The effect of mirabegron on patient-related outcomes in patients with overactive bladder: the results of post hoc correlation and responder analyses using pooled data from three randomized Phase III trials
Purpose To understand how improvements in the symptoms
of overactive bladder (OAB) seen with the b3-adrenoceptor
agonist mirabegron 50 mg, correlate with patient experience
as measured by validated and standard patient-reported
outcomes (PROs), and to identify whether there is overall
directional consistency in the responsiveness of PROs to
treatment effect.
Methods In a post hoc analysis of pooled data from three
randomized, double-blind, placebo-controlled, 12-week
Phase III trials of mirabegron 50 mg once daily, responder
rates for incontinence frequency (C50 % reduction in
incontinence episodes/24 h from baseline to final visit),
micturition frequency (B8 micturitions/24 h at final visit),
and PROs [minimally important differences in patient
perception of bladder condition (PPBC) and subsets of the
overactive bladder questionnaire (OAB-q) measuring total
health-related quality of life (HRQoL), and symptom
bother] were evaluated individually and in combination.
Results Mirabegron 50 mg demonstrated greater
improvement from baseline to final visit than placebo for
each of the responder analyses, whether for individual
objective and subjective outcomes or combinations thereof.
These improvements versus placebo were statistically significant
for all double and triple responder analyses and for
all single responder analyses except PPBC. PRO measurements
showed directional consistency and significant
correlations, and there were also significant correlations
between objective and subjective measures of efficacy.
Conclusions The improvements in objective measures
seen with mirabegron 50 mg translate into a meaningful
clinical benefit as evident by the directional consistency
seen in HRQoL measures of benefit

Structural characterization of InAlAsSb/InGaAs/InP heterostructures for solar cells
In this work, we have characterized by transmission electron microscopy techniques the structural properties of InAlAsSb/InGaAs/InP heterostructures, with target applications in high efficiency solar cells. Previous photoluminescence (PL)1 analysis suggested the existence of compositional fluctuations in the active layer of these heterostructures. 220 bright field (BF)2 diffraction contrast micrographs have revealed strong strain contrast in the InGaAs buffer layer, related to the existence of these compositional fluctuations. The effect of a decomposed buffer on the growth of the InAlAsSb layer has been analyzed through the simulation of the strain fields in the heterostructure using the finite elements method (FEM).3 These simulations have shown that the strain in the buffer layer due to the compositional fluctuations only affects the first few nm of the InAlAsSb layer. The analysis by aberration corrected high angle annular dark field scanning transmission electron microscopy (HAADF-STEM)4and electron energy loss spectroscopy (EELS)5of the composition of the InAlAsSb layer reveals that any compositional fluctuation is only observed as an average effect, rather than in the form of clustering or atomically sharp transitions. The limitations of these techniques for the detection of small 3D compositional fluctuations are discussed
Coral Reef Resilience Index for Novel Ecosystems: A Spatial Planning Tool for Managers and Decision Makers - A Case Study from Puerto Rico
Timely information is critical for coral reef managers and decision-makers to implement sustainable management measures. A Coral Reef Resilience Index (CRRI) was developed with a GIS-coupled decision-making tool applicable for Caribbean coral reef ecosystems. The CRRI is based on a five-point scale parameterized from the quantitative characterization of benthic assemblages. Separate subindices such as the Coral Index, the Threatened Species Index, and the Algal Index also provide specific information regarding targeted benthic components. This case study was based on assessments conducted in 2014 on 11 reef sites located across 3 geographic zones and 3 depth zones along the southwestern shelf of the island of Puerto Rico, Caribbean Sea. There was a significant spatial and bathymetric gradient (p < 0.05) in the distribution of CRRI values indicating higher degradation of inshore reefs. Mean global CRRI ranged from 2.78 to 3.17 across the shelf, ranking them as “fair.” The Coral Index ranged from 2.60 to 3.76, ranking reefs from “poor” to “good,” showing a general cross-shelf trend of improving conditions with increasing distance from pollution sources. Turbidity and ammonia were significantly correlated to CRRI scores. Multiple recommendations are provided based on coral reef conditions according to observed CRRI rankings
Magnetic operations: a little fuzzy physics?
We examine the behaviour of charged particles in homogeneous, constant and/or
oscillating magnetic fields in the non-relativistic approximation. A special
role of the geometric center of the particle trajectory is elucidated. In
quantum case it becomes a 'fuzzy point' with non-commuting coordinates, an
element of non-commutative geometry which enters into the traditional control
problems. We show that its application extends beyond the usually considered
time independent magnetic fields of the quantum Hall effect. Some simple cases
of magnetic control by oscillating fields lead to the stability maps differing
from the traditional Strutt diagram.Comment: 28 pages, 8 figure
Inflammatory Animal Model for Parkinson's Disease: The Intranigral Injection of LPS Induced the Inflammatory Process along with the Selective Degeneration of Nigrostriatal Dopaminergic Neurons
We have developed an animal model of degeneration of the nigrostriatal dopaminergic neurons, the neuronal system involved in Parkinson's disease (PD). The implication of neuroinflammation on this disease was originally established in 1988, when the presence of activated microglia in the substantia nigra (SN) of parkinsonians was reported by McGeer et al. Neuroinflammation could be involved in the progression of the disease or even has more direct implications. We injected 2 μg of the potent proinflammatory compound lipopolysaccharide (LPS) in different areas of the CNS, finding that SN displayed the highest inflammatory response and that dopaminergic (body) neurons showed a special and specific sensitivity to this process with the induction of selective dopaminergic degeneration. Neurodegeneration is induced by inflammation since it is prevented by anti-inflammatory compounds. The special sensitivity of dopaminergic neurons seems to be related to the endogenous dopaminergic content, since it is overcome by dopamine depletion. Compounds that activate microglia or induce inflammation have similar effects to LPS. This model suggest that inflammation is an important component of the degeneration of the nigrostriatal dopaminergic system, probably also in PD. Anti-inflammatory treatments could be useful to prevent or slow down the rate of dopaminergic degeneration in this disease
An improved method for statistical studies of the internal kinematics of HII regions: the case of M 83
We present the integrated Halpha emission line profile for 157 HII regions in
the central 3.4' x 3.4' of the galaxy M 83 (NGC 5236). Using the Fabry-Perot
interferometer GHaFaS, on the 4.2 m William Herschel on La Palma, we show the
importance of a good characterization of the instrumental response function for
the study of line profile shapes. The luminosity-velocity dispersion relation
is also studied, and in the log(L)-log(sigma) plane we do not find a linear
relation, but an upper envelope with equation log(L)=0.9 *log(sigma)+38.1. For
the adopted distance of 4.5 Mpc, the upper envelope appears at the luminosity
L=10^38.5 ergs, in full agreement with previous studies of other galaxies,
reinforcing the idea of using HII regions as standard candles.Comment: 13 pages, 9 figures, accepted for publication in MNRA
