    Degradabilidade in situ de híbridos de milho e de capim-elefante colhidos em quatro estádios de maturidade

    Dentre as forragens, a silagem de milho é amplamente utilizada pelos fazendeiros que visam explorar o máximo do potencial genético dos animais. No entanto, outros volumosos tropicais como o capim-elefante (Pennisetum purpureum) são mais produtivos e, portanto, mais baratos do que a silagem de milho. Nosso objetivo foi comparar a degradabilidade in situ do capim-elefante com a degradabilidade de híbridos de milho, colhidos em quatro estágios de maturidade. O experimento seguiu um delineamento de blocos ao acaso com sub-parcelas. Dois híbridos de milho: AG5011 e ZN8392 foram colhidos com 25, 30, 35 e 40% matéria seca (MS) na planta toda e separados na fração colmo + bainha + folhas e espigas. Capim elefante foi colhido 30, 40, 50 e 60 dias após o corte de nivelamento. As amostras secas e trituradas foram incubadas no rúmen por 0, 6, 12, 24, 48 e 72 h para cálculo da cinética da degradação ruminal da MS e da fibra em detergente neutro (FDN). O avanço da maturidade aumentou os teores de FDN e fibra em detergente ácido (FDA) do capim elefante e reduziu a degradabilidade da MS. Entretanto, a maturidade teve pouco efeito sobre os teores de fibra e a degradabilidade da MS da fração planta dos híbridos de milho. O capim elefante apresentou maior degradabilidade da FDN do que híbridos de milho, e não houve efeito da maturidade sobre a degradabilidade da FDN das duas espécies. Fiber degradability of elephant-grass was not worse than that of corn stover, and therefore the choice of forage should be made on economical analysis rather than assuming an intrinsic low production potential for elephant-grass based diet

    Formation and Evolution of Supermassive Black Holes

    The correlation between the mass of supermassive black holes in galaxy nuclei and the mass of the galaxy spheroids or bulges (or more precisely their central velocity dispersion), suggests a common formation scenario for galaxies and their central black holes. The growth of bulges and black holes can commonly proceed through external gas accretion or hierarchical mergers, and are both related to starbursts. Internal dynamical processes control and regulate the rate of mass accretion. Self-regulation and feedback are the key of the correlation. It is possible that the growth of one component, either BH or bulge, takes over, breaking the correlation, as in Narrow Line Seyfert 1 objects. The formation of supermassive black holes can begin early in the universe, from the collapse of Population III, and then through gas accretion. The active black holes can then play a significant role in the re-ionization of the universe. The nuclear activity is now frequently invoked as a feedback to star formation in galaxies, and even more spectacularly in cooling flows. The growth of SMBH is certainly there self-regulated. SMBHs perturb their local environment, and the mergers of binary SMBHs help to heat and destroy central stellar cusps. The interpretation of the X-ray background yields important constraints on the history of AGN activity and obscuration, and the census of AGN at low and at high redshifts reveals the downsizing effect, already observed for star formation. History appears quite different for bright QSO and low-luminosity AGN: the first grow rapidly at high z, and their number density decreases then sharply, while the density of low-luminosity objects peaks more recently, and then decreases smoothly.Comment: 31 pages, 13 figures, review paper for Astrophysics Update

    Single-cell RNA sequencing of liver fine-needle aspirates captures immune diversity in the blood and liver in chronic hepatitis B patients

    Background and Aims: HBV infection is restricted to the liver, where it drives exhaustion of virus-specific T and B cells and pathogenesis through dysregulation of intrahepatic immunity. Our understanding of liver-specific events related to viral control and liver damage has relied almost solely on animal models, and we lack useable peripheral biomarkers to quantify intrahepatic immune activation beyond cytokine measurement. Our objective was to overcome the practical obstacles of liver sampling using fine-needle aspiration and develop an optimized workflow to comprehensively compare the blood and liver compartments within patients with chronic hepatitis B using single-cell RNA sequencing. Approach and Results: We developed a workflow that enabled multi-site international studies and centralized single-cell RNA sequencing. Blood and liver fine-needle aspirations were collected, and cellular and molecular captures were compared between the Seq-Well S3 picowell-based and the 10× Chromium reverse-emulsion droplet–based single-cell RNA sequencing technologies. Both technologies captured the cellular diversity of the liver, but Seq-Well S3 effectively captured neutrophils, which were absent in the 10× dataset. CD8 T cells and neutrophils displayed distinct transcriptional profiles between blood and liver. In addition, liver fine-needle aspirations captured a heterogeneous liver macrophage population. Comparison between untreated patients with chronic hepatitis B and patients treated with nucleoside analogs showed that myeloid cells were highly sensitive to environmental changes while lymphocytes displayed minimal differences. Conclusions: The ability to electively sample and intensively profile the immune landscape of the liver, and generate high-resolution data, will enable multi-site clinical studies to identify biomarkers for intrahepatic immune activity in HBV and beyond.</p

    Effect of lactation stage and concurrent pregnancy on milk composition in the bottlenose dolphin

