    Role of G{alpha}12 and G{alpha}13 as Novel Switches for the Activity of Nrf2, a Key Antioxidative Transcription Factor

    G{alpha}12 and G{alpha}13 function as molecular regulators responding to extracellular stimuli. NF-E2-related factor 2 (Nrf2) is involved in a protective adaptive response to oxidative stress. This study investigated the regulation of Nrf2 by G{alpha}12 and G{alpha}13. A deficiency of G{alpha}12, but not of G{alpha}13, enhanced Nrf2 activity and target gene transactivation in embryo fibroblasts. In mice, G{alpha}12 knockout activated Nrf2 and thereby facilitated heme catabolism to bilirubin and its glucuronosyl conjugations. An oligonucleotide microarray demonstrated the transactivation of Nrf2 target genes by G{alpha}12 gene knockout. G{alpha}12 deficiency reduced Jun N-terminal protein kinase (JNK)-dependent Nrf2 ubiquitination required for proteasomal degradation, and so did G{alpha}13 deficiency. The absence of G{alpha}12, but not of G{alpha}13, increased protein kinase C {delta} (PKC {delta}) activation and the PKC {delta}-mediated serine phosphorylation of Nrf2. G{alpha}13 gene knockout or knockdown abrogated the Nrf2 phosphorylation induced by G{alpha}12 deficiency, suggesting that relief from G{alpha}12 repression leads to the G{alpha}13-mediated activation of Nrf2. Constitutive activation of G{alpha}13 promoted Nrf2 activity and target gene induction via Rho-mediated PKC {delta} activation, corroborating positive regulation by G{alpha}13. In summary, G{alpha}12 and G{alpha}13 transmit a JNK-dependent signal for Nrf2 ubiquitination, whereas G{alpha}13 regulates Rho-PKC {delta}-mediated Nrf2 phosphorylation, which is negatively balanced by G{alpha}12

    We have designed a method of harvesting electrical energy using plasmon-enhanced light pressure. A device was fabricated as a cut cone structure that optimizes light collection so that the weak incident light pressure can be sufficiently enhanced inside the cut cone to generate electrical energy. An increase in the device's current output is a strong indication that the pressure of incident light has been enhanced by the surface plasmons on a platinum layer inside the cut cone.     Therapeutic genome editing for Charcot-marie-tooth disease type 1a

    Charcot-Marie-Tooth 1A (CMT1A) is the most common inherited neuropathy without a known therapy, which is caused by a 1.4 Mb duplication on human chromosome 17, which includes the gene encoding the peripheral myelin protein of 22 kDa (PMP22). Overexpressed PMP22 protein from its gene duplication is thought to cause demyelination and subsequently axonal degeneration in the peripheral nervous system (PNS). Here, we targeted regulatory region of human PMP22 to normalize overexpressed PMP22 level in C22 mice, a mouse model of CMT1A harboring multi copies of human PMP22. Direct local intraneural delivery of CRISPR/Cas9 designed to target TATA-box of PMP22 before the onset of disease, downregulates gene expression of PMP22 and preserves both myelin and axons. Notably, the same approach was effective in partial rescue of demyelination even after the onset of disease. Collectively, our data present a potential therapeutic efficacy of CRISPR/Cas9-mediated targeting of regulatory region of PMP22 to treat CMT1A.     Analysis of Exercise Intensity Based on Cardiorespiratory Response in a Virtual Reality-Based Exercise Program for Firefighter Physical Fitness Enhancement

    OBJECTIVES Current regulations for Korean firefighters primarily emphasize exercise during working hours without specifying particular exercise methods. This study aims to evaluate the exercise intensity of the stepwise Korean Firefighter Fitness Program (KFFP) using Kinect-based mixed reality device (Virtual mate, VM), considering the demanding 24-hour shift work and high physical strength requirements. The objective is to assess its potential as one of the exercise methods and provide valuable insights. METHODS CResearchers recruited 30 participants to evaluate the exercise intensity based on the cardiopulmonary responses induced by the Stepwise Korean Firefighter Fitness Program (KFFP) using Virtual Mate (VM). Initially, the participants' resting heart rate, maximum heart rate (HRmax), and maximum oxygen consumption (VO2max) were measured. Subsequently, researchers monitored the participants' cardiopulmonary responses by employing portable gas analyzers and wireless heart rate monitors while implementing the Stepwise KFFP with VM. Exercise intensity related to cardiopulmonary responses was recorded using both absolute parameters (VO2, HR) and relative parameters (%VO2max, %HRR, %HR). Researchers utilized repeated measures analysis of variance for statistical analysis. RESULTS The analysis of the stepwise KFFP using VM showed significant increases all parameters as the program advanced(p<.001). VO2 increased from around 19.4 ml/kg/min in Step 1 to approximately 22.8 ml/kg/min in Step 3. Similarly, %HRR rose from roughly 40.6% in Step 1 to about 53.9% in Step 3. CONCLUSIONS Upon evaluating the stepwise KFFP based on the exercise intensity levels recommended by ACSM, it is confirmed that the stepwise KFFP using VM comprises both moderate-intensity and highintensity exercises. This approach is deemed effective and can serve as a fitness management method for firefighters working in 24-hour shift patterns that demand high fitness levels

