    Nest predation in an urbanizing landscape: the role of exotic shrubs

    Nest predation is considered a primary force shaping avian communities, and landscape-scale features (e.g., amount of fragmentation) are generally recognized as factors mediating nest predation. These same landscape-scale features, however, may promote invasion by exotic plants, which may, in turn, increase risk of nest predation. We examined whether the use of exotic shrubs (Lonicera spp. and Rosa multiflora Thumb.) affected nest predation across 12 riparian forest sites along a rural–urban gradient (<1– 47% urban land cover within 1 km). From 2001 to 2003, 188 Northern Cardinal (Cardinalis cardinalis) and American Robin (Turdus migratorius) nests ≤5 m tall were monitored. Nest substrate, nest height, and distance from the forest edge were recorded for each nest, whereas nest placement and nest patch characteristics were measured only for Northern Cardinal nests (n = 68). To further assess relative rates of nest predation in native vs. exotic shrubs while controlling for nest height, distance to edge, and land use, we conducted an artificial nest experiment at two rural sites. Artificial nests (n = 79) were placed at similar heights in honeysuckle, rose, and native nest substrates along a transect 50–75 m from the forest edge. Nest substrate and landscape type alone failed to account for differences in daily mortality rates. Instead, the effect of nest substrate varied with the landscape matrix, such that nests in exotic shrubs in urbanizing landscapes were twice as likely to be depredated than nests in native substrates, irrespective of distance from the edge. Artificial nests placed in exotic shrubs in rural landscapes also suffered higher rates of nest failure than artificial nests in native substrates. Daily mortality rates were greater for nests in exotic shrubs, likely due to reduced nest height and larger shrub volume surrounding the nest. Nests in exotic shrubs were 1.5–2 m lower to the ground and within patches containing 6–9 times more exotic shrub volume. These differences may improve search efficiency of mammalian predators, which appear to be the main predators at our study sites. Based on marks present on recovered clay eggs, 68% of the predation events were attributed to mammals. These findings demonstrate that exotic shrubs can reduce nesting success of forest birds and may cause increased nest failure in urbanizing landscapes. This illustrates another way that exotic plants may diminish habitat quality and limit the capacity of urban forests to contribute to wildlife conservation; therefore, restoring the native shrub community may prove beneficial

    We demonstrate coherent microwave control of the rotational, hyperfine and Zeeman states of ultracold CaF molecules, and the magnetic trapping of these molecules in a single, selectable quantum state. We trap about 5×1035\times 10^{3} molecules for 2 s at a temperature of 65(11) μ\muK and a density of 1.2×1051.2 \times 10^{5} cm3^{-3}.     Mast cells limit extracellular levels of IL-13 via a serglycin proteoglycan-serine protease axis

    Mast cell (MC) granules contain large amounts of proteases of the chymase, tryptase and carboxypeptidase A (MC-CPA) type that are stored in complex with serglycin, a proteoglycan with heparin side chains. Hence, serglycin-protease complexes are released upon MC degranulation and may influence local inflammation. Here we explored the possibility that a serglycin-protease axis may regulate levels of IL-13, a cytokine involved in allergic asthma. Indeed, we found that wild-type MCs efficiently degraded exogenous or endogenously produced IL-13 upon degranulation, whereas serglycin(-/-) MCs completely lacked this ability. Moreover, MC-mediated IL-13 degradation was blocked both by a serine protease inhibitor and by a heparin antagonist, which suggests that IL-13 degradation is catalyzed by serglycin-dependent serine proteases and that optimal IL-13 degradation is dependent on both the serglycin and the protease component of the serglycin-protease complex. Moreover, IL-13 degradation was abrogated in MC-CPA(-/-) MC cultures, but was normal in cultures of MCs with an inactivating mutation of MC-CPA, which suggests that the IL-13-degrading serine proteases rely on MC-CPA protein. Together, our data implicate a serglycin-serine protease axis in the regulation of extracellular levels of IL-13. Reduction of IL-13 levels through this mechanism possibly can provide a protective function in the context of allergic inflammation

    FcRn-mediated antibody transport across epithelial cells revealed by electron tomography

