    Get PDF

Design of Magnetic Resonance Imaging (MRI) RF Coils by Using the Method of Moments

    Get PDF
Abstract - A Method of Moments (MoM) technique is employed, to design RF coils for Magnetic Resonance Imaging (MRI) applications. In particular a full wave simulator has been usefully applied to determine the principal characteristics of the antennas used for this kind of application, i.e. the resonant modes, the Q factor and the uniformity of the magnetic field radiated by the sensor. Indeed, at the increasing of the operating frequency, magneto-static models are no longer valid and more sophisticated electromagnetic tools are needed. Some examples relevant to the design of different kind of birdcage coils are presented to demonstrate the effectiveness of the method

In vitro transcription profiling of the σS subunit of bacterial RNA polymerase: re-definition of the σS regulon and identification of σS-specific promoter sequence elements

Specific promoter recognition by bacterial RNA polymerase is mediated by σ subunits, which assemble with RNA polymerase core enzyme (E) during transcription initiation. However, σ70 (the housekeeping σ subunit) and σS (an alternative σ subunit mostly active during slow growth) recognize almost identical promoter sequences, thus raising the question of how promoter selectivity is achieved in the bacterial cell. To identify novel sequence determinants for selective promoter recognition, we performed run-off/microarray (ROMA) experiments with RNA polymerase saturated either with σ70 (Eσ70) or with σS (EσS) using the whole Escherichia coli genome as DNA template. We found that Eσ70, in the absence of any additional transcription factor, preferentially transcribes genes associated with fast growth (e.g. ribosomal operons). In contrast, EσS efficiently transcribes genes involved in stress responses, secondary metabolism as well as RNAs from intergenic regions with yet-unknown function. Promoter sequence comparison suggests that, in addition to different conservation of the −35 sequence and of the UP element, selective promoter recognition by either form of RNA polymerase can be affected by the A/T content in the −10/+1 region. Indeed, site-directed mutagenesis experiments confirmed that an A/T bias in the −10/+1 region could improve promoter recognition by Eσ

<p>Abstract</p> <p>Background</p> <p><it>Acinetobacter baumannii </it>is emerging as an important nosocomial pathogen. Multidrug resistance, as well as ability to withstand environmental stresses, makes eradication of <it>A. baumannii </it>difficult, particularly from hospital settings.</p> <p>Results</p> <p>Over a six-year period, 73 isolates of <it>A. baumannii </it>were collected from infected patients in two hospitals in Italy. While 69 out of the 73 isolates displayed identical multidrug antibiotic resistance pattern, they were susceptible to carbapenems. Genetic profiles of these 69 isolates, determined by Pulsed Field Gel Electrophoresis (PFGE), indicated that they were genetically related and could be clustered in a specific clone, called SMAL. We tested the ability of the SMAL clone to form biofilm, an important determinant for bacterial colonization of the human host and for persistence in the hospital environment. Biofilm formation by <it>A. baumannii </it>SMAL, measured as surface adhesion to polystyrene, is strongly affected by growth conditions, being impaired in rich growth media such as LB, while being favoured in glucose-based medium. Surface adhesion in glucose-based media is inhibited by treatment with cellulase, suggesting that it depends on production of cellulose or of a chemically related extracellular polysaccharide. Exposure of <it>A. baumannii </it>SMAL to subinhibitory concentrations of imipenem resulted in biofilm stimulation and increased production of iron uptake proteins. Growth in iron-supplemented medium also stimulated surface adhesion, thus suggesting that increased intracellular iron concentrations might act as an environmental signal for biofilm formation in <it>A. baumannii </it>SMAL.</p> <p>Conclusions</p> <p>Our results indicate that exposure to subinhibitory concentrations of imipenem can stimulate biofilm formation and induce iron uptake in a pathogenic strain of <it>A. baumannii</it>, with potential implications on antibiotic susceptibility and ability to persist in the human host.</p

Comparative genomics of closely related strains of Klebsiella pneumoniae reveals genes possibly involved in colistin resistance

