ATR-mediated phosphorylation of DNA polymerase η is needed for efficient recovery from UV damage
DNA polymerase η (polη) belongs to the Y-family of DNA polymerases and facilitates translesion synthesis past UV damage. We show that, after UV irradiation, polη becomes phosphorylated at Ser601 by the ataxia-telangiectasia mutated and Rad3-related (ATR) kinase. DNA damageâinduced phosphorylation of polη depends on its physical interaction with Rad18 but is independent of PCNA monoubiquitination. It requires the ubiquitin-binding domain of polη but not its PCNA-interacting motif. ATR-dependent phosphorylation of polη is necessary to restore normal survival and postreplication repair after ultraviolet irradiation in xeroderma pigmentosum variant fibroblasts, and is involved in the checkpoint response to UV damage. Taken together, our results provide evidence for a link between DNA damageâinduced checkpoint activation and translesion synthesis in mammalian cells
Clinical Data Reuse or Secondary Use: Current Status and Potential Future Progress
Objective: To perform a review of recent research in clinical data reuse or secondary use, and envision future advances in this field. Methods: The review is based on a large literature search in MEDLINE (through PubMed), conference proceedings, and the ACM Digital Library, focusing only on research published between 2005 and early 2016. Each selected publication was reviewed by the authors, and a structured analysis and summarization of its content was developed. Results: The initial search produced 359 publications, reduced after a manual examination of abstracts and full publications. The following aspects of clinical data reuse are discussed: motivations and challenges, privacy and ethical concerns, data integration and interoperability, data models and terminologies, unstructured data reuse, structured data mining, clinical practice and research integration, and examples of clinical data reuse (quality measurement and learning healthcare systems). Conclusion: Reuse of clinical data is a fast-growing field recognized as essential to realize the potentials for high quality healthcare, improved healthcare management, reduced healthcare costs, population health management, and effective clinical research
Wikipedia as an encyclopaedia of life
In his 2003 essay E O Wilson outlined his vision for an “encyclopaedia of life” comprising “an electronic page for each species of organism on Earth”, each page containing “the scientific name of the species, a pictorial or genomic presentation of the primary type specimen on which its name is based, and a summary of its diagnostic traits.” Although the “quiet revolution” in biodiversity informatics has generated numerous online resources, including some directly inspired by Wilson's essay (e.g., "http://ispecies.org":http://ispecies.org, "http://www.eol.org":http://www.eol.org), we are still some way from the goal of having available online all relevant information about a species, such as its taxonomy, evolutionary history, genomics, morphology, ecology, and behaviour. While the biodiversity community has been developing a plethora of databases, some with overlapping goals and duplicated content, Wikipedia has been slowly growing to the point where it now has over 100,000 pages on biological taxa. My goal in this essay is to explore the idea that, largely independent of the efforts of biodiversity informatics and well-funded international efforts, Wikipedia ("http://en.wikipedia.org/wiki/Main_Page":http://en.wikipedia.org/wiki/Main_Page) has emerged as potentially the best platform for fulfilling E O Wilson’s vision
Alkalinity generation from carbonate weathering in a silicate-dominated headwater catchment at Iskorasfjellet, northern Norway
The weathering rate of carbonate minerals is several
orders of magnitude higher than for silicate minerals. Therefore, small
amounts of carbonate minerals have the potential to control the dissolved
weathering loads in silicate-dominated catchments. Both weathering processes
produce alkalinity under the consumption of CO2. Given that only
alkalinity generation from silicate weathering is thought to be a long-term
sink for CO2, a misattributed weathering source could lead to incorrect
conclusions about long- and short-term CO2 fixation. In this study, we
aimed to identify the weathering sources responsible for alkalinity
generation and CO2 fixation across watershed scales in a degrading
permafrost landscape in northern Norway, 68.7â70.5ââN, and on
a temporal scale, in a subarctic headwater catchment on the mountainside of
Iskorasfjellet, characterized by sporadic permafrost and underlain mainly by
silicates as the alkalinity-bearing lithology. By analyzing total alkalinity
(AT) and dissolved inorganic carbon (DIC) concentrations, as well as the
stable isotope signature of the latter (ÎŽ13C-DIC), in
conjunction with dissolved cation and anion loads, we found that AT was
almost entirely derived from weathering of the sparse carbonate minerals. We
propose that in the headwater catchment the riparian zone is a hotspot area
of AT generation and release due to its enhanced hydrological connectivity
and that the weathering load contribution from the uphill catchment is
limited by insufficient contact time of weathering agents and weatherable
materials. By using stable water isotopes, it was possible to explain
temporal variations in AT concentrations following a precipitation event due
to surface runoff. In addition to carbonic acid, sulfuric acid, probably
originating from oxidation of pyrite or reduced sulfur in wetlands or from
acid deposition, is shown to be a potential corrosive reactant. An increased
proportion of sulfuric acid as a potential weathering agent may have
resulted in a decrease in AT. Therefore, carbonate weathering in the studied
area should be considered not only as a short-term CO2 sink but also
as a potential CO2 source. Finally, we found that AT increased with
decreasing permafrost probability, and attributed this relation to an
increased water storage capacity associated with increasing contact of
weathering agent and rock surfaces and enhanced microbial activity. As both
soil respiration and permafrost thaw are expected to increase with climate
change, increasing the availability of weathering agents in the form of
CO2 and water storage capacity, respectively, we suggest that future
weathering rates and alkalinity generation will increase concomitantly in
the study area.</p

