool, that is freely available for research use

    Differential gene expression in cisplatin-resistant and -sensitive testicular germ cell tumor cell lines

    Testicular germ cell tumors (TGCTs) represent a well curable malignity due to their exceptional response to cisplatin (CDDP). Despite remarkable treatment results, approximately 5% of TGCT patients develop CDDP resistance and die. Exceptional curability makes TGCTs a highly valuable model system for studying the molecular mechanisms of CDDP sensitivity. Our study was aimed at revealing difference in gene expression between the CDDP-resistant and -sensitive TGCT cell lines, and hence at identifying candidate genes that could serve as potential biomarkers of CDDP response. Using gene expression array, we identified 281 genes that are differentially expressed in CDDP-resistant compared to -sensitive TGCT cell lines. The expression of 25 genes with the highest fold change was validated by RT-qPCR. Of them, DNMT3L, GAL, IGFBP2, IGFBP7, L1TD1, NANOG, NTF3, POU5F1, SOX2, WNT6, ZFP42, ID2, PCP4, SLC40A1 and TRIB3, displayed comparable expression change in gene expression array and RT-qPCR, when all CDDP-resistant TGCT cell lines were pairwise combined with all -sensitive ones. Products of the identified genes are pluripotency factors, or are involved in processes, such as cell metabolism, proliferation or migration. We propose that, after clinical validation, these genes could serve as prognostic biomarkers for early detection of CDDP response in TGCT patients

    Soothing the emotional brain : modulation of neural activity to personal emotional stimulation by social touch

    Social touch may modulate emotions, but the neurobehavioral correlates are poorly understood. Here, we investigated neural responses to a picture of a deceased close person and if neural activity and connectivity are modulated by social touch from one's romantic partner. Using functional magnetic resonance imaging (fMRI) we found altered reactivity in several brain areas including the anterior cingulate cortex (ACC) and the anterior insula in response to the personal picture compared to a picture of an unfamiliar person. Hand holding with the romantic partner, compared to being alone, reduced reactivity in the ACC and cerebellum and provided subjective comfort. To separate physical touch from the emotional effect of partner presence, we evaluated hand holding with the partner relative to a stranger, and found reduced reactivity in the anterior insula. Connectivity between the anterior insula and the ACC was reduced during partner touch, and the connectivity strength were negatively related to attachment security, with higher reported partner security associated with weaker connectivity. Overall, holding hands with one's partner attenuates reactivity in emotional brain areas and reduces between-region connectivity

    Integrated Microarray-Based Data Analysis of miRNA Expression Profiles: Identification of Novel Biomarkers of Cisplatin-Resistance in Testicular Germ Cell Tumours

    Testicular germ cell tumours (TGCTs) are the most common solid malignancy among young men, and their incidence is still increasing. Despite good curability with cisplatin (CDDP)-based chemotherapy, about 10% of TGCTs are non-responsive and show a chemoresistant phenotype. To further increase TGCT curability, better prediction of risk of relapse and early detection of refractory cases is needed. Therefore, to diagnose this malignancy more precisely, stratify patients more accurately and improve decision-making on treatment modality, new biomarkers are still required. Numerous studies showed association of differential expressions of microRNAs (miRNAs) with cancer. Using microarray analysis followed by RT-qPCR validation, we identified specific miRNA expression patterns that discriminate chemoresistant phenotypes in TGCTs. Comparing CDDP-resistant vs. -sensitive TGCT cell lines, we identified miR-218-5p, miR-31-5p, miR-125b-5p, miR-27b-3p, miR-199a-5p, miR-214-3p, let-7a and miR-517a-3p as significantly up-regulated and miR-374b-5p, miR-378a-3p, miR-20b-5p and miR-30e-3p as significantly down-regulated. In patient tumour samples, we observed the highest median values of relative expression of miR-218-5p, miR-31-5p, miR-375-5p and miR-517a-3p, but also miR-20b-5p and miR-378a-3p, in metastatic tumour samples when compared with primary tumour or control samples. In TGCT patient plasma samples, we detected increased expression of miR-218-5p, miR-31-5p, miR-517a-3p and miR-375-5p when compared to healthy individuals. We propose that miR-218-5p, miR-31-5p, miR-375-5p, miR-517-3p, miR-20b-5p and miR-378a-3p represent a new panel of biomarkers for better prediction of chemoresistance and more aggressive phenotypes potentially underlying metastatic spread in non-seminomatous TGCTs.
