    Age-related response to playback of calls of sharp-shinned hawks (Accipiter striatus) by American redstart (Setophaga ruticilla) males.

    BirdsAbstract We investigated whether the response of American Redstart (Setophaga ruticilla) is influenced by age. We recorded song rates before and after playback of a Sharp-shinned Hawk (Accipiter striatus) call, and how long after playback redstarts took to sing again. Our results showed no significant differences in response between first year and second year males, but significant reduction in song rate for all redstarts was found immediately post-playback.     Repeatability and Reproducibility of Decisions by Latent Fingerprint Examiners

    The interpretation of forensic fingerprint evidence relies on the expertise of latent print examiners. We tested latent print examiners on the extent to which they reached consistent decisions. This study assessed intra-examiner repeatability by retesting 72 examiners on comparisons of latent and exemplar fingerprints, after an interval of approximately seven months; each examiner was reassigned 25 image pairs for comparison, out of total pool of 744 image pairs. We compare these repeatability results with reproducibility (inter-examiner) results derived from our previous study. Examiners repeated 89.1% of their individualization decisions, and 90.1% of their exclusion decisions; most of the changed decisions resulted in inconclusive decisions. Repeatability of comparison decisions (individualization, exclusion, inconclusive) was 90.0% for mated pairs, and 85.9% for nonmated pairs. Repeatability and reproducibility were notably lower for comparisons assessed by the examiners as “difficult” than for “easy” or “moderate” comparisons, indicating that examiners' assessments of difficulty may be useful for quality assurance. No false positive errors were repeated (n = 4); 30% of false negative errors were repeated. One percent of latent value decisions were completely reversed (no value even for exclusion vs. of value for individualization). Most of the inter- and intra-examiner variability concerned whether the examiners considered the information available to be sufficient to reach a conclusion; this variability was concentrated on specific image pairs such that repeatability and reproducibility were very high on some comparisons and very low on others. Much of the variability appears to be due to making categorical decisions in borderline cases

    Pre-validation of a MALDI MS proteomics-based method for the reliable detection of blood and blood provenance

    Abstract: The reliable identification of blood, as well as the determination of its origin (human or animal) is of great importance in a forensic investigation. Whilst presumptive tests are rapid and deployed in situ, their very nature requires confirmatory tests to be performed remotely. However, only serological tests can determine blood provenance. The present study improves on a previously devised Matrix Assisted Laser Desorption Ionisation Mass Spectrometry (MALDI MS)—proteomics based method for the reliable detection of blood by enabling the determination of blood provenance. The overall protocol was developed to be more specific than presumptive tests and faster/easier than the gold standard liquid chromatography (LC) MS/MS analysis. This is considered a pre-validation study that has investigated stains and fingermarks made in blood, other biofluids and substances that can elicit a false-positive response to colorimetric or presumptive tests, in a blind fashion. Stains and marks were either untreated or enhanced with a range of presumptive tests. Human and animal blood were correctly discriminated from other biofluids and non-biofluid related matrices; animal species determination was also possible within the system investigated. The procedure is compatible with the prior application of presumptive tests. The refined strategy resulting from iterative improvements through a trial and error study of 56 samples was applied to a final set of 13 blind samples. This final study yielded 12/13 correct identifications with the 13th sample being correctly identified as animal blood but with no species attribution. This body of work will contribute towards the validation of MALDI MS based methods and deployment in violent crimes involving bloodshed

    Nitrate and a nitrate-reducing Rothia aeria strain as potential prebiotic or synbiotic treatments for periodontitis

    A few studies indicate that nitrate can reduce dysbiosis from a periodontitis point of view. However, these experiments were performed on samples from healthy individuals, and it is unknown if nitrate will be effective in periodontal patients, where the presence of nitrate-reducing bacteria is clearly reduced. The aim of this study was to test the effect of nitrate and a nitrate-reducing R. aeria (Ra9) on subgingival biofilms of patients with periodontitis. For this, subgingival plaque was incubated with 5 mM nitrate for 7 h (n = 20) or 50 mM nitrate for 12 h (n = 10), achieving a ~50% of nitrate reduction in each case. Additionally, Ra9 was combined with 5 mM nitrate (n = 11), increasing the nitrate reduced and nitrite produced (both p &lt; 0.05). The addition of nitrate to periodontitis communities decreased biofilm mass (50 mM &gt; 5 mM, both p &lt; 0.05). Five millimolar nitrate, 50 mM nitrate and 5 mM nitrate + Ra9 led to 3, 28 and 20 significant changes in species abundance, respectively, which were mostly decreases in periodontitis-associated species. These changes led to a respective 15%, 63% (both p &lt; 0.05) and 6% (not significant) decrease in the dysbiosis index. Using a 10-species biofilm model, decreases in periodontitis-associated species in the presence of nitrate were confirmed by qPCR (all p &lt; 0.05). In conclusion, nitrate metabolism can reduce dysbiosis and biofilm growth of periodontitis communities. Five millimolar nitrate (which can be found in saliva after vegetable intake) was sufficient, while increasing this concentration to 50 mM (which could be achieved by topical applications such as a periodontal gel) increased the positive effects. Ra9 increased the nitrate metabolism of periodontitis communities and should be tested in vivo.</p

