    Observations of Spontaneous Raman Scattering in Silicon Slow-light Photonic Crystal Waveguides

    We report the observations of spontaneous Raman scattering in silicon photonic crystal waveguides. Continuous-wave measurements of Stokes emission for both wavelength and power dependence is reported in single line-defect waveguides in hexagonal lattice photonic crystal silicon membranes. By utilizing the Bragg gap edge dispersion of the TM-like mode for pump enhancement and the TE-like fundamental mode-onset for Stokes enhancement, the Stokes emission was observed to increase by up to five times in the region of slow group velocity. The results show explicit nonlinear enhancement in a silicon photonic crystal slow-light waveguide device.Comment: 12 pages, 4 figure

    A Field Test Study on under Water Vacuum Preloading Method

    Vacuum preloading technique incorporating prefabricated vertical drains is one of the most widely used ground improvement methods in the world for improving the engineering properties of soft clays. Although many successful on-shore cases on application of the technique have been reported, the effectiveness of applying the technique under water has not yet been investigated. Moreover, many technical and operation factors, that are playing important roles in vacuum consolidation, are also not yet fully understood. To study the feasibility of under water vacuum preloading, a large-scale field test was conducted. A 50 m by 50 m geo-membrane was laid under water in an 80 m wide by 100 m long pond. The geo-membrane was custom made with drainage outlet pipes to release the trapped air bubbles during the placement under water. Prefabricated vertical drains were installed on an equilateral triangular grid at a spacing of 1.2 m to a depth of 7 m. Internal drainage pipes were provided in the sand cushion layers to provide a passage for the prefabricated vertical drains with the external vacuum pumps. Instruments such as piezometers, vacuum sensors, inclinometers, settlement plates and extensometers were installed to monitor the performance of the system. An automatic and remote wireless monitoring system was installed for data collection because of difficult access. Vane shear tests and cone penetration tests were conducted before and after vacuum preloading to determine the effectiveness of the operation. This paper documents the constructions of the field test and reports the major observations from the monitored readings. The operation has demonstrated that under water vacuum preloading is feasible and that with proper design and construction procedure, a very tight seal can be provided by the geo-membrane separating the water and the underlying prefabricated vertical drains through out the test. The monitored results demonstrate that the stiffness and strength of the soft clay can be improved effectively

    Iterative in Situ Click Chemistry Assembles a Branched Capture Agent and Allosteric Inhibitor for Akt1

    We describe the use of iterative in situ click chemistry to design an Akt-specific branched peptide triligand that is a drop-in replacement for monoclonal antibodies in multiple biochemical assays. Each peptide module in the branched structure makes unique contributions to affinity and/or specificity resulting in a 200 nM affinity ligand that efficiently immunoprecipitates Akt from cancer cell lysates and labels Akt in fixed cells. Our use of a small molecule to preinhibit Akt prior to screening resulted in low micromolar inhibitory potency and an allosteric mode of inhibition, which is evidenced through a series of competitive enzyme kinetic assays. To demonstrate the efficiency and selectivity of the protein-templated in situ click reaction, we developed a novel QPCR-based methodology that enabled a quantitative assessment of its yield. These results point to the potential for iterative in situ click chemistry to generate potent, synthetically accessible antibody replacements with novel inhibitory properties

    Childhood obesity management shifting from health care system to school system: Intervention study of school-based weight management programme

The aim of this study is evaluation of the effectiveness of a multi-component school-based weight management programme for overweight and obese primary school children via a home-school joint venture. Methods: This study made use of variety of behavioural modification strategies integrating into the Health Promoting School approach to promote healthy lifestyles. The participants were overweight and obese students aged between 8 and 12 from six participating schools. The interventions involved students attending ten 75 minutes after-school sessions and one 3-hour week-end session of practical interactive and fun activities on healthy eating and exercise, and meal plan together with parents and printed tailor-made management advices. Parents received an introductory seminar with 2 sets of specially designed exercise for their overweight children. The tools to measure bodyweight and fat percentage and standing height were bio-impedance body fat scale and a portable stadiometer. Self-administered questionnaire was used to measure knowledge, attitudes and behaviours. McNemar test was utilized to compare the proportions of behaviour changes within the same group to assess for the trends of changes. BMI z-score and body fat percentage of intervention participants at baseline, 4 month and 8 month were compared pair-wisely using tests of within subject contrasts in repeated measures ANOVA to assess for programme sustainability. Results: Those students in the intervention group reduced their BMI z-score (-0.21, 95% CI -0.34 to -0.07, P = 0.003) and body fat (-2.67%, 95% CI -5.12 to -0.22, P = 0.033) compared to wait list control group with statistical significant, and the intervention group also had a significant reduction in BMI z-score (-0.06, 95% CI -0.11, -0.007, P = 0.028) and body fat (-1.71%, 95% CI, -3.44 to 0.02, P = 0.052) after a 4 month maintenance period. Improvement of dietary habits and positive attitudes towards exercise were observed among the intervention group. Conclusion: School based weight management programme integrated into a Health Promoting School approach with improved school policies and environment in supporting individual skills of obese students and their parents appears to be a promising practice for sustaining weight control.Link_to_subscribed_fulltex

    NF-κB p65 Subunit Is Modulated by Latent Transforming Growth Factor-β Binding Protein 2 (LTBP2) in Nasopharyngeal Carcinoma HONE1 and HK1 Cells

