Portuguese Children's Sleep Habits Questionnaire - validation and cross-cultural comparison
OBJECTIVE:
to validate the Portuguese version of the Children's Sleep Habits Questionnaire (CSHQ-PT) and compare it to the versions from other countries.
METHODS:
the questionnaire was previously adapted to the Portuguese language according to international guidelines. 500 questionnaires were delivered to the parents of a Portuguese community sample of children aged 2 to 10 years old. 370 (74%) valid questionnaires were obtained, 55 children met exclusion criteria and 315 entered in the validation study.
RESULTS:
the CSHQ-PT internal consistency (Cronbach's α) was 0.78 for the total scale and ranged from 0.44 to 0.74 for subscales. The test-retest reliability for subscales (Pearson's correlations, n=58) ranged from 0.59 to 0.85. Our data did not adjust to the original 8 domains structure in Confirmatory Factor Analysis but the Exploratory Factor Analysis extracted 5 factors that have correspondence to CSHQ subscales.
CONCLUSION:
the CSHQ-PT evidenced psychometric properties that are comparable to the versions from other countries and adequate for the screening of sleep disturbances in children from 2 to 10 years old
Microplastics and nanoplastics in haemodialysis waters: Emerging threats to be in our radar
Microplastics are present in the environment, in drinking water, in human blood and there is evidence of nanoplastics in tap water. The objective of this work was to analyze the possibility of hemodialysis patients being contaminated by micro and nanoplastics (MNPs) during dialysis treatment. The motivation for this investigation is the fact that hemodialysis patients use about 300–600 L of drinking water per week, which may be contaminated by MNPs. A literature review, a field investigation in a London hospital and an estimation of MNPs intake in patients were carried out. The results showed potential points of risk of contamination of patients by MNPs in hemodialysis. It was also estimated that for a filtration efficiency of 99 % for MNPs, the amount of microplastics that can penetrate the kidneys of patients is 0.0021–3768 particles/week. The assessment concludes that hemodialysis patients are at high risk of MNP contamination
Adesão à terapêutica antirretroviral de pessoas vivendo com HIV/aids em um municÃpio do interior paulista
RESUMO Objetivo Avaliar a adesão aos antirretrovirais de pessoas vivendo com o HIV/AIDS e identificar sua associação com variáveis sociodemográficas e clÃnicas. Métodos Estudo analÃtico transversal que utilizou instrumento sociodemográfico e o CEAT-HIV, com dados coletados no perÃodo de 2014 a 2015. Resultados Identificou-se 75,0% com grau de adesão bom/adequado. Verificou-se que os indivÃduos com idade entre 40 e 59 anos (p=0,029) e com mais de oito anos de estudo (p=0,043) obtiveram maior grau de adesão, assim como aqueles com diagnóstico de HIV/AIDS há mais de 10 anos (p=0,002), contagem de TCD4 >350 células/mm3 (p<0,001) e carga viral indetectável (p=0,025). Conclusão Nesse estudo, identificou-se uma boa adesão entre os sujeitos e observou-se que indivÃduos de maior faixa etária, maior grau de escolaridade, maior tempo de diagnóstico, elevada contagem de células TCD4 e carga viral indetectável estiveram associados a uma maior adesão ao tratamento
Long-term tobacco exposure and immunosenescence: paradoxical effects on T-cells telomere length and telomerase activity
Immunosenescence are alterations on immune system that occurs throughout an individual life. The main characteristic of this process is replicative senescence, evaluated by telomere shortening. Several factors implicate on telomere shortening, such as smoking. In this study, we evaluated the influence of smoking and Chronic Obstructive Pulmonary Disease (COPD) on cytokines, telomere length and telomerase activity.
Blood samples were collected from subjects aged over 60 years old: Healthy (never smokers), Smokers (smoking for over 30 years) and COPDs (ex-smokers for ≥15 years). A young group was included as control. PBMCs were cultured for assessment of telomerase activity using RT-PCR, and cytokines secretion flow cytometry. CD4+ and CD8+ purified lymphocytes were used to assess telomere length using FlowFISH.
We observed that COPD patients have accelerated telomere shortening. Paradoxically, smokers without lung damage showed preserved telomere length, suggesting that tobacco smoking may affect regulatory mechanisms, such as telomerase. Telomerase activity showed diminished activity in COPDs, while Smokers showed increased activity compared to COPDs and Healthy groups.