    Anterior bridging bronchus

    Previously reported patients with a bridging bronchus (BB) presented with respiratory distress. In addition, each patient had one or more associated anomalies. All but two patients progressed to cardiopulmonary failure and death. We describe a case of an anterior BB without associated anomalies, who did well without operative intervention. This patient presented with a cough at 6 months of age. Chest X-ray was normal, but due to suspicion of foreign body aspiration, bronchoscopy was performed, which revealed a third bronchus at the carina. Bronchography demonstrated the anatomy of the BB. The patient has continued to do well without further intervention.     A proteomic approach for the rapid, multi-informative and reliable identification of blood

    Blood evidence is frequently encountered at the scene of violent crimes and can provide valuable intelligence in the forensic investigation of serious offences. Because many of the current enhancement methods used by crime scene investigators are presumptive, the visualisation of blood is not always reliable nor does it bear additional information. In the work presented here, two methods employing a shotgun bottom up proteomic approach for the detection of blood are reported; the developed protocols employ both an in solution digestion method and a recently proposed procedure involving immobilization of trypsin on hydrophobin Vmh2 coated MALDI sample plate. The methods are complementary as whilst one yields more identifiable proteins (as biomolecular signatures), the other is extremely rapid (5 minutes). Additionally, data demonstrate the opportunity to discriminate blood provenance even when two different blood sources are present in a mixture. This approach is also suitable for old bloodstains which had been previously chemically enhanced, as experiments conducted on a 9-year-old bloodstain deposited on a ceramic tile demonstrate

    Latent Print Proficiency Testing: An Examination of Test Respondents, Test‐Taking Procedures, and Test Characteristics

    Proficiency testing is a key component of quality assurance programs within crime laboratories and can help improve laboratory practices. However, current proficiency testing procedures contain significant limitations and can be misinterpreted by examiners and court personnel (Garrett & Mitchell, 2018). To evaluate some of these limitations, we surveyed latent print examiners (n = 198) after they completed a Collaborative Testing Services, Inc. proficiency test. Additionally, we evaluated test performance and used a quality metric algorithm to evaluate the quality of test prints. Results do not suggest that respondents are dissimilar to the broader examiner population, although they may engage in different behaviors when completing tests versus casework. Findings show that proficiency testing contains prints of high quality and is perceived as both relatively easy and representative of casework. The test discriminated between inexperienced and experienced respondents, and verification procedures were largely ineffective in reducing errors. Objective quality metrics may provide a path forward to improving proficiency testing in a measurable manner

    The Middle Cambrian Wheeler Formation: Sequence Stratigraphy and Geochemistry Across a Ramp-to-Basin Transition

    The Middle Cambrian Wheeler Formation is interpreted as having been deposited in the shallow ramp and deeper basin environments of the House Range embayment (HRE), presumably, during a single third-order sequence. In the Drum Mountains, the Wheeler Formation (295 m thick) is dominated by proximal and distal ramp deposits; at Ma~um Pass, the Wheeler Formation (190m thick) is dominated by basinal shale deposits. The Wheeler Formation contains only one biozone marker; the first appearance of Ptyhagnostus atavus. Lack of other chronostratigraphic markers and distinctive stratal patterns in the basinal facies makes correlation along this ramp-to-basin transect difficult. Therefore, carbon-isotope stratigraphy and total organic carbon analysis were tested for their utility as intra basinal correlation tools. 813Ccarbonate isotope values range from -1.7% to 0.07%o (PDB) at Marjum Pass and -1.1% to 1.4% (PDB) in the Drum Mountains; previously reported 813Ccarbonate values in the Great Basin for this time interval range between -2% to 2% (PDB). Both localities show small-scale isotope variability, however, this variability is thought to be the result of local isotopic effects and was not used for correlation. TOC values obtained from both sections increase upsection, define a distinct peak, then decrease upsection. These peaks are associated with shale facies and occur near the maximum flooding surface in both sections, indicating that the TOC results could be used for correlation between sections. The lithologic cyclicity recognized in the shallow-water deposits at the Drum Mountains locality have also been recognized in the deeper-water deposits at Ma~um Pass. At each locality the meter-scale cycles shallow upward and display similar stacking patterns. Because cyclicity is preserved in both sections and the total stratigraphic thickness and cycle thickness decrease toward the embayment-controlling fault, it is probable that the cyclicity was the result of small-scale eustatic changes in sea level rather than episodic tectonism. This ramp-to-basin correlation also supportS the validity of P. atavus as a global biostratigraphic marker. The first appearance of Ptydnagostus atavus has been found below the interpreted maximum flooding surface and was coeval with transgression in both localities, indicating that its appearance was likely synchronous
