Determination of molecular groups involved in the interaction of annexin A5 with lipid membrane models at the air–water interface

AbstractAnnexin A5 (AnxA5) is a member of a family of homologous proteins sharing the ability to bind to negatively charged phospholipid membranes in a Ca2+-dependent manner. In this paper, we used polarization-modulated infrared reflection absorption spectroscopy (PMIRRAS), Brewster angle microscopy (BAM), and ellipsometry to investigate changes both in the structure of AnxA5 and phospholipid head groups associated with membrane binding. We found that the secondary structure of AnxA5 in the AnxA5/Ca2+/lipid ternary complex is conserved, mainly in α-helices and the average orientation of the α-helices of the protein is slightly tilted with respect to the normal to the phospholipid monolayer. Upon interaction between AnxA5 and phospholipids, a shift of the νas PO2− band is observed by PMIRRAS. This reveals that the phosphate group is the main group involved in the binding of AnxA5 to phospholipids via Ca2+ ions, even when some carboxylate groups are accessible (PS). PMIRRAS spectra also indicate a change of carboxylate orientation in the aspartate and glutamate residues implicated in the association of the AnxA5, which could be linked to the 2D crystallization of protein under the phospholipid monolayer. Finally, we demonstrated that the interaction of AnxA5 with pure carboxylate groups of an oleic acid monolayer is possible, but the orientation of the protein under the lipid is completely different

Building Decision Tools for Sustainable Grassland Management: A Case Study of Participatory Research in La Réunion

    Get PDF
During the last 30 years, cattle breeding has developed in La Reunion Island (France, Indian Ocean) with strong support from local authorities and extension structures. The Union des Associations Foncières Pastorales (UAFP) initiated and still sustain a large effort on grassland improvement. During the same period, CIRAD managed different research programmes on agronomic aspects of pasture management). Scarcity of space, the volcanic soil and tropical climatic conditions contributed to repeated forage shortages during the dry season, making forage production a major issue for cattle breeders. In 1991, CIRAD launched a research programme to elaborate decision tools for assessing and improving pasture management. A partnership was developed with UAFP, with the local institution being involved in the elaboration of the tools and now being completely in charge of their use to advise cattle breeders on pasture management

Effects of 900 MHz radiofrequency on corticosterone, emotional memory and neuroinflammation in middle-aged rats

International audienceThe widespread use of mobile phones raises the question of the effects of electromagnetic fields (EMF, 900 MHz) on the brain. Previous studies reported increased levels of the glial fibrillary acidic protein (GFAP) in the rat's brain after a single exposure to 900 MHz global system for mobile (GSM) signal, suggesting a potential inflammatory process. While this result was obtained in adult rats, no data is currently available in older animals. Since the transition from middle-age to senescence is highly dependent on environment and lifestyle, we studied the reactivity of middle-aged brains to EMF exposure. We assessed the effects of a single 15 min GSM exposure (900 MHz ; specific absorption rate (SAR)=6 W/kg) on GFAP expression in young adults (6 week-old) and middle-aged rats (12 month-old). Brain interleukin (IL)-1ß and IL-6, plasmatic levels of corticosterone (CORT), and emotional memory were also assessed. Our data indicated that, in contrast to previously published work, acute GSM exposure did not induce astrocyte activation. Our results showed an IL-1ß increase in the olfactory bulb and enhanced contextual emotional memory in GSM-exposed middle-aged rats, and increased plasmatic levels of CORT in GSMexposed young adults. Altogether, our data showed an age dependency of reactivity to GSM exposure in neuro-immunity, stress and behavioral parameters. Reproducing these effects and studying their mechanisms may allow a better understanding of mobile phone EMF effects on neurobiological parameters

    Salvador Dali and jewellery design

The history of design is an important aspect for considering the heritage of individual members of this activity. The term “art” is partly formulated from the assumption, that theexistence of this phenomenon is seen in the temporary space.Rethinking the aesthetic perception of the world, as an individual artist and as the creative discoveries and the era of the arts in general is made by literary translation. One of the brightest occasions in the history of design in 20 century was the appearance of the Spanish artist, painter, graphic artist, sculptor, director, writer and designer, Salvador Dali. The cultural situation of the 20th century as well as considering surrealismto be a special phenomenon in the development of Europeanculture cause the interest to historians and theorists of culture

Inter- and Intragranular Stress Determination with Kossel Microdiffraction in a Scanning Electron Microscope

    Get PDF
A Kossel microdiffraction experimental set up is under development inside a Scanning Electron Microscope (SEM) in order to determine the crystallographic orientation as well as the inter- and intragranular strains and stresses on the micron scale, using a one cubic micrometer spot. The experimental Kossel line patterns are obtained by way of a CCD camera and are then fully indexed using a home-made simulation program. The so-determined orientation is compared with Electron Back-Scattered Diffraction (EBSD) results, and in-situ tests are performed inside the SEM using a tensile/compressive machine. The aim is to verify a 50MPa stress sensitivity for this technique and to take advantage from this microscope environment to associate microstructure observations (slip lines, particle decohesion, crack initiation) with determined stress analyses

The tRNA-like domains of E coli and A.aeolicus transfer-messenger RNA: structural and functional studies.

International audienceTransfer-messenger RNA (tmRNA, 10Sa RNA or ssrA) acts to rescue stalled bacterial ribosomes while encoding a peptide tag added trans-translationally to the nascent peptide, targeting it for proteolysis. The understanding at molecular level of this ubiquitous quality control system in eubacteria requires structural information. Here, we describe the purification and structural analysis of a functional fragment of both Aquifex aeolicus and Escherichia coli tmRNA, recapitulating their tRNA-like domain, which were expressed in vivo from synthetic genes. Both recombinant RNA are correctly processed at both 5' and 3' ends and are produced in quantities suitable for structural analysis by NMR and/or X-ray crystallography. The sequence and solution structure of the tRNA-like domains were analysed by various methods including structural mapping with chemical and enzymatic probes and 2D NMR spectroscopy. The minimalist RNAs contain two post-transcriptional base modifications, 5-methyluridine and pseudouridine, as the full-length tmRNA. Both RNAs fold into three stems, a D-analogue, a T-loop and a GAAA tetra-loop. 2D NMR analysis of the imino proton resonances of both RNAs allowed the assignment of the three stems and of a number of tertiary interactions. It shows the existence of interactions between the TPsiC-loop and the D-analogue, exhibiting a number of similarities and also differences with the canonical tRNA fold, indicating that RNA tertiary interactions can be modulated according to the sequence and secondary structure contexts. Furthermore, the E.coli minimalist RNA is aminoacylatable with alanine with a catalytic efficiency an order of magnitude higher than that for full-length tmRNA
