    On dual canonical bases

    The dual basis of the canonical basis of the modified quantized enveloping algebra is studied, in particular for type AA. The construction of a basis for the coordinate algebra of the n×nn\times n quantum matrices is appropriate for the study the multiplicative property. It is shown that this basis is invariant under multiplication by certain quantum minors including the quantum determinant. Then a basis of quantum SL(n) is obtained by setting the quantum determinant to one. This basis turns out to be equivalent to the dual canonical basis

    Real-time RT-PCR analysis of mRNA decay: half-life of Beta-actin mRNA in human leukemia CCRF-CEM and Nalm-6 cell lines

    BACKGROUND: We describe an alternative method to determine mRNA half-life (t(1/2)) based on the Real-Time RT-PCR procedure. This approach was evaluated by using the β-actin gene as a reference molecule for measuring of mRNA stability. RESULTS: Human leukemia Nalm-6 and CCRF-CEM cells were treated with various concentrations of Actinomycin D to block transcription and aliquots were removed periodically. Total RNA was isolated and quantified using the RiboGreen(®) fluorescent dye with the VersaFluor Fluorometer System. One μg of total RNA was reverse transcribed and used as template for the amplification of a region of the β-actin gene (231 bp). To generate the standard curve, serial ten-fold dilutions of the pBactin-231 vector containing the cDNA amplified fragment were employed, β-actin mRNAs were quantified by Real-Time RT-PCR using the SYBR(®) Green I fluorogenic dye and data analyzed using the iCycle iQ system software. Using this method, the β-actin mRNA exhibited a half-life of 6.6 h and 13.5 h in Nalm-6 and CCRF-CEM cells, respectively. The t(1/2) value obtained for Nalm-6 is comparable to those estimated from Northern blot studies, using normal human leukocytes (5.5 h). CONCLUSIONS: We have developed a rapid, sensitive, and reliable method based on Real-Time RT-PCR for measuring mRNA half-life. Our results confirm that β-actin mRNA half-life can be affected by the cellular growth rate

    Inferring the rules of social interaction in migrating caribou

    Social interactions are a significant factor that influence the decision-making of species ranging from humans to bacteria. In the context of animal migration, social interactions may lead to improved decision-making, greater ability to respond to environmental cues, and the cultural transmission of optimal routes. Despite their significance, the precise nature of social interactions in migrating species remains largely unknown. Here we deploy unmanned aerial systems to collect aerial footage of caribou as they undertake their migration from Victoria Island to mainland Canada. Through a Bayesian analysis of trajectories we reveal the fine-scale interaction rules of migrating caribou and show they are attracted to one another and copy directional choices of neighbours, but do not interact through clearly defined metric or topological interaction ranges. By explicitly considering the role of social information on movement decisions we construct a map of near neighbour influence that quantifies the nature of information flow in these herds. These results will inform more realistic, mechanism-based models of migration in caribou and other social ungulates, leading to better predictions of spatial use patterns and responses to changing environmental conditions. Moreover, we anticipate that the protocol we developed here will be broadly applicable to study social behaviour in a wide range of migratory and non-migratory taxa.     SUSIG: an on-line signature database, associated protocols and benchmark results

    We present a new online signature database (SUSIG). The database consists of two parts that are collected using different pressure-sensitive tablets ( one with and the other without an LCD display). A total of 100 people contributed to each part, resulting in a database of more than 3,000 genuine signatures and 2,000 skilled forgeries. The genuine signatures in the database are real signatures of the contributors. In collecting skilled forgeries, forgers were shown the signing process on the monitor and were given a chance to practice. Furthermore, for a subset of the forgeries ( highly skilled forgeries), this animation was mapped onto the LCD screen of the tablet so that the forgers could trace over the mapped signature. Forgers in this group were also informed of how close they were to the reference signature, so that they could improve their forgery quality. We describe the signature acquisition process and several verification protocols for this database. We also report the performance of a state-of-the-art signature verification system using the associated protocols. The results show that the highly skilled forgery set is significantly more difficult compared to the skilled forgery set, providing researchers with challenging forgeries.
