    The effect of increase in the number of fishing trips on the economic efficiency of 9.82 m and 11 m fishing trawlers along Kerala coast

    Cost-benefit analysis of a 9.82 m and 11 m fishing trawlers based on the number of fishing trips is presented. The number of fishing trips per year determines the profit and loss of the trawler. With the increase in the number of fishing trips, the profit also increase for both the sizes of trawlers. The minimum quantity of prawn and fish to be landed for 0-20% profit for varying number of fishing trips are worked out. The break-even for 9.82 and 11m trawlers was observed to be 185 and 210 fishing trips respectively during 1980-81

    Survey on the idle capacity of fish processing (freezing) plants in India - 2. East coast

    The extent of idle capacity in the fish processing (freezing) plants in the east coast of India estimated by stratified random sampling and the factors responsible for the same are reported. The estimates of idle capacity of fish processing plants in the east coast for the years 1978 and 1979 were respectively 75.9% and 72.5% on the basis of 250 working days per annum and double shift per day. The percentage errors of estimates worked out to 6.9 for 1978 and 4.7 for 1979. The corresponding figures were worked out on the basis of 200 working days also. Substantial under-utilisation of plants in all the maritime states in the east coast accounted mainly to non-availability of raw material, high cost of production, shortage of power, scarcity of ice and potable water during peak season and frequent labour troubles

    Controlled packing and single-droplet resolution of 3D-printed functional synthetic tissues

    3D-printing networks of droplets connected by interface bilayers are a powerful platform to build synthetic tissues in which functionality relies on precisely ordered structures. However, the structural precision and consistency in assembling these structures is currently limited, which restricts intricate designs and the complexity of functions performed by synthetic tissues. Here, we report that the equilibrium contact angle (θDIB) between a pair of droplets is a key parameter that dictates the tessellation and precise positioning of hundreds of picolitre-sized droplets within 3D-printed, multi-layer networks. When θDIB approximates the geometrically-derived critical angle (θc) of 35.3°, the resulting networks of droplets arrange in regular hexagonal close-packed (hcp) lattices with the least fraction of defects. With this improved control over droplet packing, we can 3D-print functional synthetic tissues with single-droplet-wide conductive pathways. Our new insights into 3D droplet packing permit the fabrication of complex synthetic tissues, where precisely positioned compartments perform coordinated tasks

ES is the recipient of a Cunningham Trust PhD studentship (PhD-CT-18-41).The enzyme m1A22-tRNA methyltransferase (TrmK) catalyzes the transfer of a methyl group to the N1 of adenine 22 in bacterial tRNAs. TrmK is essential for Staphylococcus aureus survival during infection but has no homolog in mammals, making it a promising target for antibiotic development. Here, we characterize the structure and function of S. aureus TrmK (SaTrmK) using X-ray crystallography, binding assays, and molecular dynamics simulations. We report crystal structures for the SaTrmK apoenzyme as well as in complexes with methyl donor SAM and co-product product SAH. Isothermal titration calorimetry showed that SAM binds to the enzyme with favorable but modest enthalpic and entropic contributions, whereas SAH binding leads to an entropic penalty compensated for by a large favorable enthalpic contribution. Molecular dynamics simulations point to specific motions of the C-terminal domain being altered by SAM binding, which might have implications for tRNA recruitment. In addition, activity assays for SaTrmK-catalyzed methylation of A22 mutants of tRNALeu demonstrate that the adenine at position 22 is absolutely essential. In silico screening of compounds suggested the multifunctional organic toxin plumbagin as a potential inhibitor of TrmK, which was confirmed by activity measurements. Furthermore, LC-MS data indicated the protein was covalently modified by one equivalent of the inhibitor, and proteolytic digestion coupled with LC-MS identified Cys92 in the vicinity of the SAM-binding site as the sole residue modified. These results identify a cryptic binding pocket of SaTrmK, laying a foundation for future structure-based drug discovery.Publisher PDFPeer reviewe

    Variation in structure and properties of poly(glycerol adipate) via control of chain branching during enzymatic synthesis