    Evaluation of barley to replace milk by-product in weaning pig’s diet

    The supplementation level of barley was limited because of high contents of fiber in monogastric animals. Barley contained high soluble fiber, thus it could prevent to diarrhea of weaning pigs. Moreover, as the barley break down by enzymes, free sugars come out from the barley, which could be used as an energy source in weaning pigs and replace milk by-products in weaning pig’s diet. Therefore, present study was conducted to investigate the influence of barley to replace milk by-product in weaning pig’s diet on growth performance, blood profile, nutrient digestibility, diarrhea incidence, and economic analysis in weaning pigs. A total of 112 crossbred ([York-shire × Landrace] × Duroc, weaned at 28 days of age) piglets were allotted to 4 treatments in a randomized complete block (RCB) design. Each treatment has 7 replications with 4 pigs per pen. Pigs were fed each treatment diet which containing different levels of barley (0%, 10%, 20%, and 30%) at the expense of whey powder and lactose. Three phase feeding programs were used for 6 weeks of growth trial (phase 1: 0–2 weeks; phase 2: 3–4 weeks; phase 3: 5–6 weeks). During 0–2 week, body weight (BW), average daily gain (ADG) and G:F ratio were decreased as barley level increased in the diet (linear response, p < 0.01). In blood profile, blood urea nitrogen was decreased as the barley level increased in the diet (linear, p < 0.01). However, no significant differences were observed in blood glucose level. In nutrient digestibility, crude fat digestibility was linearly increased as barley increased (linear, p < 0.01). The incidence of diarrhea was improved as increasing barley contents in all phases (linear, p < 0.01). These results demonstrated that supplementation of barley to replace milk by-product influenced negatively on growth performance during 0–2 week. However, the incidence of diarrhea and later growth performance from 3 week postweaning were improved as dietary barley level increased

    NaCl plus chitosan as a dietary salt to prevent the development of hypertension in spontaneously hypertensive rats

    The effect of NaCl plus 3% chitosan on the systolic blood pressure of spontaneously hypertensive rats (SHR) were evaluated and compared with NaCl plus KCl (NaCl, 49.36% + KCl 49.36%) and chitosan or NaCl treatment alone. In SHR, administration of NaCl plus chitosan (44 mM Na/day) for two months significantly decreased the systolic blood pressure greater than of NaCl plus KCl and NaCl alone. NaCl plus chitosan resulted, though not statistically significant, in decreased urinary Na+ excretion and decreased blood urea nitrogen levels. Urinary creatinine of NaCl plus chitosan was slightly decreased compared to 3 treated groups. Serum electrolytes levels, however, remained unchanged. The combination of NaCl and chitosan may be superior to the conventional use of NaCl plus KCl or NaCl alone in the prevention of hypertension. Even though these supplementary diets have demonstrated potential anti-hypertensive effects in the experimental animal model, further research is needed before any recommendations can be made

Intelligent and smart systems require freedom of form factor, unrestricted design, and unlimited scale. Initial attempts to develop conductive fibers and textile electronics failed to achieve reliable integration and performance required for industrial-scale manufacturing of technical textiles by standard weaving technologies. Here, we present a textile electronic system with functional one-dimensional devices, including fiber photodetectors (as an input device), fiber supercapacitors (as an energy storage device), fiber field-effect transistors (as an electronic driving device), and fiber quantum dot light-emitting diodes (as an output device). As a proof of concept applicable to smart homes, a textile electronic system composed of multiple functional fiber components is demonstrated, enabling luminance modulation and letter indication depending on sunlight intensity.publishersversionpublishe

    Impact of COVID-19 on cardiovascular testing in the United States versus the rest of the world

