    The synthesis and purification of aromatic hydrocarbons IV : 1,2,3-trimethylbenzene

    A six gallon quantity of 1,2,3-trimethylbenezene was prepared and purified in a four-step synthesis involving the condensation of 1,3-pentadiene with crotonaldehyde. The dimethylcyclohexencarboxaldehydes formed were hydrogenated to give the corresponding isomeric dimethylcyclohexylcarbinols. The dehydration of the carbinols and the subsequent dehydrogenation of the trimethylcyclohexenes yielded the 1,2,3-trimethylbenzene.     Contaminant biotransport by Pacific salmon to Lake Michigan tributaries

    The Great Lakes are ideal systems for evaluating the synergistic components of environmental change, such as exotic species introductions and legacy pollutants. Introduced Pacific Salmon (Oncorhynchus spp.) represent an intersection of these drivers because they are non-native species of economic importance that bioaccumulate contaminants during the open water phase of their life cycle. Furthermore, Pacific salmon can deliver a significant pulse of contaminated tissue to tributaries during spawning and subsequent death. Thus, salmon represent a key pathway by which contaminants accumulated in Lake Michigan are transported inland to tributaries that otherwise lack point source pollution. Our research has revealed that salmon exhibit basin-specific persistent organic pollutant (POP) and mercury (Hg) concentrations reflecting pollutant inputs from both current and historic sources. Overall, Lake Michigan salmon were more contaminated with POPs and Hg than conspecifics from Lakes Huron or Superior. Consequently, Lake Michigan salmon pose a higher risk and magnitude of contaminant biotransport and transfer. Resident stream fish (e.g., brook trout) sampled from salmon spawning reaches had higher pollutant concentrations than fish sampled from upstream reaches lacking salmon, but the extent of fish contamination varied among lake basins and streams. In general, Lake Michigan tributaries were the most impacted, suggesting a direct relationship between the extent of salmon-derived contaminant inputs and resident fish contaminant levels. Within and among lake basins, contaminant biotransport by salmon is context dependent and likely reflects a suite of ecological characteristics such as species identity and trophic position, dynamics of the salmon run, watershed land-use, and instream geomorphology such as sediment size. We suggest that future management of salmon-mediated contaminant biotransport to stream communities in the Great Lakes basin should consider biological, chemical, and physical factors that constitute the environmental context

    Restoration of an Indiana, USA, stream: bridging the gap between basic and applied lotic ecology

Articles in this series Abstract. Stream restoration attempts to reverse the global degradation of rivers and streams, but rigorous evaluations are needed to advance the science. We evaluated a 3 rd -order channelized Indiana (USA) stream that was restored in 1997 by constructing two meanders, each ϳ400 m long. Pool and riffle sequences were constructed, coarse substrate and wood were added to the channel, banks were stabilized and revegetated, and sedimentation was reduced by creating a sediment retention basin upstream. Habitat, periphyton, macroinvertebrates, and fishes were measured before restoration and for 5 y after restoration in the restored reaches and in an upstream, unrestored reach. Restoration improved habitat conditions (e.g., more pools, fewer fine sediments) in both restored reaches compared to the unrestored reach. Within 1 y after restoration, major trophic groups (i.e., periphyton, macroinvertebrates, and fishes) recovered to or exceeded levels in the degraded, unrestored reach. However, biotic responses varied with time, trophic level, and community parameter measured. Five years after the restoration, habitat quality, algal abundance, and macroinvertebrate density remained higher in the restored reaches, whereas macroinvertebrate diversity and fish abundance in the restored reaches were similar to or below levels in the unrestored, channelized reach. Although biotic recovery was relatively rapid, long-term persistence is uncertain because of continued sedimentation at a watershed scale. In many instances, reach-scale restorations may be ineffective in the face of basin-wide degradation. This study illustrates the importance of conducting long-term assessments of stream restorations, which can improve both knowledge and management of stream ecosystems

    Acidic microenvironment plays a key role in human melanoma progression through a sustained exosome mediated transfer of clinically relevant metastatic molecules

    Background: Microenvironment cues involved in melanoma progression are largely unknown. Melanoma is highly influenced in its aggressive phenotype by the changes it determinates in its microenvironment, such as pH decrease, in turn influencing cancer cell invasiveness, progression and tissue remodelling through an abundant secretion of exosomes, dictating cancer strategy to the whole host. A role of exosomes in driving melanoma progression under microenvironmental acidity was never described. Methods: We studied four differently staged human melanoma lines, reflecting melanoma progression, under microenvironmental acidic pHs pressure ranging between pH 6.0-6.7. To estimate exosome secretion as a function of tumor stage and environmental pH, we applied a technique to generate native fluorescent exosomes characterized by vesicles integrity, size, density, markers expression, and quantifiable by direct FACS analysis. Functional roles of exosomes were tested in migration and invasion tests. Then we performed a comparative proteomic analysis of acid versus control exosomes to elucidate a specific signature involved in melanoma progression. Results: We found that metastatic melanoma secretes a higher exosome amount than primary melanoma, and that acidic pH increases exosome secretion when melanoma is in an intermediate stage, i.e. metastatic non-invasive. We were thus able to show that acidic pH influences the intercellular cross-talk mediated by exosomes. In fact when exposed to exosomes produced in an acidic medium, pH naïve melanoma cells acquire migratory and invasive capacities likely due to transfer of metastatic exosomal proteins, favoring cell motility and angiogenesis. A Prognoscan-based meta-analysis study of proteins enriched in acidic exosomes, identified 11 genes (HRAS, GANAB, CFL2, HSP90B1, HSP90AB1, GSN, HSPA1L, NRAS, HSPA5, TIMP3, HYOU1), significantly correlating with poor prognosis, whose high expression was in part confirmed in bioptic samples of lymph node metastases. Conclusions: A crucial step of melanoma progression does occur at melanoma intermediate -stage, when extracellular acidic pH induces an abundant release and intra-tumoral uptake of exosomes. Such exosomes are endowed with pro-invasive molecules of clinical relevance, which may provide a signature of melanoma advancement