    The neonatal Fc receptor (FcRn) transports maternal IgG across epithelial barriers, thereby providing the fetus or newborn with humoral immunity before its immune system is fully functional. In newborn rats, FcRn transfers IgG from milk to blood by apical-to-basolateral transcytosis across intestinal epithelial cells. The pH difference between the apical (pH 6.0–6.5) and basolateral (pH 7.4) sides of intestinal epithelial cells facilitates the efficient unidirectional transport of IgG, because FcRn binds IgG at pH 6.0–6.5 but not at pH 7 or more. As milk passes through the neonatal intestine, maternal IgG is removed by FcRn-expressing cells in the proximal small intestine (duodenum and jejunum); remaining proteins are absorbed and degraded by FcRn-negative cells in the distal small intestine (ileum). Here we use electron tomography to make jejunal transcytosis visible directly in space and time, developing new labelling and detection methods to map individual nanogold-labelled Fc within transport vesicles and simultaneously to characterize these vesicles by immunolabelling. Combining electron tomography with a nonperturbing endocytic label allowed us to conclusively identify receptor-bound ligands, resolve interconnecting vesicles, determine whether a vesicle was microtubule-associated, and accurately trace FcRn-mediated transport of IgG. Our results present a complex picture in which Fc moves through networks of entangled tubular and irregular vesicles, only some of which are microtubule-associated, as it migrates to the basolateral surface. New features of transcytosis are elucidated, including transport involving multivesicular body inner vesicles/tubules and exocytosis through clathrin-coated pits. Markers for early, late and recycling endosomes each labelled vesicles in different and overlapping morphological classes, revealing spatial complexity in endo-lysosomal trafficking

    Positive relationships between association strength and phenotypic similarity characterize the assembly of mixed-species bird flocks worldwide

    Competition theory predicts that local communities should consist of species that are more dissimilar than expected by chance. We find a strikingly different pattern in a multicontinent data set (55 presence-absence matrices from 24 locations) on the composition of mixed-species bird flocks, which are important sub-units of local bird communities the world over. By using null models and randomization tests followed by meta-analysis, we find the association strengths of species in flocks to be strongly related to similarity in body size and foraging behavior and higher for congeneric compared with noncongeneric species pairs. Given the local spatial scales of our individual analyses, differences in the habitat preferences of species are unlikely to have caused these association patterns; the patterns observed are most likely the outcome of species interactions. Extending group-living and social-information-use theory to a heterospecific context, we discuss potential behavioral mechanisms that lead to positive interactions among similar species in flocks, as well as ways in which competition costs are reduced. Our findings highlight the need to consider positive interactions along with competition when seeking to explain community assembly

    Testing Alternative Hypotheses for the Cause of Population Declines: The Case of the Red-Headed Woodpecker

    The Red-headed Woodpecker (Melanerpes erythrocephalus) has experienced strong population declines during the past 3 decades. Using North American Breeding Bird Survey (BBS) and Audubon Christmas Bird Count (CBC) data, we investigated 4 hypotheses that may explain this decline, including: (1) interspecific competition with native Red-bellied Woodpeckers (Melanerpes carolinus) and nonnative European Starlings (Sturnus vulgaris); (2) predation by Cooper\u27s Hawks (Accipiter cooperii) and Sharp-shinned Hawks (Accipiter striatus); (3) climate change; and (4) changes in forested area within their range. In analyses of both the breeding and overwintering periods, our results indicated a role of increased accipiter populations in driving Red-headed Woodpecker declines through increased predation. We also found evidence for significant effects of warmer winter temperatures and increased forest cover, both directly and indirectly through their effects on enhancing accipiter populations. In contrast, our results failed to support the hypothesis that interspecific competition with either Red-bellied Woodpeckers or European Starlings has played a role in Red-headed Woodpecker declines. Despite considerable evidence for nest-site competition and aggression between Red-headed Woodpeckers and both Red-bellied Woodpeckers and European Starlings, these interactions do not appear to be limiting Red-headed Woodpecker populations

    Group B Streptococcus vaccine development: present status and future considerations, with emphasis on perspectives for low and middle income countries.

    Globally, group B Streptococcus (GBS) remains the leading cause of sepsis and meningitis in young infants, with its greatest burden in the first 90 days of life. Intrapartum antibiotic prophylaxis (IAP) for women at risk of transmitting GBS to their newborns has been effective in reducing, but not eliminating, the young infant GBS disease burden in many high income countries. However, identification of women at risk and administration of IAP is very difficult in many low and middle income country (LMIC) settings, and is not possible for home deliveries. Immunization of pregnant women with a GBS vaccine represents an alternate pathway to protecting newborns from GBS disease, through the transplacental antibody transfer to the fetus in utero. This approach to prevent GBS disease in young infants is currently under development, and is approaching late stage clinical evaluation. This manuscript includes a review of the natural history of the disease, global disease burden estimates, diagnosis and existing control options in different settings, the biological rationale for a vaccine including previous supportive studies, analysis of current candidates in development, possible correlates of protection and current status of immunogenicity assays. Future potential vaccine development pathways to licensure and use in LMICs, trial design and implementation options are discussed, with the objective to provide a basis for reflection, rather than recommendations