    Objectives: This study sought to quantify and compare the decline in volumes of cardiovascular procedures between the United States and non-US institutions during the early phase of the coronavirus disease-2019 (COVID-19) pandemic. Background: The COVID-19 pandemic has disrupted the care of many non-COVID-19 illnesses. Reductions in diagnostic cardiovascular testing around the world have led to concerns over the implications of reduced testing for cardiovascular disease (CVD) morbidity and mortality. Methods: Data were submitted to the INCAPS-COVID (International Atomic Energy Agency Non-Invasive Cardiology Protocols Study of COVID-19), a multinational registry comprising 909 institutions in 108 countries (including 155 facilities in 40 U.S. states), assessing the impact of the COVID-19 pandemic on volumes of diagnostic cardiovascular procedures. Data were obtained for April 2020 and compared with volumes of baseline procedures from March 2019. We compared laboratory characteristics, practices, and procedure volumes between U.S. and non-U.S. facilities and between U.S. geographic regions and identified factors associated with volume reduction in the United States. Results: Reductions in the volumes of procedures in the United States were similar to those in non-U.S. facilities (68% vs. 63%, respectively; p = 0.237), although U.S. facilities reported greater reductions in invasive coronary angiography (69% vs. 53%, respectively; p < 0.001). Significantly more U.S. facilities reported increased use of telehealth and patient screening measures than non-U.S. facilities, such as temperature checks, symptom screenings, and COVID-19 testing. Reductions in volumes of procedures differed between U.S. regions, with larger declines observed in the Northeast (76%) and Midwest (74%) than in the South (62%) and West (44%). Prevalence of COVID-19, staff redeployments, outpatient centers, and urban centers were associated with greater reductions in volume in U.S. facilities in a multivariable analysis. Conclusions: We observed marked reductions in U.S. cardiovascular testing in the early phase of the pandemic and significant variability between U.S. regions. The association between reductions of volumes and COVID-19 prevalence in the United States highlighted the need for proactive efforts to maintain access to cardiovascular testing in areas most affected by outbreaks of COVID-19 infection

    Determination of Building Model Key Points Using Multidirectional Shaded Relief Images Generated from Airborne LiDAR Data

    Light detection and ranging (LiDAR) data collected from airborne laser scanner system is one of the major sources to reconstruct Earth’s surface features. This paper presents a method for detecting model key points (MKPs) of the buildings using LiDAR point clouds. The proposed approach utilizes shaded relief images (SRIs) derived from the LiDAR data. The SRIs based on the concept of the shape from shading could provide unique information about individual surface patches of the building roofs. The main advantage of the proposed approach is to detect directly MKPs, which are primitives for 3D building modeling, without segmenting point clouds. Depending on the location of the light source, the SRIs are created differently. Therefore, integration of the multidirectional SRIs created from different locations of the light source could provide more reliable results. In addition, the vertical exaggeration (i.e., scaling Z-coordinates) is also beneficial because constituent surface patches of the roofs in the SRIs created with vertically exaggerated LiDAR data are more distinguishable. To determine the MKPs of the roofs, building data was separated from other objects using modified marker-controlled watershed algorithm in accordance with criteria to specify buildings such as area, height, and standard deviation. This process could remove the unnecessary objects such as trees, vegetation, and cars. The curvature scale space (CSS) corner detector was used to determine MKP since this method is robust to geometric changes such as rotation, translation, and scale. The proposed method was applied to simulated and real LiDAR datasets with various roof types. The experimental results show that the proposed method is effective in determining MKPs of various roof types with high level of detail (LoD)

    Land Cover Classification Using SegNet with Slope, Aspect, and Multidirectional Shaded Relief Images Derived from Digital Surface Model

    Most object detection, recognition, and classification are performed using optical imagery. Images are unable to fully represent the real-world due to the limited range of the visible light spectrum reflected light from the surfaces of the objects. In this regard, physical and geometrical information from other data sources would compensate for the limitation of the optical imagery and bring a synergistic effect for training deep learning (DL) models. In this paper, we propose to classify terrain features using convolutional neural network (CNN) based SegNet model by utilizing 3D geospatial data including infrared (IR) orthoimages, digital surface model (DSM), and derived information. The slope, aspect, and shaded relief images (SRIs) were derived from the DSM and were used as training data for the DL model. The experiments were carried out using the Vaihingen and Potsdam dataset provided by the German Society for Photogrammetry, Remote Sensing and Geoinformation (DGPF) through the International Society for Photogrammetry and Remote Sensing (ISPRS). The dataset includes IR orthoimages, DSM, airborne LiDAR data, and label data. The motivation of utilizing 3D data and derived information for training the DL model is that real-world objects are 3D features. The experimental results demonstrate that the proposed approach of utilizing and integrating various informative feature data could improve the performance of the DL for semantic segmentation. In particular, the accuracy of building classification is higher compared with other natural objects because derived information could provide geometric characteristics. Intersection-of-union (IoU) of the buildings for the test data and the new unseen data with combining all derived data were 84.90% and 52.45%, respectively
