    Polymorphic genetic characterization of the ORF7 gene of porcine reproductive and respiratory syndrome virus (PRRSV) in China

    <p>Abstract</p> <p>Background</p> <p>Porcine reproductive and respiratory syndrome virus (PRRSV) exhibits extensive genetic variation. The outbreak of a highly pathogenic PRRS in 2006 led us to investigate the extent of PRRSV genetic diversity in China. To this end, we analyzed the Nsp2 and ORF7 gene sequences of 98 Chinese PRRSV isolates.</p> <p>Results</p> <p>Preliminary analysis indicated that highly pathogenic PRRSV strains with a 30-amino acid deletion in the Nsp2 protein are the dominant viruses circulating in China. Further analysis based on ORF7 sequences revealed that all Chinese isolates were divided into 5 subgroups, and that the highly pathogenic PRRSVs were distantly related to the MLV or CH-1R vaccine, raising doubts about the efficacy of these vaccines. The ORF7 sequence data also showed no apparent associations between geographic or temporal origin and heterogeneity of PRRSV in China.</p> <p>Conclusion</p> <p>These findings enhance our knowledge of the genetic characteristics of Chinese PRRSV isolates, and may facilitate the development of effective strategies for monitoring and controlling PRRSV in China.</p

    A confidence limit for the empirical mode decomposition and Hilbert spectral analysis

    The confidence limit is a standard measure of the accuracy of the result in any statistical analysis. Most of the confidence limits are derived as follows. The data are first divided into subsections and then, under the ergodic assumption, the temporal mean is substituted for the ensemble mean. Next, the confidence limit is defined as a range of standard deviations from this mean. However, such a confidence limit is valid only for linear and stationary processes. Furthermore, in order for the ergodic assumption to be valid, the subsections have to be statistically independent. For non‐stationary and nonlinear processes, such an analysis is no longer valid. The confidence limit of the method here termed EMD/HSA (for empirical mode decomposition/Hilbert spectral analysis) is introduced by using various adjustable stopping criteria in the sifting processes of the EMD step to generate a sample set of intrinsic mode functions (IMFs). The EMD technique acts as a pre‐processor for HSA on the original data, producing a set of components (IMFs) from the original data that equal the original data when added back together. Each IMF represents a scale in the data, from smallest to largest. The ensemble mean and standard deviation of the IMF sample sets obtained with different stopping criteria are calculated, and these form a simple random sample set. The confidence limit for EMD/HSA is then defined as a range of standard deviations from the ensemble mean. Without evoking the ergodic assumption, subdivision of the data stream into short sections is unnecessary; hence, the results and the confidence limit retain the full‐frequency resolution of the full dataset. This new confidence limit can be applied to the analysis of nonlinear and non‐stationary processes by these new techniques. Data from length‐of‐day measurements and a particularly violent recent earthquake are used to demonstrate how the confidence limit is obtained and applied. By providing a confidence limit for this new approach, a stable range of stopping criteria for the decomposition or sifting phase (EMD) has been established, making the results of the final processing with HSA, and the entire EMD/HSA method, more definitive

    Carvacrol protects mice against LPS-induced sepsis and attenuates inflammatory response in macrophages by modulating the ERK1/2 pathway

    Abstract Macrophages play an important role in the development of life-threatening sepsis, which is characterized by multiorgan dysfunction, through their ability to produce inflammatory cytokines. Carvacrol is a phenolic compound that has been confirmed to possess strong anti‑inflammatory activity. In this study, we mainly investigated the effect of carvacrol on lipopolysaccharide (LPS)-induced macrophage proinflammatory responses and endotoxic shock. The results showed that carvacrol significantly reduced mouse body weight loss and ameliorated pathological damage to the liver, lung, and heart under LPS-induced sepsis. Carvacrol attenuated inflammatory responses by inhibiting the LPS-induced production of inflammatory cytokine interleukin-6 (IL-6) in vivo and in vitro. Mechanistically, carvacrol inhibited IL-6 production mainly through the ERK1/2 signalling pathway in macrophages. Furthermore, carvacrol improved the survival of septic mice. This study sheds light on the role of carvacrol in the pathogenesis of LPS-induced sepsis, and thus, its potential in treating sepsis patients may be considered

    Unmet Needs and Treatment Preference of Systemic Treatments for Moderate-to-Severe Psoriasis from the Perspectives of Patients and Dermatologists in China

    Abstract Introduction The treatment options for moderate to severe psoriasis (msPsO) in China have been greatly increased with the approvals of biologics. However, the unmet needs and treatment preferences of systemic treatments for msPsO in China remain unclarified. Methods Fifty dermatologists and 300 patients with msPsO (41% with severe psoriasis) were surveyed for effectiveness, safety, treatment convenience, and treatment preferences (using a choice-based conjoint questionnaire). Descriptive statistics and conjoint simulation analyses were employed to summarize survey information and assess treatment preferences. Results Both patients and dermatologists reported shorter treatment duration for oral drugs (2.7–6.2 months) than that for biologics (9.5–17.0 months). The most frequently reported treatment discontinuation reasons by the surveyed patients and dermatologists were unsatisfactory effectiveness (average 84.5%) for oral drugs and loss of efficacy over time (average 68.5%) for biologics. Commonly reported treatment inconveniences included regular lab tests for traditional oral drugs (average 71.5%) and administration assistance for biologics (average 58.0%). Injection site reactions (average 51.5%) and needle fear (average 35.5%) were frequently reported for biologics among the surveyed patients and dermatologists. Once-daily oral treatment was preferred over biweekly subcutaneous injection treatment when the two had comparable attributes (average in patients 87.1% vs. 12.9%; average in dermatologists 93.4% vs. 6.6%). Conclusions Unmet needs of systemic treatments remain for msPsO in China. Once-daily oral treatment is preferred over biweekly subcutaneous injections to treat msPsO when other treatment attributes are comparable