    NF-kappa B is a well-characterized transcription factor, widely known as a key player in tumor-derived inflammation and cancer development. Herein, we present the functional and molecular relevance of the canonical NF-kappa B p65 subunit in nasopharyngeal carcinoma (NPC). Loss-and gain-of-function approaches were utilized to reveal the functional characteristics of p65 in propagating tumor growth, tumor-associated angiogenesis, and epithelial-to-mesenchymal transition in NPC cells. Extracellular inflammatory stimuli are critical factors that trigger the NF-kappa B p65 signaling; hence, we investigated the components of the tumor microenvironment that might potentially influence the p65 signaling pathway. This led to the identification of an extracellular matrix (ECM) protein that was previously reported as a candidate tumor suppressor in NPC. Our studies on the Latent Transforming Growth Factor-beta Binding Protein 2 (LTBP2) protein provides substantial evidence that it can modulate the p65 transcriptional activity. Re-expression of LTBP2 elicits tumor suppressive effects that parallel the inactivation of p65 in NPC cells. LTBP2 was able to reduce phosphorylation of p65 at Serine 536, inhibit nuclear localization of active phosphorylated p65, and impair the p65 DNA-binding ability. This results in a consequential down-regulation of p65-related gene expression. Therefore, the data suggest that the overall up-regulation of p65 expression and the loss of this candidate ECM tumor suppressor are milestone events contributing to NPC development.published_or_final_versio

    Paclitaxel induces immunogenic cell death in ovarian cancer via TLR4/IKK2/SNARE-dependent exocytosis

    Emerging evidence shows that the efficacy of chemotherapeutic drugs are reliant on their capability to induce immunogenic cell death (ICD), thus transforming dying tumor cells into antitumor vaccines. We wanted to uncover potential therapeutic strategies that target ovarian cancer by having a better understanding of the standard-of-care chemotherapy treatment. Here, we showed in ovarian cancer that paclitaxel induced ICD-associated DAMPs (i.e. damage-associated molecular patterns, such as CALR exposure, ATP secretion and HMGB1 release) in vitro and elicited significant antitumor responses in tumor vaccination assays in vivo. Paclitaxel-induced TLR4 signaling was essential to the release of DAMPs, which lead to the activation of NF-κB-mediated CCL2 transcription and IKK2-mediated SNARE-dependent vesicle exocytosis, thus exposing CALR on the cell surface. Paclitaxel induced ER stress, which triggered PERK activation and eIF2α phosphorylation independent of TLR4. Paclitaxel chemotherapy induced T cell infiltration in ovarian tumors of the responsive patients; CALR expression in primary ovarian tumors also correlated with patients' survival and patient response to chemotherapy. These findings suggest that the effectiveness of paclitaxel relied upon the activation of antitumor immunity through ICD via TLR4 and highlighted the importance of CALR expression in cancer cells as an indicator of response to paclitaxel chemotherapy in ovarian cancer

    The aim of this special issue is to examine more closely of the implementations of international human resource management (IHRM) practices in the contexts of high uncertainties. It seeks contexts of relevance, encompassing those experiencing financial crisis, economic sanctions, political and civil uncertainty, environmental collapse and/or deep recession. It aims to supplement the Danger and Risk as Challenges for HRM in the IJHRM special issue which encompasses terrorism, violent disorder, crime and other physical risks, by focusing on initially seemingly peaceful forms of uncertainty, even if their consequences might lead to societal collapse. While appreciating that these contexts are very different, the key theme that cuts across all of these contexts are the unexpected changes that they brought, creating considerable ambiguity for businesses, and how they manage their people. Businesses will face the challenges of coping in such contexts, with unpredictability in demand, and in supplier relations, in adding greater time pressure to the decision-making process, and in terms of work and employment relations (Pearson & Clair, 1998). Through operating in different settings, multinational enterprises (MNEs) may be able to hedge risk, but at the same time protecting their own interest from a distance can be extremely difficult (Cantwell, Dunning, & Lundan, 2010). They will also impact on MNE decisions to invest and reinvest in particular settings (Oh & Oetzel, 2011). However, reducing or eliminating their presence in the host location is not always possible. MNEs may have substantial resources and infrastructural interests in the host location that need to be protected. Again, there is often a pressing need of MNEs to use expatriates on international assignments to complete strategically critical tasks, but the same time managing expatriate staff becomes much more difficult when countries of domicile become less certain. However, these situations often present golden opportunity for businesses. Studies have found that the option value of MNEs in entering a country under the uncertain conditions can be high (Miller, 1998). This is because government and international bodies often inject considerable amount of investments into the affected countries in aiding the recovery and rebuilding process and, in turn, pumped up the local aggregate demands, opening new opportunities for MNEs in relevant industries (Vigdor, 2008). At the same time, consumers’ demand for products and services may change; demand may not necessarily decline, but what consumers may want may be different, and this will impact on the demands placed on a firm’s human resources.     Age-related grey matter volume correlates of response inhibition and shifting in Attention Deficit Hyperactivity Disorder

    Background Children with attention-deficit hyperactivity disorder (ADHD) have difficulties with executive function and impulse control which may improve with age. Aims To map the brain correlates of executive function in ADHD and determine age-related changes in reaction times and brain volumes. Method Attention-deficit hyperactivity disorder and control groups were compared on the change task measures of response inhibition (stop signal reaction time, SSRT) and shifting (change response reaction time, CRRT). voxel-wise magnetic resonance imaging (MRI) correlations of reaction times and grey matter volume were determined, along with bivariate correlations of reaction times, brain volumes and age.Results Individuals in the ADHD group had longer SSRTs and CRRTs. Anterior cingulate, striatal and medial temporal volumes highly correlated with SSRT. Striatal and cerebellar volumes strongly correlated with CRRT. Older children had faster reaction times and larger regional brain volumes. In controls, orbitofrontal, medial temporal and cerebellar volumes correlated with CRRT but not SSRT. Neither reaction times nor regional brain volumes were strongly age- dependent. Conclusions Our evidence supports delayed brain maturation in ADHD and implies that some features of ADHD improve with age.link_to_subscribed_fulltex