Expectation values of four-quark operators in pions
The values of four-quark operators averaged over pions are expressed through
those averaged over vacuum. The specific values are obtained in the framework
of the factorization assumption. For the condensates of the light quarks of the
same flavour \bar q\Gamma q\bar q\Gamma q the scalar condensate is shown to be
an order of magnitude larger than the other ones. The condensates containing
the strange quarks \bar q q\bar s s appear to be only about twice smaller than
those of the light quarks. The degeneracy of the ground state in the

Implications of Electronics Constraints for Solid-State Quantum Error Correction and Quantum Circuit Failure Probability
In this paper we present the impact of classical electronics constraints on a
solid-state quantum dot logical qubit architecture. Constraints due to routing
density, bandwidth allocation, signal timing, and thermally aware placement of
classical supporting electronics significantly affect the quantum error
correction circuit's error rate. We analyze one level of a quantum error
correction circuit using nine data qubits in a Bacon-Shor code configured as a
quantum memory. A hypothetical silicon double quantum dot quantum bit (qubit)
is used as the fundamental element. A pessimistic estimate of the error
probability of the quantum circuit is calculated using the total number of
gates and idle time using a provably optimal schedule for the circuit
operations obtained with an integer program methodology. The micro-architecture
analysis provides insight about the different ways the electronics impact the
circuit performance (e.g., extra idle time in the schedule), which can
significantly limit the ultimate performance of any quantum circuit and
therefore is a critical foundation for any future larger scale architecture

Epileptic Seizure Detection on an Ultra-Low-Power Embedded RISC-V Processor Using a Convolutional Neural Network
The treatment of refractory epilepsy via closed-loop implantable devices that act on seizures either by drug release or electrostimulation is a highly attractive option. For such implantable medical devices, efficient and low energy consumption, small size, and efficient processing architectures are essential. To meet these requirements, epileptic seizure detection by analysis and classification of brain signals with a convolutional neural network (CNN) is an attractive approach. This work presents a CNN for epileptic seizure detection capable of running on an ultra-low-power microprocessor. The CNN is implemented and optimized in MATLAB. In addition, the CNN is also implemented on a GAP8 microprocessor with RISC-V architecture. The training, optimization, and evaluation of the proposed CNN are based on the CHB-MIT dataset. The CNN reaches a median sensitivity of 90% and a very high specificity over 99% corresponding to a median false positive rate of 6.8 s per hour. After implementation of the CNN on the microcontroller, a sensitivity of 85% is reached. The classification of 1 s of EEG data takes t=35 ms and consumes an average power of Pâ140 ÎŒW. The proposed detector outperforms related approaches in terms of power consumption by a factor of 6. The universal applicability of the proposed CNN based detector is verified with recording of epileptic rats. This results enable the design of future medical devices for epilepsy treatment
The behaviour of political parties and MPs in the parliaments of the Weimar Republic

Remarkably, this is stable throughout the entire lifespan of the Republic and not just in the later years and despite the varying content of votes across the lifespan of the Republic, and (2) that nearly all parties were troubled by intra-party divisions, though, in particular, the national socialists and communists became homogeneous in the final years of the Republic.Zukunftskolleg, University of Konstan
Electronic Health Record-Based Recruitment and Retention and Mobile Health App Usage: Multisite Cohort Study.
BACKGROUND: To address the obesity epidemic, there is a need for novel paradigms, including those that address the timing of eating and sleep in relation to circadian rhythms. Electronic health records (EHRs) are an efficient way to identify potentially eligible participants for health research studies. Mobile health (mHealth) apps offer available and convenient data collection of health behaviors, such as timing of eating and sleep.
OBJECTIVE: The aim of this descriptive analysis was to report on recruitment, retention, and app use from a 6-month cohort study using a mobile app called Daily24.
METHODS: Using an EHR query, adult patients from three health care systems in the PaTH clinical research network were identified as potentially eligible, invited electronically to participate, and instructed to download and use the Daily24 mobile app, which focuses on eating and sleep timing. Online surveys were completed at baseline and 4 months. We described app use and identified predictors of app use, defined as 1 or more days of use, versus nonuse and usage categories (ie, immediate, consistent, and sustained) using multivariate regression analyses.
RESULTS: Of 70,661 patients who were sent research invitations, 1021 (1.44%) completed electronic consent forms and online baseline surveys; 4 withdrew, leaving a total of 1017 participants in the analytic sample. A total of 53.79% (n=547) of the participants were app users and, of those, 75.3% (n=412), 50.1% (n=274), and 25.4% (n=139) were immediate, consistent, and sustained users, respectively. Median app use was 28 (IQR 7-75) days over 6 months. Younger age, White race, higher educational level, higher income, having no children younger than 18 years, and having used 1 to 5 health apps significantly predicted app use (vs nonuse) in adjusted models. Older age and lower BMI predicted early, consistent, and sustained use. About half (532/1017, 52.31%) of the participants completed the 4-month online surveys. A total of 33.5% (183/547), 29.3% (157/536), and 27.1% (143/527) of app users were still using the app for at least 2 days per month during months 4, 5, and 6 of the study, respectively.
CONCLUSIONS: EHR recruitment offers an efficient (ie, high reach, low touch, and minimal participant burden) approach to recruiting participants from health care settings into mHealth research. Efforts to recruit and retain less engaged subgroups are needed to collect more generalizable data. Additionally, future app iterations should include more evidence-based features to increase participant use