    Poly (glycerol adipate) (PGA) can be produced from divinyl adipate and unprotected glycerol by an enzymatic route to generate a polymer with relatively low molar mass (12 kDa). PGA bears a pendant hydroxyl group which imparts a hydrophilic character to this water insoluble polymer. We have examined the effect of synthesis temperature on polymer characteristics through various techniques including FT-IR, 1H and 13C NMR, surface and thermal analysis, both to expand the data already present in the literature about this material and to understand better its properties for potential pharmaceutical applications. The use of a lipase (Novozym 435) as a catalyst suppresses cross-linking at the pendant glyceryl hydroxyl through steric hindrance at the active site, thus producing polymers with low degrees of branching (5–30%), and removes the need for any pre- or post-polymerization protection/deprotection reactions. Careful temperature control during synthesis can give polymers with reproducible molecular weights and reduced amounts of polymer branching compared to synthesis at higher temperatures. Due to the ability of the synthetic route to produce a range of structures, PGA generated by enzymatic routes may emerge as a useful biodegradable polymer platform to engineer solid dispersions or nanoparticles for healthcare applications

    Multiwavelength studies of MHD waves in the solar chromosphere: An overview of recent results

    The chromosphere is a thin layer of the solar atmosphere that bridges the relatively cool photosphere and the intensely heated transition region and corona. Compressible and incompressible waves propagating through the chromosphere can supply significant amounts of energy to the interface region and corona. In recent years an abundance of high-resolution observations from state-of-the-art facilities have provided new and exciting ways of disentangling the characteristics of oscillatory phenomena propagating through the dynamic chromosphere. Coupled with rapid advancements in magnetohydrodynamic wave theory, we are now in an ideal position to thoroughly investigate the role waves play in supplying energy to sustain chromospheric and coronal heating. Here, we review the recent progress made in characterising, categorising and interpreting oscillations manifesting in the solar chromosphere, with an impetus placed on their intrinsic energetics.Comment: 48 pages, 25 figures, accepted into Space Science Review

    Titrimetric and spectrophotometric assay of diethylcarbamazine citrate in formulations using iodate and iodide mixture as reagents

    One titrimetric and two spectrophotometric methods are proposed for the determination of diethylcarbamazine citrate (DEC) in bulk drug and in formulations using potassium iodate and potassium iodide as reagent. The methods employ the well-known analytical reaction between iodate and iodide in the presence of acid. In titrimetry (method A), the drug was treated with a measured excess of thiosulfate in the presence of unmeasured excess of iodate-iodide mixture and after a standing time of 10 min, the surplus thiosulfate was determined by back titration with iodine towards starch end point. Titrimetric assay is based on a 1:3 reaction stoichiometry between DEC and iodine and the method is applicable over 2.0-10.0 mg range. The liberated iodine is measured spectrophotometrically at 370 nm (method B) or the iodine-starch complex measured at 570 nm (method C). In both methods, the absorbance is found to be linearly dependent on the concentration of iodine, which in turn is related to DEC concentration. The calibration curves are linear over 2.5-50 and 2.5-30 µg mL-1 DEC for method B and method C, respectively. The calculated molar absorptivity and Sandell sensitivity values were 6.48×103 L mol-1 cm-1 and 0.0604 µg cm-2, respectively, for method B, and their respective values for method C are 9.96×103 L mol-1 cm-1 and 0.0393 µg cm-2. The intra-day and inter-day accuracy and precision studies were carried out according to the ICH guidelines. The methods were successfully applied to the analysis of DEC formulations

    Espacialização da precipitação pluvial por meio de krigagem e cokrigagem

    O objetivo deste trabalho foi avaliar a cokrigagem ordinária, com dados de altitude e distância do mar, em comparação à krigagem ordinária, na espacialização da precipitação pluvial dos períodos anual, seco e úmido, no Estado do Espírito Santo. Os dados de altitude e distância do mar foram obtidos de pontos de amostragem em grades regulares e irregulares. Foram utilizados dados de 108 postos pluviométricos. A avaliação dos métodos e das variáveis foi realizada com base na validação cruzada, tendo-se considerado os erros dos valores preditos e o ajuste dos modelos de regressão linear para valores observados e preditos. A grade regular para amostragem das covariáveis apresentou melhor acurácia de predição em comparação à grade irregular. A cokrigagem produziu resultados mais acurados do que a krigagem, verificados por pequenas diferenças nos erros médios absolutos, capazes de produzir mapas estatisticamente diferentes. A interpolação por cokrigagem e o uso de grades regulares para amostragem são preferíveis, principalmente se as covariáveis são de fácil obtenção e de baixo custo