Strains of colistin-resistant Klebsiella pneumoniae are emerging worldwide, due to the increased use of this molecule in antibiotic-resistant nosocomial infections. Comparative genomics was performed on three closely related K. pneumoniae strains isolated from three patients in a single hospital in Bologna, Italy. Two of these isolates are colistin-resistant, while the third is sensitive to this antibiotic. The designed bioinformatic approach detected, among the three analyzed genomes, single nucleotide polymorphisms, insertions and deletions, specific patterns of gene presence and absence, in a total of 270 genes. These genes were analyzed by automatic and manual methods, to identify those potentially involved in colistin resistance, based on the data available in the literature and on the mechanism of action of colistin, the alteration of the outer membrane. Three of the identified genes (waaL, rfbA, vacJ), all presenting non-synonymous substitutions in the colistin resistant strains, resulted to be of special interest, due to the specific function of their protein products, involved in the biosynthesis of the outer bacterial membrane

A Molecular Epidemiology Case Control Study on Pleural Malignant Mesothelioma

Abstract Pleural malignant mesothelioma is an uncommon neoplasm usually associated with asbestos exposure. The increasing incidence of malignant mesothelioma cases involving individuals with low levels of asbestos exposure suggests a complex carcinogenetic process with the involvement of other cofactors. Cytogenetic studies revealed the complexity of the genetic changes involved in this neoplasm reflecting the accumulation of genomic damage. One of the most used methodologies for assessing genomic damage is the cytokinesis-blocked micronucleus test applied in peripheral blood lymphocytes (PBL). This approach allows the detection of chromosomal alterations expressed in binucleated cells after nuclear division in vitro. This marker could provide a tool for assessing genetically determined constitutional differences in chromosomal instability. A biomonitoring study was carried out to evaluate the micronuclei frequency in PBLs of patients with pleural malignant mesothelioma with respect to lung cancer, healthy, and risk controls as a marker of cancer susceptibility in correlation with the presence of SV40. A significant increased micronuclei frequency was observed in patients with malignant mesothelioma in comparison with all the other groups, the mean micronuclei frequency was double in patients with malignant mesothelioma compared with healthy controls, risk controls, and patients with lung adenocarcinoma (median 11.4 binucleated cells with micronuclei/1,000 binucleated cells versus 6.2, 6.1, and 5.1, respectively). Our data indicate that human T lymphocyte samples carry DNA sequences coding for SV40 large T antigen at low prevalence, both in cancer cases and controls. Evidence of cytogenetic damage revealed as micronuclei frequency in mesothelioma cancer patients could be related to exogenous and endogenous cofactors besides asbestos exposure

Stratigraphic framework of the late Miocene to Pliocene Pisco Formation at Cerro Colorado (Ica Desert, Peru)

This paper describes a 200 m-thick section of the Pisco Formation exposed at Cerro Colorado, an important fossiliferous site in the Ica desert. In order to properly place the fauna in its correct relative position, this study establishes the stratigraphic framework within which the different fossil-bearing intervals of this site can be compared and may prove invaluable in future high-resolution studies on the faunal change. Most of the Pisco Formation deposits exposed at Cerro Colorado consist of gently dipping fine-grained sandstones, diatomaceous siltstones and diatomites with minor ash layers and dolomites deposited within nearshore and offshore settings. To facilitate detailed stratigraphic correlations within the Pisco strata for a 30 km2 area, eight marker beds have been defined and large-scale (1:10,000 scale) geological mapping conducted to determine fault positions, styles and offsets. The geological map shows that there are two important angular unconformities in the study area. The first one is the interformational basal unconformity of the Pisco Formation against folded, faulted, and planated Oligo-Miocene rocks of the Chilcatay Formation. The second is a low-angle intraformational erosional discontinuity of up to 48 angular discordance that allows the subdivision of the Pisco stratigraphy exposed in the study area into two informal allomembers. Dating of the exposed succession by diatom biostratigraphy suggests that the age of the lower allomember is late Miocene, whereas the upper allomember is late Miocene or younger