Extracellular environment reflected this unbalance, indicated by an anti-inflammatory profile in Smokers, while COPDs showed an inflammatory prone profile. Further studies focusing on telomeric maintenance may unveil mechanisms that are associated with cancer under long-term smoking
Field Theory for a Deuteron Quantum Liquid
Based on general symmetry principles we study an effective Lagrangian for a
neutral system of condensed spin-1 deuteron nuclei and electrons, at
greater-than-atomic but less-than-nuclear densities. We expect such matter to
be present in thin layers within certain low-mass brown dwarfs. It may also be
produced in future shock-wave-compression experiments as an effective fuel for
laser induced nuclear fusion. We find a background solution of the effective
theory describing a net spin zero condensate of deuterons with their spins
aligned and anti-aligned in a certain spontaneously emerged preferred
direction. The spectrum of low energy collective excitations contains two spin
waves with linear dispersions -- like in antiferromagnets -- as well as gapped
longitudinal and transverse modes related to the Meissner effect -- like in
superconductors. We show that counting of the Nambu-Goldstone modes of
spontaneously broken internal and space-time symmetries obeys, in a nontrivial
way, the rules of the Goldstone theorem for Lorentz non-invariant systems. We
discuss thermodynamic properties of the condensate, and its potential

Mulheres internadas por agressão em um hospital de pronto socorro: (in)visibilidade da violência
Trata-se de um estudo do tipo transversal, com o objetivo de caracterizar a hospitalização de mulheres vÃtimas de agressão, no Hospital de Pronto Socorro de Porto Alegre, Rio Grande do Sul, durante o ano de 2005. A coleta dos dados foi realizada nos prontuários de mulheres com idade a partir de 18 anos, vÃtimas de agressão. A análise foi descrita a partir da utilização do Software Epi-Info, com a categorização dos eventos, do cruzamento de variáveis descritivas e de Ãndices frequenciais absolutos e relativos. No perÃodo do estudo, foram hospitalizadas 73 mulheres, vÃtimas de agressão; 49,3% na faixa etária de 18 a 29; 41,1% foram agredidas com arma de fogo e 37% com arma branca. Na maioria dos prontuários, os dados estavam incompletos e/ou faltavam registros, o que pode indicar que muitos casos de agressões não foram identificados durante o perÃodo da hospitalização e que o tratamento ficou restrito à s lesões fÃsicas
Rede de cuidado em saúde mental: visão dos coordenadores da estratégia saúde da famÃlia
RESUMO Objetivo Objetivou-se analisar a visão de coordenadores da Estratégia Saúde da FamÃlia (ESF) sobre a conformação da rede de saúde mental no municÃpio de Pelotas/RS. Método Estudo descritivo e exploratório, com abordagem qualitativa, realizado com seis coordenadores de ESF no ano de 2012. Para a coleta dos dados, foi utilizada a entrevista semiestruturada, e os resultados foram avaliados por análise temática. Resultados A rede construÃda pelos coordenadores é composta por poucos serviços formais e focada no Centro de Atenção Psicossocial (CAPS), e a rede informal é descrita com maiores pontos de cuidado. As trocas de corresponsabilidade no cuidado em rede ocorrem de forma frágil, sem o diálogo necessário. Conclusão Os coordenadores demonstram pouco conhecimento da rede formal de cuidado e uma atenção focada nos serviços da ESF e no CAPS. Reconhecem a importância de haver polÃticas públicas que valorizem e ampliem os espaços informais de cuidado no território
Splice Site Mutations in the ATP7A Gene
Menkes disease (MD) is caused by mutations in the ATP7A gene. We describe 33 novel splice site mutations detected in patients with MD or the milder phenotypic form, Occipital Horn Syndrome. We review these 33 mutations together with 28 previously published splice site mutations. We investigate 12 mutations for their effect on the mRNA transcript in vivo. Transcriptional data from another 16 mutations were collected from the literature. The theoretical consequences of splice site mutations, predicted with the bioinformatics tool Human Splice Finder, were investigated and evaluated in relation to in vivo results. Ninety-six percent of the mutations identified in 45 patients with classical MD were predicted to have a significant effect on splicing, which concurs with the absence of any detectable wild-type transcript in all 19 patients investigated in vivo. Sixty-seven percent of the mutations identified in 12 patients with milder phenotypes were predicted to have no significant effect on splicing, which concurs with the presence of wild-type transcript in 7 out of 9 patients investigated in vivo. Both the in silico predictions and the in vivo results support the hypothesis previously suggested by us and others, that the presence of some wild-type transcript is correlated to a milder phenotype
MIR376A is a regulator of starvation-induced autophagy
Background: Autophagy is a vesicular trafficking process responsible for the degradation of long-lived, misfolded or abnormal proteins, as well as damaged or surplus organelles. Abnormalities of the autophagic activity may result in the accumulation of protein aggregates, organelle dysfunction, and autophagy disorders were associated with various diseases. Hence, mechanisms of autophagy regulation are under exploration.
Methods: Over-expression of hsa-miR-376a1 (shortly MIR376A) was performed to evaluate its effects on autophagy. Autophagy-related targets of the miRNA were predicted using Microcosm Targets and MIRanda bioinformatics tools and experimentally validated. Endogenous miRNA was blocked using antagomirs and the effects on target expression and autophagy were analyzed. Luciferase tests were performed to confirm that 3’ UTR sequences in target genes were functional. Differential expression of MIR376A and the related MIR376B was compared using TaqMan quantitative PCR.
Results: Here, we demonstrated that, a microRNA (miRNA) from the DlkI/Gtl2 gene cluster, MIR376A, played an important role in autophagy regulation. We showed that, amino acid and serum starvation-induced autophagy was blocked by MIR376A overexpression in MCF-7 and Huh-7 cells. MIR376A shared the same seed sequence and had overlapping targets with MIR376B, and similarly blocked the expression of key autophagy proteins ATG4C and BECN1 (Beclin 1). Indeed, 3’ UTR sequences in the mRNA of these autophagy proteins were responsive to MIR376A in luciferase assays. Antagomir tests showed that, endogenous MIR376A was participating to the control of ATG4C and BECN1 transcript and protein levels. Moreover, blockage of endogenous MIR376A accelerated starvation-induced autophagic activity. Interestingly, MIR376A and MIR376B levels were increased with different kinetics in response to starvation stress and tissue-specific level differences were also observed, pointing out to an overlapping but miRNA-specific biological role.
Conclusions: Our findings underline the importance of miRNAs encoded by the DlkI/Gtl2 gene cluster in stress-response control mechanisms, and introduce MIR376A as a new regulator of autophagy