    Although many toothed whales (Cetacea: Odontoceti) lactate for 2–3 years or more, it is not known whether milk composition is affected by lactation stage in any odontocete species. We collected 64 pooled milk samples spanning 1–30 months postpartum from three captive bottlenose dolphins Tursiops truncatus. Milks were assayed for water, fat, crude protein (TN × 6.38) and sugar; gross energy was calculated. Ovulation and pregnancy were determined via monitoring of milk progesterone. Based on analysis of changes in milk composition for each individual dolphin, there were significant increases (P<0.05) in fat (in all three dolphins) and crude protein (in two of three), and a decrease (P<0.05) in water (in two of three) over the course of lactation, but the sugar content did not change. In all three animals, the energy content was positively correlated with month of lactation, but the percentage of energy provided by crude protein declined slightly but significantly (P<0.05). At mid-lactation (7–12 months postpartum, n=17), milk averaged 73.0±1.0% water, 12.8±1.0% fat, 8.9±0.5% crude protein, 1.0±0.1% sugar, 1.76±0.09 kcal g−1 (=7.25 kJ g−1) and 30.3±1.3% protein:energy per cent. This protein:energy per cent was surprisingly high compared with other cetaceans and in relation to the growth rates of calves. Milk progesterone indicated that dolphins ovulated and conceived between 413 and 673 days postpartum, following an increase in milk energy density. The significance of these observed compositional changes to calf nutrition will depend on the amounts of milk produced at different stages of lactation, and how milk composition and yield are influenced by sampling procedure, maternal diet and maternal condition, none of which are known

    Topological R4R^4 Inflation

    We consider the possibility that higher-curvature corrections could drive inflation after the compactification to four dimensions. Assuming that the low-energy limit of the fundamental theory is eleven-dimensional supergravity to the lowest order, including curvature corrections and taking the descent from eleven dimensions to four via an intermediate five-dimensional theory, as favored by recent considerations of unification at some scale around 1016\sim 10^{16} GeV, we may obtain a simple model of inflation in four dimensions. The effective degrees of freedom are two scalar fields and the metric. The scalars arise as the large five-dimensional modulus and the self-interacting conformal mode of the metric. The effective potential has a local maximum in addition to the more usual minimum. However, the potential is quite flat at the top, and admits topological inflation. We show that the model can resolve cosmological problems and provide a mechanism for structure formation with very little fine tuning.Comment: 25 pages, latex, 2 eps figures, minor changes, accepted for publication in Phys.     Perfilhamento, área foliar e produtividade do milho sob diferentes arranjos espaciais

    O objetivo deste trabalho foi avaliar o efeito de variações no arranjo espacial de plantas sobre o perfilhamento, a área foliar e a produtividade do milho. Os experimentos foram implantados na primavera/ verão dos anos agrícolas 2007/2008 e 2008/2009. Os tratamentos consistiram de quatro densidades (três, cinco, sete e nove plantas por metro quadrado) e de três espaçamentos entre linhas (0,4, 0,7 e 1,0 m). Foram avaliados o índice de área foliar (IAF) e a produtividade de grãos do híbrido P30F53, além da contribuição dos perfílhos para esses caracteres. Em 2007/2008, não houve deficiência hídrica, o IAF na floração foi superior a 7 e os perfilhos contribuíram com 65% do IAF total, na menor densidade de plantas. Nesse ano, a produtividade de grãos (13,7 Mg ha-1) não foi afetada pelos tratamentos, e os perfilhos contribuíram com 44% da produtividade, na densidade de três plantas por metro quadrado. Em 2008/2009, houve restrição hídrica na pré‑floração e no enchimento de grãos, o que diminuiu o perfilhamento e a contribuição dos perfilhos ao IAF. A produtividade de grãos, nesse ano, aumentou de 9,7 para 11,7 Mg ha-1 com o aumento na densidade de plantas, mas a contribuição dos perfilhos à produtividade foi menor. O perfilhamento aumenta a estabilidade fenotípica da produtividade de grãos frente a variações no arranjo de plantas

    Techniques for measuring aerosol attenuation using the Central Laser Facility at the Pierre Auger Observatory

    The Pierre Auger Observatory in Malargüe, Argentina, is designed to study the properties of ultra-high energy cosmic rays with energies above 10(18) eV. It is a hybrid facility that employs a Fluorescence Detector to perform nearly calorimetric measurements of Extensive Air Shower energies. To obtain reliable calorimetric information from the FD, the atmospheric conditions at the observatory need to be continuously monitored during data acquisition. In particular, light attenuation due to aerosols is an important atmospheric correction. The aerosol concentration is highly variable, so that the aerosol attenuation needs to be evaluated hourly. We use light from the Central Laser Facility, located near the center of the observatory site, having an optical signature comparable to that of the highest energy showers detected by the FD. This paper presents two procedures developed to retrieve the aerosol attenuation of fluorescence light from CLF laser shots. Cross checks between the two methods demonstrate that results from both analyses are compatible, and that the uncertainties are well understood. The measurements of the aerosol attenuation provided by the two procedures are currently used at the Pierre Auger Observatory to reconstruct air shower data