    Modeling the drug release from hydrogel-based matrices

    In this work the behavior of hydrogel-based matrices, the most widespread systems for oral controlled release of pharmaceuticals, has been mathematically described. In addition, the calculations of the model have been validated against a rich set of experimental data obtained working with tablets made of hydroxypropyl methylcellulose (a hydrogel) and theophylline (a model drug). The model takes into account water uptake, hydrogel swelling, drug release, and polymer erosion. The model was obtained as an improvement of a previous code, describing the diffusion in concentrated systems, and obtaining the erosion front (which is a moving boundary) from the polymer mass balance (in this way, the number of fitting parameters was also reduced by one). The proposed model was found able to describe all the observed phenomena, and then it can be considered a tool with predictive capabilities, useful in design and testing of new dosage systems based on hydrogels

    Measurement of inclusive D*+- and associated dijet cross sections in photoproduction at HERA

    Inclusive photoproduction of D*+- mesons has been measured for photon-proton centre-of-mass energies in the range 130 < W < 280 GeV and a photon virtuality Q^2 < 1 GeV^2. The data sample used corresponds to an integrated luminosity of 37 pb^-1. Total and differential cross sections as functions of the D* transverse momentum and pseudorapidity are presented in restricted kinematical regions and the data are compared with next-to-leading order (NLO) perturbative QCD calculations using the "massive charm" and "massless charm" schemes. The measured cross sections are generally above the NLO calculations, in particular in the forward (proton) direction. The large data sample also allows the study of dijet production associated with charm. A significant resolved as well as a direct photon component contribute to the cross section. Leading order QCD Monte Carlo calculations indicate that the resolved contribution arises from a significant charm component in the photon. A massive charm NLO parton level calculation yields lower cross sections compared to the measured results in a kinematic region where the resolved photon contribution is significant.Comment: 32 pages including 6 figure

    Measurement of Jet Shapes in Photoproduction at HERA

    The shape of jets produced in quasi-real photon-proton collisions at centre-of-mass energies in the range 134277134-277 GeV has been measured using the hadronic energy flow. The measurement was done with the ZEUS detector at HERA. Jets are identified using a cone algorithm in the ηϕ\eta - \phi plane with a cone radius of one unit. Measured jet shapes both in inclusive jet and dijet production with transverse energies ETjet>14E^{jet}_T>14 GeV are presented. The jet shape broadens as the jet pseudorapidity (ηjet\eta^{jet}) increases and narrows as ETjetE^{jet}_T increases. In dijet photoproduction, the jet shapes have been measured separately for samples dominated by resolved and by direct processes. Leading-logarithm parton-shower Monte Carlo calculations of resolved and direct processes describe well the measured jet shapes except for the inclusive production of jets with high ηjet\eta^{jet} and low ETjetE^{jet}_T. The observed broadening of the jet shape as ηjet\eta^{jet} increases is consistent with the predicted increase in the fraction of final state gluon jets.Comment: 29 pages including 9 figure

    Measurement of the diffractive structure function in deep inelastic scattering at HERA

    This paper presents an analysis of the inclusive properties of diffractive deep inelastic scattering events produced in epep interactions at HERA. The events are characterised by a rapidity gap between the outgoing proton system and the remaining hadronic system. Inclusive distributions are presented and compared with Monte Carlo models for diffractive processes. The data are consistent with models where the pomeron structure function has a hard and a soft contribution. The diffractive structure function is measured as a function of \xpom, the momentum fraction lost by the proton, of β\beta, the momentum fraction of the struck quark with respect to \xpom, and of Q2Q^2. The \xpom dependence is consistent with the form \xpoma where a = 1.30 ± 0.08 (stat)  0.14+ 0.08 (sys)a~=~1.30~\pm~0.08~(stat)~^{+~0.08}_{-~0.14}~(sys) in all bins of β\beta and Q2Q^2. In the measured Q2Q^2 range, the diffractive structure function approximately scales with Q2Q^2 at fixed β\beta. In an Ingelman-Schlein type model, where commonly used pomeron flux factor normalisations are assumed, it is found that the quarks within the pomeron do not saturate the momentum sum rule.Comment: 36 pages, latex, 11 figures appended as uuencoded fil
